Erythrocyte membrane protein glycosylation increase by 3.4 fold in diabetes. Insulin or sulfonylurea treatment did not reduce the extent of glycosylation. The serum protein glycosylation was comparable in all the groups including control. Erythrocyte membrane Na+,K+-ATPase activity decreased in the diabetics; only insulin treatment partly restored the activity. Erythrocyte membrane acetylcholinesterase activity decreased only in the sulfonylurea treated group. Serum butyrylcholinesterase activity was relatively low in the diabetic and insulin treated diabetic groups. The Km and Vmax of the two components of Na+,K+-ATPase from erythrocyte membranes were differently affected in the diabetic and the two treatment groups. The Vmax of acetylcholinesterase decreased only in the sulfonylurea treated group. Diabetic states resulted in decreased Vmax of components I and II of serum butyrylcholinesterase. In insulin-treated diabetics, component II was absent. Sulfonylurea group resembled diabetics.In vitro incubation with insulin differentially affected the Na+,K+-ATPase and serum butyrylcholinesterase activities.  相似文献

Plasma levels of lipids, lipoproteins and lipid peroxides and erythrocyte Na+−K+ ATPase, Mg2+ATPase and antioxidant enzymes were measured in type-2 diabetic patients. A significant decrease in Na+-K+-ATPase activity was observed in diabetic patients which was negatively correlated with blood glucose and lipid peroxides, while the Mg2+-ATPase activity was increased. In the diabetic subjects the plasma concentrations of Na+ and K+ were increased where as erythrocyte levels of Na+ were increased and K+ were decreased. Hyperlipidaemia and increased levels of lipid peroxides were observed in the diabetic subjects. There was a significant increase in erythrocyte catalase activity in diabetics which positively correlated with their lipid peroxides. There was no change in GPx activities between controls and diabetics.  相似文献

The effect of DL α-lipoic acid, a potent antioxidant was studied in relation to certain erythrocyte membrane parameters in calcium oxalate stone forming rats. Induction of calcium oxalate lithiasis was done by feeding a diet containing 3% w/w sodium glycollate. Erythrocyte membrane (Na+, K+)-ATPase showed a significant decrease in stone formers whereas (Ca2+)-ATPase showed a significant increase. Lipoic acid administration brought about an elevation in the activity of (Ca2+)-ATPase. Changes in membrane (Mg2+)-ATPase was minimal. Membrane cholesterol and phospholipids were found raised significantly in lithogenic rats. The changes may be attributed to enhanced lipid peroxidative mechanisms and altered serum lipid profile observed in this group. Treatment with lipoic acid reduced membrane cholesterol levels. Phospholipids were also decreased moderately. The above observations suggest that lipoic acid administration to calculogenic rats reduces the erythrocyte strucutral changes observed in this condition.  相似文献

The sarcoplasmic reticulum was isolated and characterized in the pure form from normal caprine (Capra hircus) hearts. Under ischemic condition, fatty acids like palmitic acid, palmitoyl carnitine and oleic acid accumulatein vivo. These are known to have a detrimental effect on membrane components. The effects of simulated ischemia on the Ca2+-ATPase were studied using these fatty acidsin vitro at concentrations at which they occurin vivo in the ischemic heart. All three fatty acids inhibited Ca2+-ATPase activity. At lower concentrations, palmitoyl carnitine was the most potent, followed by oleic acid while palmitic acid displayed the least potency. Electron microscopy studies with fatty acids showed morphological disruptions in SR vesicles. The decline in Ca2+-ATPase activity could be attributed to the change in membrane morphology.  相似文献

In view of several reports that there is a lack of balance in free radicals in case of bronchial asthma (1) the effect of free radicals on cell membrane was studied by estimating the membrane bound protein Na+, K+-ATPase activity and the intracellular sodium level in patients of bronchial asthma before and after a short course (one week) of oral corticosteroid (prednisolone 0.75–1mg/kg body weight) therapy. Results showed that there is a definite statistically significant rise in free radical level and intracellular sodium level and a significant lowering of Na+, K+-ATPase activity in case of untreated bronchial asthma. After short course of therapy with oral corticosteroids, the free radical level and intracellular sodium level decreased significantly, together with a significant rise of the Na+, K+-ATPase activity. Also, a significant negative correlation (r=−0.74) between the lipid peroxide level and the Na+,-K+-ATPase activity was found in these cases.  相似文献

Increased oxidative stress is a widely accepted participant in the development and progression of diabetes and its complications. The present study has been undertaken to evaluate oxidative stress in type 2 diabetes mellitus and effect of vitamin E supplementation on oxidative stress. In all 120 subjects were enrolled in the present study, 40 subjects are age and sex matched controls. Test group comprised of clinically diagnosed (n = 80) type 2 diabetic patients. Biochemical parameters like serum MDA, nitric oxide, superoxide dismutase, erythrocyte reduced glutathione and platelet aggregation were analyzed in control and diabetic group. Test group is further categorized as Group I (n = 40) diabetics were treated by only hypoglycemic drugs and Group II (n = 40) diabetics were treated by hypoglycemic drugs with vitamin E supplementation. All above biochemical parameters were again reassessed after 3 months follow-up in both group and its values were compared with its respective baseline levels. The study shows, reduction of oxidative stress, improvement in antioxidant enzymes and endothelial dysfunction in group II, those were on treatment of hypoglycemic drugs along with vitamin E supplementation. Hence the present study may conclude that vitamin E supplementation along with hypoglycemic drugs may be beneficial to type 2 DM patients to minimize vascular complications.  相似文献

Measurement of serum free (ionized) calcium (Ca++) reflects true calcium status of the body in health and disease. Present study evaluates efficacy of Ca++ over total calcium (CaT) in serum for calcium status. 52 subjects were enrolled for study. Anaerobic fasting blood sample for Ca++ measurement and autoclaved plain bulb for estimation of CaT, Total protein (TP) and Albumin was used. CaT, Corrected CaT, Ca++, Calculated Ca++ were measured and correlated. Corrected CaT and calculated Ca++ were derived from the measured parameters. Study group showed significant difference between CaT and corrected CaT (p<0.006), Ca++ and calculated Ca++ (p<0.001). Negligible correlation was observed between Ca++ and serum protein. Positive correlation was observed between CaT and calculated Ca++, TP and albumin. Findings indicate that Ca++ levels are independent of serum protein status. With scrupulous sampling, Ca++ may be a better parameter than presently used CaT for assessing calcium status in serum.  相似文献

Prostaglandins and (PG) have been reported to be an important gastric acid suppressive factor. However, the mechanism underlying is yet to be clearly established. In vitro study with gastric microsomes in presence of both PGE2 and PGI2 shows a stimulation of gastric H+ K+-ATPase activity below 1X10−6M and 2.5X10−7M concentrations respectively. However, with further increase in concentrations of both PGE2 and PGI2, H+, K+-ATPase activity shows an inhibition but PGI2 completely obliterates the K+ stimulated part of H+, K+-ATPase activity at higher concentration. The H+-ion transport study using chambered frog gastric mucosa shows that both PGE2 and PGI2 inhibit H+-ion transport at 5X10−6 M and 10X10−6M concentrations respectively but the effect of PGI2 is reversible. These differential effects of PGE2 and PGI2 on microsomal H+, K+-ATPase and on H+ transport my be caused by the differential effects of these phospholipid mediators with the gastric mucosal cell membrane. This in vitro investigation shows the role of prostaglandin (s) as a physiological switch/regulator of gastric H+ ion transport leading to the cessation of gastric acid secretion.  相似文献

The present study deals with the evaluation of glycemic profile of aqueous extract of Cajanus cajan leaves in streptozotocin induced type 2 diabetic rats. Single oral administration of graded doses of aqueous extract of Cajanus cajan leaves showed significant increment of 14.3 % in fasting blood glucose levels of normal rats. The sub diabetic and mild diabetic models have also shown hyperglycemic effect from the same variable doses of the extract. The dose of 1000 mg/kg showed the maximum rise of 17.1, 71.2 and 50.7 % in Blood glucose levels of normal, sub and mild diabetic rats respectively during glucose tolerance test. The study of leaves was taken into consideration on the basis of earlier reported hypoglycemic activity of Cajanus cajan seeds. However the results observed were found just opposite and therefore it may be useful in controlling hypoglycemia, occasionally caused due to excess of insulin and other hypoglycemic drugs.  相似文献

The effect of sodium pentosan polysulphate (SPP), was studied in relation to certain blood and erythrocyte membrane parameters in calcium oxalate stone forming rats. Calcium oxalate stones were induced by feeding the rats with 3% w/w sodium glycollate. Fibrinogen, haemoglobin and serum protein levels did not show any variation with the treatment procedures. Serum mucoprotein and protein bound carbohydrates-hexosamine and sialic acid-were increased significantly in the rats receiving calculogenic (CPD) and attained nearly normal levels with SPP treatment. In contrast, hexuronic acid level was decreased in the CPD group and SPP administration increased the level of hexuronic acid in the treated groups. Erythrocyte membrane Ca2+-ATPase activity was increased in stone forming rats and SPP administration brought a reduction in the above enzyme activity. Changes in Membrane Mg2+- and Na+, K+-ATPases were minimal. Membrane cholesterol and phospholipids were also raised significantly in stone formers, SPP treatment reduced the membrane cholesterol levels in both controls and stone formers. Phospholipids were also decreased moderately. The above observations suggest that SPP is safe for administration in urolithiatic condition without adverse effects.  相似文献

Ca++ was not found to be essential for the activation of the latent elastase in normal lenses. However, the activities increased 70–100% in human, bovine, sheep and goat lenses at a concentration of 5.0×10?3M of CaCl2. On the other hand, in cataractous human lenses, Ca++ inhibited the process of activation. The activation process however, seems to be a divalent metal ion dependent process since ethylene diamine tetra acetate (2.0×10?3M) blocked the activation of the elastase-like enzyme in both normal and cataractous lenses. p-Hydroxy mercuribenzoate (10?4M) and benzamidine (1.65×10?2M) also blocked the activation process, indicating that a trypsin-like enzyme requiring sulfhydryl groups, is involved in the activation of latent elastase.  相似文献

This study was aimed at isolating, in its pure form, and characterizing the sarcoplasmic reticulum from caprine (Capra hircus) heart. The sarcoplasmic reticulum from thirty caprine heart ventricular homogenates was isolated and purified. It was characterized on the basis of both, its protein and lipid composition. The protein content was 142±10 mg/g of tissue. Ca2+-ATPase activity equaled 3.75±1.06mmol Pi/mg protein/min while the uptake rate was 24±1.14 nmol/mg protein/min. 205kD, 110kD, 90kD, 84kD, 66kD, 55kD and 29kD molecular weight proteins were seen on an SDS polyacrylamide gel. Triglyceride, Cholesterol and Phospholipids (phosphatidylethanolamine, phosphatidylinositol, phosphatidylcholine, sphingomyelin and phosphatidylserine) were present in increasing order of their concentration. Long chain fatty acids predominated over the unsaturated ones. The ryanodine receptor displayed two binding sites for ryanodine. Characterisation encompassing the above biochemical aspects of normal caprine cardiac sarcoplasmic reticulum was thus achieved after isolating it in the pure form.  相似文献

Elevated plasma triglyceride and non-esterified fatty acid concentrations may cause insulin resistance and type 2 diabetes mellitus. Lipoprotein lipase (LPL) is a rate-determining enzyme in lipid metabolism. A variant in the LPL gene has been identified which alters the penultimate amino acid Serine at 447 to a stop codon (S447X), and results in a truncated LPL molecule lacking the C-terminal dipeptide Ser–Gly. The present study was designed to evaluate the frequency of S447X variant in the LPL gene and its effect on the lipid and lipoprotein levels in type 2 diabetic subjects. The genotype frequency distributions of type 2 diabetes patients and controls were in Hardy–Weinberg equilibrium. Comparison of the genotype and allelic frequencies of S447X in subjects with type 2 diabetics compared to controls demonstrated no significant difference. In subjects with type 2 diabetics having hypertriglyceridemia (TG ≥ 150 mg/dl) compared to diabetics with TG level <150 mg/dl, significant difference in genotype frequency was found among these groups, while allelic frequency of X was significantly differed. Logistic regression analysis showed the negative association of LPL S447X variant with TG and VLDL cholesterol, while no association with total cholesterol, HDL cholesterol and LDL cholesterol was found. The lipid levels except for HDL cholesterol were found to be significantly lower in carriers for S447X than wild type in diabetes group. The decreased level of TG and TG rich lipoprotein in subjects with SNP S447X in LPL, predicts anti-atherogenic activity of carriers for S447X variant in general population as well as type 2 diabetic patients.  相似文献

A distinguishable feature of type 2 diabetes besides hyperglycemia and deranged lipid profile is an impaired insulin secretion, peripheral insulin resistance and obesity which has become a major health concern worldwide. India with an estimated 31million diabetics in 2000 and 79mllions by the yr 2030 has the highest number of type 2 diabetics in the world. In this study, we aimed to see if yoga-asanas and pranayamas have any influence in modifying certain biochemical parameters. Sixty patients of uncomplicated type 2 diabetes (age 35–60 yrs of 1–10 yrs duration) were divided into two groups: Group 1 (n=30): performed yoga along with the conventional hypoglycemic medicines and group 2 (n=30): patients who only received conventional medicines. Duration of the study was 45 days. Basal recordings of blood glucose (fasting and post-prandial), lipid profile and serum insulin were taken at the time of recruitment and the second reading after forty five days. Results showed a significant improvement in all the biochemical parameters in group 1 while group 2 showed significant improvement in only few parameters, thus suggesting a beneficial effect of yoga regimen on these parameters in diabetic patients.  相似文献

In this study, differences in lipid levels amongst diabetics with and without complications were assessed to determine lipid disorders that are associated with diabetic complications other than cardiovascular diseases. A Cross sectional study design was employed. The study included 288 diabetics and 108 non diabetics with different types of complications such as hypertension, nephropathy, neuropathy, and retinopathy. The mean serum total cholesterol was higher in patients with complications compared to those without complications and the non-diabetic controls. The normotensive diabetic patients had the lowest total cholesterol among the diabetic patients’ groups (4.65 ± 0.17 mmol/l) compared to the diabetics with hypertension (6.051 ± 0.20 mmol/l), retinopathy (6.26 ± 0.29 mmol/l), neuropathy (5.80 ± 0.17 mmol/l) and nephropathy patients 5.74 ± 0.26 mmol/l (P < 0.05). The prevalence of dyslipidaemia among diabetic subjects was between 19.2 and 84.0%. The study shows that, in addition to macrovascular complications, dyslipidaemia is common in type 2 diabetes mellitus patients with microvascular complications.  相似文献

Water extract of dry fruits ofTerminalia chebula (Hindi-Harda, Telugu-Karakkaya) at a dose of 200 mg/kg body weight improved the glucose tolerance as indicated by 44% of reduction in the peak blood glucose at 2nd hour in glucose tolerance test in diabetic (streptozotocin induced) rats. Treatment of diabetic rats with an initial fasting blood glucose of 253±9.4 mg/dl daily once with the water extract (200 mg/kg) for two weeks brought down the fasting blood glucose to 123±8.4 mg/dl which is only slightly above the normal value. These results indicate that water extract of Terminalia chebula improves glucose tolerance and brings down fasting blood glucose in diabetic rats.  相似文献

Serum levels of cholesterol, triglyceride, free fatty acid (FFA) and high density lipoprotein (HDL) cholesterol were studied in 10 undernourished and 10 better nourished diabetic patients at different periods of insulin treatment and in non-diabetic controls. Serum concentrations of cholesterol, triglyceride and FFA were significantly higher in untreated diabetic patients than in control but the elevated lipid levels gradually shifted towards normal on insulin treatment. The HDL cholesterol, on the other hand, was significantly lower in untreated diabetic subjects than those in controls and the HDL cholesterol also gradually modified on insulin therapy. However, the improvement of all the lipid parameters on insulin treatment was somewhat delayed in undernourished compared to those of better nourished diabetics. These results, therefore, suggest that malnutrition interferes with the serum lipid improvement in diabetics on insulin treatment and may make them more prone to develop vascular complications.  相似文献

The influence of Crataeva nurvala bark decoction was studied in calcium oxalate stone forming rats, in relation to oxalate metabolism in liver. The activities of the major oxalate synthesizing enzymes in liver namely, glycollate oxidase (GAO) and lactate dehydrogenase (LDH) were significantly increased in the calculogenic group. Bark decoction treatment lowered the liver GAO activity considerably. Transport ATPases (Na+, K+ and Ca2+-ATPases) and alkaline phosphatase were enhanced in rats fed calculi producing diet, while the activities of acid phosphatase, inorganic pyrophosphatase and aminotransferases were slightly reduced. Bark decoction administration produced a marginal decrease in Na+, K+-ATPase and increase in aspartate aminotransferase activities, without significantly altering other enzyme activities. The decrease in liver GAO activity seen during bark decoction treatment, with concomitant decrease in kidney oxalate level, may prove beneficial as a prophylactic measure in preventing stone recurrence.  相似文献

There was increase in erythrocyte lipid peroxidation and adenosine deaminase (ADA) activity in red blood cells (RBC's), white blood cells (WBC's) and plasma, in 30 patients of confirmed diabetes mellitus, 10 each of insulin dependent diabetes mellitus (IDDM or type I), non-insulin dependent diabetes mellitus (NIDDM or type II) and diabetes with ketoacidosis when compared with 20 healthy individuals (controls). Glycosylated hemoglobin A1c%, plasma free fatty acid (FFA) and glycerol levels were also elevated in all the cases when compared with controls. The increase was more in diabetic ketoacidosis group. Significant positive correlations were seen between erythrocyte lipid peroxidation and ADA activities in RBC's, WBC's and plasma. It is suggested that decreased tissue adenosine levels due to increase in ADA activity, is related to the severity of hyperglycemia and lipid peroxidation in diabetes mellitus.  相似文献

Type 2 diabetes mellitus consists of dysfunctions characterized by hyperglycemia and resulting from combination of resistance to insulin action and inadequate insulin secretion. Most of diabetic patients report significant gastrointestinal symptoms. Entire GI tract can be affected by diabetes from oral cavity to large bowel and anorectal region. Proteins, carbohydrates, fats, and most fluids are absorbed in small intestine. Malabsorption may occurs when proper absorption of nutrients does not take place due to bacterial overgrowth or altered gut motility. The present study was planned to measure various malabsorption parameters in type 2 diabetic patients. 175 patients and 175 age and sex matched healthy controls attending Endocrinology Clinic in PGI, Chandigarh were enrolled. Lactose intolerance was measured by using non-invasive lactose hydrogen breath test. Urinary d-xylose and fecal fat were estimated using standard methods. Orocecal transit time and small intestinal bacterial overgrowth were measured using non-invasive lactulose and glucose breath test respectively. Out of 175 diabetic patients enrolled, 87 were males while among 175 healthy subjects 88 were males. SIBO was observed in 14.8 % type 2 diabetic patients and in 2.8 % of controls. There was statistically significant increase (p < 0.002) in OCTT in type 2 diabetic patients compared with controls. OCTT was observed to be more delayed (p < 0.003) in patients who were found to have SIBO than in patients without SIBO. Lactose intolerance was observed in 60 % diabetic patients and 39.4 % in controls. Urinary d-xylose levels were also lower in case of diabetic patients but no significant difference was found in 72 h fecal fat excretion among diabetic patients and controls. Urinary d-xylose and lactose intolerance in SIBO positive type 2 diabetic patients was more severe as compared to SIBO negative diabetic patients. From this study we can conclude that delayed OCTT may have led to SIBO which may have instigated the process of malabsorption among type 2 diabetic patients.  相似文献
