Abstract

The purpose of this study was to analyse the effect of regular exercise on spleen and peritoneal exudate reactive oxygen species (ROS) and lymphocyte proliferation by splenocytes. Twenty-four female BALB/c mice were randomly divided into trained (n = 12) and untrained (n = 12) groups. These two groups were further divided into mice that were studied at rest (trained/rest, n = 5; untrained/rest, n = 6) and immediately after a 2 h acute bout of exercise (trained/exercise, n = 6; untrained/exercise, n = 6). The animals were bred in the animal facility of the Yonsei University College of Medicine, where they were housed in a temperature- (22 – 24°C) and humidity- (50 – 60%) controlled environment, with a 12 h photoperiod, and provided with food and water ad libitum. The trained mice underwent 10 weeks of endurance swimming training (5 days per week) in water at 26 – 29°C for 60 min. Changes in body mass, proliferative activity and the production of reactive oxygen species from spleen lymphocytes and peritoneal exudate cells were determined. The splenic lymphocytes of the trained mice had much greater proliferative activity than those of the untrained mice (P < 0.05). Trained mice had lower ROS production in splenic lymphocytes and peritoneal exudate cells than untrained mice. In both groups, there was substantial inhibition of proliferative activity stimulated with medium, concanavalin A and lipopolysaccharide following the acute bout of exercise. This may have been caused by excessive ROS production following the acute exercise session.  相似文献

There is a growing interest in exploring irisin response to acute exercise; however, the associations of acute exercise-induced irisin release with training status and exercise mode are not fully understood. This study was primarily designed to evaluate these associations. Sixteen healthy adults (8 trained versus 8 untrained) underwent a bout of cycling at 80% of maximal oxygen uptake (VO2max) for 50?min, with blood drawn pre-, 10-, and 180-min post-exercise. Another 17 healthy adults performed 2 bouts of graded exercise (cycling and running) until exhaustion on separate days using a randomized cross-over design, with blood taken pre-, 0-, 10-, and 60-min post-exercise. Circulating irisin, creatine kinase (CK), aspartate aminotransferase (AST), and myoglobin (Mb) were measured, and their respective areas under the curves (AUCs) were calculated. Irisin increased 10-min after 50?min of cycling at 80% of VO2max, while its changes from baseline to post-exercise and the amount of exercise-induced irisin release (presented as AUC) were comparable between trained and untrained adults (all P?>?.05). Irisin remained elevated 10-min post-exhausting running but decreased towards baseline 10-min post-exhausting cycling. Exhausting running induced an increase in irisin release for the whole course of exercise and recovery periods, but cycling did not. Acute exercise-induced irisin changes seemed not related to changes of CK, aspartate AST, and Mb in general. In conclusion, acute exercise-induced irisin release is not associated with training status but might be affected by training mode. Future studies are required to investigate which exercise mode might be most efficient in altering irisin.  相似文献

目的:探讨不同运动对小鼠心脏形态结构及心肌微小RNA(miRNA)的影响,了解miRNA在运动性心脏肥大发生过程中基因转录后的调节作用。方法:C57BL/6雄性小鼠随机分为3组,每组8只,为安静对照组(C组)、一次性力竭运动组(E组)和8周耐力训练组(T组)。E组进行一次力竭性游泳运动;T组进行8周游泳训练,每周训练5天,前5周,每晚运动1次,后3周每天运动两次,间隔6h;第1周30min开始训练,每周增加10min直至90min,最后2周均维持在90min。最后1周通过超声心动图检测心脏结构。训练结束后48hT组取心肌,C组和力竭后的E组取心肌,使用荧光定量PCR检测miRNA-1、133、208a、499。结果:T组游泳训练明显提高了心室内径和心壁厚度,运动诱导产生心脏肥大。E组miRNA-1、miRNA-133a、miRNA-208a、miRNA-499表达全部显著性高于C组(P<0.01);T组不同miRNA表达有差异,与C组比较,miRNA-1、133a经历8周训练后显著性下降(P<0.01),而miRNA-208a显著性提高(P<0.05),但提高幅度不及力竭性运动。而T组miRNA-499相比C组升高并不显著。结论:8周游泳耐力训练可以诱导C57BL/6小鼠心肌肥大,力竭性游泳运动能提高miRNA表达,而耐力训练降低miRNA-1、133a表达同时提高了miRNA-208a、499的调节通路,miRNA的变化可能参与了心脏重塑的适应过程,有利于心肌肥大的形成。  相似文献

The purpose of this study was to describe substrate changes in blood and muscles during submaximal exercise and to look at the relationship of these changes to some enzyme activities important in lipid and carbohydrate metabolism. A group of six trained and six untrained men were exercised for 1 hour at 50% [Vdot]O2 max; three from each group cycled and the rest ran on a treadmill. During the exercise, blood-free fatty acid (FFA) levels increased twofold, glycerol by 3.6 times, triglyceride (TG) by 11/100 ml, and glucose by a mean 11 mg/100 ml in the trained men. In the untrained subjects, however, FFA increased 1.8 fold, glycerols 1.6 fold, and TG by a mean 24/100 ml. Glucose fell in every subject by an average 5 mg%. Muscle glycogen depletion averaged 34 mmoles/kg in the untrained and 23 mmoles/kg in the trained group. The activities of succinic acid dehydrogenase (SDH), malic acid dehydrogenase (MDH), and carnitine palmityl transferase (CPT) were greater by 177%, 93%, and 64%, respectively, in the trained group than in the untrained subjects. The results indicated that at the same percent [Vdot]O2 max, the trained men seemed to “preferentially” use FFA as an energy source, possibly due to a well developed Krebs cycle and increased CPT activities. This preference would tend to inhibit the major rate-limiting enzymes of glycolysis, thus slowing even further the use of carbohydrates as an energy source. The differences in response to exercise were independent of fiber type and indicate that these catabolic responses were primarily due to aerobic training.  相似文献

目的:观察少年和成年男子业余跑运动员一次急性耐力运动后心率变异(HRV)变化特征,并探讨运动后HRV变化生理学意义。方法:少年(14.4±1.3岁)与成年(25.7±2.8岁)受试者各10名,进行1次90min、强度为95%个体无氧阈强度跑台运动。在运动前、运动后15min以及运动后6h测定HRV。结果:运动后15min,TPLn、HFnu和HRV各时域指标明显低于运动前水平,LFnu和LF/HF显著升高;少年受试者HFnu降幅、LFnu和LF/HF升幅明显低于成年受试者(P<0.05)。运动后6h,HRV各指标均已恢复,其中,TPLn、SDNN、HFnu和RMSSD有反弹的迹象。运动后HFnu相对运动前的降幅与受试者年龄呈负相关关系(r=-0.645,P<0.01,n=20);另外,HFnu、TPLn相对运动前降低程度与运动前水平高度负相关(P<0.05,n=20)。结论:一次急性耐力运动后HRV出现短暂抑制现象,少年受试者运动后HRV降低幅度相对成年较低,可能与发育成熟度不同有关,急性运动后HRV变化可能是运动适应的生理表现。  相似文献

The purpose of this study was to assess the effect of carbohydrate (CHO) feeding during different periods of two 90-min cycling bouts (the first bout began at 09:00?h and the second bout began at 13:30 h) at 60% maximal oxygen uptake(VO2max) on saliva flow rate and saliva immunoglobulin A (sIgA) responses to the second exercise bout. The study consisted of three investigations: carbohydrate supplementation during (1) the first hour of the recovery interval (CHO-REC), (2) during the first bout of exercise and (3) during the second bout of exercise. Each investigation included two trials completed in a counterbalanced order and separated by at least 4 days. Participants consumed a lemon-flavoured 10% w/v carbohydrate beverage or placebo (22 ml.kg-1 body mass) in the first hour of the recovery interval (n=8) and 500 ml just before exercise, followed by 250 ml every 20 min during exercise in the first (n=9) and second exercise bouts (n=9). Timed unstimulated saliva samples were collected at 10 min before exercise, after 48-50 min of exercise and during the last 2 min of exercise, at 1 h post exercise, 2 h post exercise (first exercise bout only), and 18 h post exercise (second exercise bout only). Venous blood samples were taken 5 min before exercise and immediately after exercise for both exercise bouts in all trials. The main findings of the present study were as follows. First, carbohydrate ingestion during both exercise bouts, but not during the recovery interval, better maintained plasma glucose concentrations and attenuated the increase in plasma adrenaline and cortisol concentrations after the second exercise bout compared with placebo. Second, carbohydrate feeding had no effect on saliva flow rate and sIgA secretion rate compared with placebo. Third, saliva flow rate and sIgA concentration returned to pre-exercise bout 1 values within 2 h in all trials. Fourth, there was no delayed effect of exercise on oral immunity. These findings suggest that carbohydrate ingestion during the first or second bout of exercise, but not during the recovery interval, is likely to better maintain plasma glucose concentrations and attenuate the responses of plasma stress hormones to a second exercise bout than ingestion of fluid alone. Two bouts of 90 min cycling at 60% VO2max on the same day appears to inhibit saliva flow rate during the second exercise bout but does not alter sIgA transcytosis. Our results show that carbohydrate ingestion during any period of two prolonged exercise bouts does not induce different effects on oral immunity compared with placebo.  相似文献

In order to describe the kinetics of the reaction of the heart rate (HR) to the onset of exercise of constant intensity, the half-time (t1/2) of HR can be used. First in a study of exercise of intensity corresponding to 2 W kg-1 on a cycle ergometer, the t1/2 in 15 trained male rowers and 11 untrained male students was determined. In the trained subjects t1/2 was smaller than in untrained students, mean (+/- S.D.) values being 24.10 (+/- 3.36) s and 47.12 (+/- 4.08) s respectively. In both groups t1/2 was positively correlated with resting HR, r = 0.774 and 0.846 and negatively correlated with maximal oxygen uptake (VO2max), r = -0.728 and -0.871 respectively (P less than 0.01). The regulation of HR in the transition range was concluded to be very similar to the regulation of VO2 and energy requirements. The second part of this work was concerned with responses to graded exercise. The linearity of the HR-exercise intensity relationship is maintained up to a submaximal exercise intensity beyond which the increase in exercise intensity exceeds the increase in HR. It was hypothesized that the point where HR departs from linearity in an incremental exercise test may be employed as a predictor of the ventilatory threshold (Tvent). To examine this, 28 trained male long-distance runners were tested on a treadmill and 17 untrained young male subjects were tested on a cycle ergometer using a continuous incremental protocol. The Tvent was determined from the dependence of VE on VO2 and/or VCO2. The VO2, HR and exercise intensity at Tvent were compared with the same parameters determined from the dependence of HR on exercise intensity. No significant differences were found between Tvent and HR break point levels. It was concluded from this second study that the HR break point level coincides with Tvent.  相似文献

The purpose of this investigation was to study the effects of an acute bout of aerobic exercise on state anxiety of women while controlling for iron status (hemoglobin and serum ferritin). Participants were 24 active women, ages 18-20 years (n = 12) and 35-45 years (n = 12). In addition to a nonexercise control condition, participants completed one exercise bout at 60% maximal oxygen uptake (VO2max) and one at 80% VO2max. Each exercise session consisted of a 33-min bout in which participants exercised at their target intensities for a 20-min segment. Immediately before each exercise trial, participants were given the Spielberger State Anxiety Inventory (SAI). The SAI was again administered immediately following the exercise session and at 30, 60, and 90 min postexercise. Data were analyzed using an Age x Intensity x Time (2 x 3 x 5) repeated measures analysis of covariance (ANCOVA) with iron status serving as the covariate. The ANCOVA on state anxiety yielded significant effects for time (p < .0001, eta2(p) = .48), the Intensity x Time interaction (p = .0006, eta2(p) = .19), and the Intensity x Age interaction (p = .04, eta2(p) = .15). All three exercise conditions (including control) showed a decline in state anxiety across time, but the 80% VO2max condition showed a sharper decline. Intensity of exercise conditions did not differ in state anxiety at baseline or immediately after exercise, but a difference favoring the 80% VO2max condition over the control condition emerged at 30 min postexercise. After controlling for iron status, older women who exercised at 80% VO2max exhibited lower SAI scores compared to the control condition.  相似文献

The first purpose of this study was to determine a possible explanation for the variability in the response to eccentric exercise by having participants repeat the same exercise 1 year apart. The second purpose was to examine whether initial injury in response to eccentric exercise was associated with the extent of the repeated bout effect (RBE). Male students performed 30 eccentric contractions (ECC) of the elbow flexors using a dumbbell set at 80% of the pre-exercise maximal isometric force (MIF). Participants were then classified into low (LR; n=6), medium (MR; n=6), high (HR; n=5), and higher (HrR; n=7) based on the increase in blood creatine kinase (CK) activity. A year later, participants repeated this exercise (ECC30). Four days after ECC30, participants performed 70 eccentric contractions (ECC70). Range of motion, MFI upper arm circumference, soreness, and blood CK activity were measured before and up to 9 days after each bout. The change in the criterion measures following ECC and ECC30 were similar for each group. There were no further changes in all parameters after ECC70 for MR, HR, and HrR, although there was a small increase in CK after ECC70 for LR. LR showed a smaller RBE after ECC70 compared with the other groups. It is concluded that participants who exercised 1 year apart showed remarkably similar responses between the bouts. The extent of the RBE following the second bout for the LR group is less for participants who demonstrate the least evidence of muscle damage after a first exercise bout.  相似文献

The aim of the present study was to determine the combined effects of pre-cooling and water ingestion on thermoregulatory responses and exercise capacity at 32 degrees C and 80% relative humidity. Nine untrained males exercised for 60 min on a cycle ergometer at 60% maximal oxygen uptake (VO2max) (first exercise bout) under four separate conditions: No Water intake, Pre-cooling, Water ingestion, and a combination of pre-cooling and water ingestion (Combined). To evaluate the efficacy of these conditions on exercise capacity, the participants exercised to exhaustion at 80% VO2max (second exercise bout) following the first exercise bout. Rectal and mean skin temperatures before the first exercise bout in the Pre-cooling and Combined conditions were significantly lower than in the No Water and Water conditions. At the end of the first exercise bout, rectal temperature was lower in the Combined condition (38.5 +/- 0.1 degrees C) than in the other conditions (No Water: 39.1 +/- 0.1 degrees C; Pre-cooling: 38.7 +/- 0.1 degrees C; Water: 38.8 +/- 0.1 degrees C) (P < 0.05). Heat storage was higher following pre-cooling than when there was no pre-cooling (P < 0.05). The final rectal temperature in the second exercise bout was similar between the four conditions (39.1 +/- 0.1 degrees C). However, exercise time to exhaustion was longer (P < 0.05) in the Combined condition than in the other conditions. Total sweat loss was less following pre-cooling than when there was no pre-cooling (P < 0.001). Evaporative sweat loss in the Water and Combined conditions was greater (P < 0.01) than in the No Water and Pre-cooling conditions. Our results suggest that the combination of pre-cooling and water ingestion increases exercise endurance in a hot environment through enhanced heat storage and decreased thermoregulatory and cardiovascular strain.  相似文献

为了解耐力训练大鼠进行一次定量运动后红细胞的运动氧化应激是否降低以及是否减轻了自由基对其的影响。将60只SD大鼠随机分为不训练组(C)、每次训练20 min组(T1)和每次训练40 min组(T2),训练组在跑台上以28 m/min的速度训练6周。取样前各组又随机分为安静组和定量运动组,测试大鼠红细胞中GPX、SOD及Na -K -ATP酶活性和MDA含量。结果发现定量运动后,未训练大鼠SOD、GPX活性及MDA含量升高,Na -K -ATP酶活性下降;20 min训练大鼠SOD及Na -K -ATP酶活性下降,MDA含量和GPX活性升高且变化程度较未训练大鼠小;40 min训练大鼠各指标变化不大。结果提示有氧耐力运动训练能降低定量运动后红细胞的氧化应激,减轻自由基对红细胞的损伤。  相似文献

研究中等负荷运动训练对心理应激大鼠淋巴细胞凋亡及其调控基因的影响。将健康雄性SD大鼠32只随机分为4组,即对照组(n=8)、心理应激组(n=8)、运动组(n=8)和运动心理应激组(n=8),实验后测定各大鼠外周血中淋巴细胞凋亡及其相关调控基因的变化情况。结果表明,与对照组比较,心理应激导致大鼠外周血淋巴细胞凋亡显著增加,中等负荷的运动训练可以抑制大鼠外周血淋巴细胞的凋亡,运动心理应激组淋巴细胞凋亡与心理应激组比较显著下降,且其发生与相关调控基因的表达一致。提示:中等负荷运动训练可通过影响相关调控凋亡基因降低心理应激导致的大鼠外周血淋巴细胞凋亡,其机制可能是中等负荷的运动训练通过降低心理应激的反应强度来降低大鼠淋巴细胞的凋亡。  相似文献

目的:探索长期运动训练和急性运动的心脏保护作用及心电指标的变化规律,为进一步研究运动预适应机制提供依据.方法:32只3月龄雌性SD大鼠随机分成3大组:间训模型组(n=8)、急性运动模型组和对照组(n=8),间歇运动训练组进行高强度间歇运动训练,急性间歇运动组仅进行急性高强度的间歇运动,对照组(n=16)不运动,在心脏模型制备时对照组随机分成对照模型组(n=8)和对照假手术组(n=8).运动训练结束后,各组大鼠均采用结扎左冠状动脉制备在体大鼠心肌缺血/再灌注损伤模型,但对照假手术组只开胸穿线而不结扎.缺血/再灌注过程实行全程心电监控,缺血30 min、再灌注40 min后,测定血清心肌酶.结果:经间歇运动训练和急性间歇运动处理的缺血/再灌注大鼠血清心肌酶明显低于对照模型组(P<0.05或P<0.01);缺血30 min及再灌注40 min间歇运动训练组心电图ST段、T波面积和QT间期明显低于对照模型组(P<0.05或P<0.01);再灌注40min急性间歇运动组心电图ST段、T波和QT间期明显低于对照模型组(P<0.05).结论:长期间歇运动训练产生心肌抗缺血/再灌注损伤的保护作用,急性运动主要产生抗心脏再灌注损伤的保护作用.  相似文献

探讨不同时间负荷训练大鼠红细胞自由基代谢和Na -K -ATP酶活性。结果表明急性运动时红细胞产生氧化应激,而有氧训练使红细胞自由基代谢得以改善。运动训练改善红细胞自由基代谢和Na -K -ATP酶活性和每日训练持续时间有一定的关系,在一定程度上红细胞自由基代谢水平影响着红细胞膜功能蛋白活性。  相似文献

目的探究长期运动训练对青年男性淋巴细胞凋亡及线粒体DNA(mtDNA)修复酶(8-氧鸟嘌呤DNA糖基化酶,OGG1)的影响及可能机制。方法训练组为20名男子足球专业运动员,对照组为20名男子大学生。2组均在功率自行车上完成递增负荷力竭运动。运动前和运动后即刻采集静脉血。流式细胞法检测淋巴细胞凋亡率,比色法检测超氧阴离子和羟自由基含量和Caspase-3,Caspase-9活性,高效液相色谱法检测mtDNA中8-氧鸟嘌呤(8-oxodG)含量,Western-blotting法检测线粒体OGG1蛋白(mtOGG1)表达水平。结果运动前和运动后,训练组与对照组比较,超氧阴离子、羟自由基、Caspase-3,Caspase-9活性、8-oxodG降低,OGG1升高;运动后,训练组与对照组比较,淋巴细胞凋亡率降低。对照组和训练组在运动后与运动前比较,淋巴细胞凋亡率、超氧阴离子、羟自由基、Caspase-3,Caspase-9活性和8-oxodG均升高,但训练组升高幅度低于对照组;运动后对照组OGG1降低,而训练组OGG1升高。结论一次性力竭运动诱导产生高水平ROS,并抑制mtOGG1表达,使mtDNA氧化损伤,促进淋巴细胞凋亡;长期耐力训练可通过抑制ROS生成,并提高mtOGG1表达,抑制运动性淋巴细胞凋亡。  相似文献

训练组(男子足球运动员,n=16)和对照组(男子大学生,n=16)在功率自行车上完成一次递增负荷力竭运动。运动前、运动后以及运动后24 h采集静脉血,测定外周血淋巴细胞计数、淋巴细胞凋亡率、羟自由基、8-羟基脱氧鸟嘌呤(8-OHdG)和8-氧鸟嘌呤DNA糖基化酶(OGG1)。结果发现,运动后即刻,训练组和对照组外周血淋巴细胞计数(P<0.01;P<0.05)、淋巴细胞凋亡率(P<0.05;P<0.01)、羟自由基(P<0.01;P<0.01)、8-OHdG(P<0.01;P<0.01)和OGG1(P<0.01;P<0.01)均高于运动前水平,而训练组OGG1升高幅度高于对照组(P<0.01),其他各指标升高幅度则均明显低于对照组(均为P<0.01)。运动后24h,对照组外周血淋巴细胞计数低于运动前(P<0.01),淋巴细胞凋亡率高于运动前(P<0.01),而训练组则恢复至运动前;两组8-OHdG(P<0.05;P<0.01)均高于运动前,但升高幅度明显低于对照组(P<0.01);OGG1在对照组恢复至运动前,而训练组仍高于运动前水平(P<0.05)。因此,长期运动训练通过降低氧化应激水平、提高DNA修复能力、减轻DNA氧化损伤,抑制运动性淋巴细胞凋亡。  相似文献

Although much research has examined the relationship between exercise and affect, few studies have considered the effects of exercise bouts of different durations and few researchers have questioned the longer term effects that might be associated with acute exercise. The aim of this study was to compare the effects of the standard health recommended exercise duration of 30 min with a shorter (15 min) bout of exercise upon individuals' affect scores both during and after exercise. Twenty-three (mean age 22.4 years) physically active participants engaged in two counterbalanced cycle ergometer exercise conditions for 15 min (short bout) and 30 min (moderate bout). The participants completed the Subjective Exercise Experience Scale before, during, 5 min, 30 mins, 1 h and 2 h after both exercise conditions. A series of 2 x 6 within-participant repeated-measures analyses of variance for positive well-being (P <0.01), psychological distress (P <0.01) and fatigue (P <0.01) scores revealed significant time main effects, with improved scores over time. No significant differences were noted between the 15-min and 30-min exercise bouts. These results indicate that positive affective responses are experienced by exercisers after relatively short bouts of acute exercise and these effects can still be evident some time later.  相似文献

Abstract

The present experiment evaluated the effects of a 27-day sit up exercise training program on adipose cell size and adiposity. Fat biopsies were taken from the abdomen, subscapular, and gluteal sites by needle aspiration in 13 experimental and 6 control male subjects before and after a five days/week progressive training regimen. Day 1 consisted of 10 bouts of 10-sec exercise, 7 sit ups/bout, with 10-sec rest intervals; on day 27, 14 bouts of 30-sec exercise were performed, 24 sit ups/bout with 10-sec rest intervals. The total number of sit ups done was 5004. Fat cells were isolated using collagenase and photographed to determine cell diameter. Repeated measures ANOVA revealed significant decreases in cell diameter at the three biopsy sites (p<.01), but no significant differences in the rate of change (pre to post) in cell diameter between sites for the experimental and control groups (p>.05). Body weight, total body fat (underwater weighing), and fatfolds and girths remained unaltered. The results demonstrate that (1) the conventional sit up exercise does not preferentially reduce adipose cell size or subcutaneous fat thickness in the abdominal region to a greater extent compared to other adipose sites, and (2) significant changes in fat cell size may occur in the absence of changes in fatfolds, girths or total body composition.  相似文献
