表观遗传学是研究基因的核苷酸序列不发生改变的情况下,基因表达或蛋白表达发生可遗传的变化,但又可以通过细胞发育和增殖而稳定遗传现象的一门遗传学分支学科.表观遗传学的研究内容包括DNA甲基化、组蛋白共价修饰、mRNA编辑、染色体重塑、基因沉默等.其调控机制是:细胞内功能基因的选择性激活或灭活.表观基因相对于整个基因组本身,提供了更有序、更特定的基因信息.表观遗传的异常会导致表型的改变,表现为人类疾病,如肿瘤.因此,随着人们对表观遗传学认识的深入,表观遗传学将成为生物医学研究领域的重点.  相似文献

DNA is the hereditary material in humans and almost all other organisms. It is essential for maintaining accurate transmission of genetic information. In the life cycle, DNA replication, cell division, or genome damage, including that caused by endogenous and exogenous agents, may cause DNA aberrations. Of all forms of DNA damage, DNA double-strand breaks(DSBs) are the most serious. If the repair function is defective, DNA damage may cause gene mutation, genome instability, and cell chromosome loss, which in turn can even lead to tumorigenesis. DNA damage can be repaired through multiple mechanisms. Homologous recombination(HR) and non-homologous end joining(NHEJ) are the two main repair mechanisms for DNA DSBs. Increasing amounts of evidence reveal that protein modifications play an essential role in DNA damage repair.Protein deubiquitination is a vital post-translational modification which removes ubiquitin molecules or polyubiquitinated chains from substrates in order to reverse the ubiquitination reaction. This review discusses the role of deubiquitinating enzymes(DUBs) in repairing DNA DSBs. Exploring the molecular mechanisms of DUB regulation in DSB repair will provide new insights to combat human diseases and develop novel therapeutic approaches.  相似文献

Systemic splits between pre-compulsory and compulsory early years education impact on transitions to school through discontinuities in children’s experience. This paper presents data from a critical participatory action research project about transitions between pre-compulsory and compulsory early education schooling in Australia. The project aim was to investigate how transitions to school might be enhanced by developing deeper professional relationships and shared understandings between teachers from both sectors. Within the communicative space afforded by a professional learning community, the participants engaged in critical conversations about their understandings of transitions practices and conditions, including systemic differences. Data analysis provides a snapshot of changes in teachers’ thinking about professional relationships, continuity and factors influencing cross-sectoral professional relationships. Findings suggest that affording opportunities for teachers to re-frame cross-sectoral professional relationships has led to transformative changes to transitions practices, understandings and conditions.  相似文献

确立从作者的“自审”角度来解读作品,是源于笔者认为杨绛在干校中当时的生活状态,同回京八年后通过回忆来记录的状态,虽在叙述事实上不存在太大差异但在情感上会有一定的变化.如,社会生活的变化,历史语境的变化,自我生活状态的变化,人生经验的变化,知识心理构成等等诸多因素都会影响作家的创作.即使这些影响是细微的,但笔者认为不应该忽视.因此,本文试图通过文本分析来寻找这种差异.而在20世纪80年代初期弥漫着关于反思、自审、忏悔、伤痕这些命题的文学环境下,但凡经历过文化大革命的这些作家们都会将这一人生经历用这些情感来消化.而杨绛的家庭环境、知识构成、人生阅历则将她的这一遭遇淡化,而且不断在从自身找原因,从而确立了她从知识分子“自审”角度展开.  相似文献

Variations in phenotype reflect the influence of environmental conditions during development on cellular functions, including that of the genome. The recent integration of epigenetics into developmental psychobiology illustrates the processes by which environmental conditions in early life structurally alter DNA, providing a physical basis for the influence of the perinatal environmental signals on phenotype over the life of the individual. This review focuses on the enduring effects of naturally occurring variations in maternal care on gene expression and phenotype to provide an example of environmentally driven plasticity at the level of the DNA, revealing the interdependence of gene and environmental in the regulation of phenotype.  相似文献

ABSTRACT

Knowledge and human identity are socially constructed phenomena. Like other aspects of human identity, teacher identity is shaped through social influences. Some of the social influences that contribute to teacher identity development happen through critical incidents that teachers or teacher candidates experience. The current study, adopting a critical incident analysis perspective, attempts to ascertain the nature of the critical incidents that 49 pre-service teachers of EFL have experienced throughout their English learning process. It also tries to uncover the effects of these critical incidents on their English learning and teacher identity. The data for the research was collected using a Critical Incident Form developed by the researchers and focus group interviews. The findings indicated that pre-service ELT teachers’ early learning experiences included certain critical incidents. An inductive quantitative analysis of these incidents revealed that they can be categorized into three themes: people, context, and outcome. Exploring the nature of critical incidents and their role in the construction of English language teacher identity will hopefully be a benefit to teachers and teacher educators.  相似文献

It is generally agreed that adipocytes originate from mesenchymal stem cells in what can be divided into two processes: determination and differentiation. In the past decade, many factors associated with epigenetic signals have been proved to be pivotal for the appropriate timing of adipogenesis progression. A large number of coregulators at critical gene promoters set up specific patterns of DNA methylation, histone acetylation and methylation, and nucleosome rearrangement, that act as an epigenetic code to modulate the correct progress of adipocyte differentiation and adipogenesis during adipogenesis. In this review, we focus on the functions and roles of epigenetic processes in preadipocyte differentiation and adipogenesis.  相似文献

Abstract

Problem‐posing teaching using multicultural children's literature nourishes an integrated literacy curriculum that supports young children's meaningful learning. This method encourages integrated learning that is both developmentally and culturally meaningful through interacting with story, reading literature, and participating in related learning activities. The problem‐posing method was developed by Paulo Freire [Education for Critical Consciousness, Seabury, New York] and critical pedagogists. The method leads students of any age, experience or ability level to base new learning on personal experience in a way that encourages critical reflection. This method has not been widely used with younger learners, but lends itself well to integrated early childhood literacy development.

This article shows selected qualitative data samples from case studies of early childhood teacher education students as they experience the method in a literacy course and as they use the method with young children. A critical analysis of the students’ work draws out key points regarding literacy development in a rapidly changing world. The teacher education students’ work provides an arena for developing the theory further as they implement theoretically‐based pedagogy with young learners. Data reveal issues regarding critical literacies and postmodern approaches to early childhood education.  相似文献

This brief synopsis of an organizational perspective on early development represents an integration of three major areas of the author's research: that of a detailed observational study of early mother-infant interaction over the first three years of life; that of a continuous neonatal state and caregiving interactional monitoring method over the first two months of life; and that of a 25- to 30-year follow-up on the same infants observed initially. From these data a ground plan is proposed for our thinking about the individual's life span trajectory as a unique construction within a unique context for that individual's interactional and adaptive self-regulatory strategies. Beginning with a review of biological principles, the paradoxical integration of complexity and unity in living systems is traced from the conceptualization of infant state and observations on its regulation through the role of state in the origins of awareness of inner experience, to the consolidation and validation of the self as agent in self-regulation. Central to integration is the recognition process, stemming from one's awareness that another is aware of what one is aware of within oneself. The constructionist perspective on the critical role of specificity in this organizing process is illustrated by reference to the negotiation between caregiver and infant of seven issues of adaptive coordination over the first three years of life. Finally, five propositions are formulated describing such specificity in this organizing process, as one that underlies integration of levels of the living system relating the biological, the developmental, the shaping of the life span trajectory, and the reconstructive therapeutic process.  相似文献

Maintenance of cellular homeostasis and genome integrity is a critical responsibility of DNA double-strand break(DSB)signaling.P53-binding protein 1(53BP1)plays a critical role in coordinating the DSB repair pathway choice and promotes the non-homologous end-joining(NHEJ)-mediated DSB repair pathway that rejoins DSB ends.New insights have been gained into a basic molecular mechanism that is involved in 53BP1 recruitment to the DNA lesion and how 53BP1 then recruits the DNA break-responsive effectors that promote NHEJ-mediated DSB repair while inhibiting homologous recombination(HR)signaling.This review focuses on the up-and downstream pathways of 53BP1 and how 53BP1 promotes NHEJ-mediated DSB repair,which in turn promotes the sensitivity of poly(ADP-ribose)polymerase inhibitor(PARPi)in BRCA1-deficient cancers and consequently provides an avenue for improving cancer therapy strategies.  相似文献

Several concepts from the adult learning literature are described that relate to working with parents of young children who are deaf. It is proposed that an adult learner perspective makes an important contribution to the family-centered philosophy and knowledge base that guide the development of practice in early intervention. Three areas are discussed: the context for adult learning, that is, the life situation of adults, including their social roles and responsibilities and experience; learning related to transitions in the adult life cycle; and transformative learning, which examines the ways people make changes in how they understand their experience. 本文分析总结了回归十年来香港幼教改革和发展的四点经验(﹕1)改革未动﹐师训先行(。2)设立基金﹐资助科研。(3)三角架构﹐全面保障(。4)实施教育券制度﹐全面资助幼儿及幼儿园。据此得出六点重要启示(﹕1)发展幼教是政府不可推卸的责任(。2)经费投入是保证幼教健康发展的关键。(3)建构科学合理的幼教管治架构是保证幼教健康发展的前提(。4)将幼教推向市场并不意味着政府可以减少投入或完全不干预市场(。5)专家到园支持园本发展是教改成功的保障(。6)提供专业培训﹐提升教师素质是当务之急。最后,笔者展望内地幼教体制改革之路,指出当前内地幼教改革有四大急务(﹕1)明“确幼教是政府不可推卸的责任”(。2)保证政府对幼教的经费投入(。3)建构科学合理的幼教管治架构(。4)委托学术机构﹐开展科学研究和科学规划。  相似文献

马克思的辩证法是关于人自我诞生的辩证法,也就是对人迄今为止的异化存在现实的描述,所以,它必然是批判性的,但不止于观念的批判,它试图恢复人的已经被异化的感性存在,恢复人与世界的感性关联。马克思的辩证法与人的具体生存经验和历史具有一种批判性的关联,这种关联是来自生活本身的超越性指向,并且这种超越性是要返回到现实存在中去,批判地澄清资本主义社会对人的感性生命的异化与颠倒,并通过革命性的实践活动来改造这种现实,也就是恢复人的感性生命自我实现的要求,这就是马克思辩证法的批判本性。  相似文献
