建立了D8S1477和D8S592基因座的分型方法,获得了华东汉族群体D8S1477和D8S592基因座的遗传学数据,并对华东汉族群体数据与华西及印度四个群体的群体遗传学数据做了构成比较．结果表明：D8S1477基因座等位基因频率分布较好,对我国华东地区汉族人群具有较高的非父排除率和个人识别能力,D8S592基因座属非高度多态性STR基因座,不适于法医学应用． 相似文献

目的研究山东地区汉族人群4个STR位点（D10S1248、D14S1434、D22S1045、SE33）的遗传多态性.方法运用PCR扩增、8%非变性聚丙烯酰胺凝胶电泳结合银染技术对152个无关汉族人群个体进行多态性研究.结果4个位点分别检测出72、80、72、136个等位基因片段,共有53个基因型,其频率分布在0.0526~0.1579之间,经检测其多态性分布符合Hardy-weinberg平衡定律.4个STR位点多态信息量（Polymorphism information content,PIC）分别为0.8493、0.8785、0.8173、0.9157.期望杂合度（Heterozygosity,HET）分别为0.8643、0.8892、0.8374、0.9211.结论山东地区汉族该4个STR基因座位点的多态性数据显示其基因分布特征具有特异性. 相似文献

目的:研究新疆维吾尔族人群19个X染色体短串联重复序列(X-STR)基因座和他们组成的7组连锁基因座的单倍型多样性,评价19个X-STR新组合位点的多态信息量和累积个体识别力。为群体遗传学和法医学的应用基础研究提供数据支持;并比较维吾尔族和11个民族在共有的X-STR基因座的遗传学差异。创新点:首次应用一个新的复合扩增检测体系,研究19个X-STR基因座新的组合(DXS8378、DXS7423、DXS10148、DXS10159、DXS10134、DXS7424、DXS10164、DXS10162、DXS7132、DXS10079、DXS6789、DXS101、DXS10103、DXS10101、HPRTB、DXS6809、DXS10075、DXS10074和DXS10135)在新疆维吾尔族的遗传多态性。方法:从233个新疆维吾尔族无关、健康个体的血痕中提取基因组DNA。应用一个新的复合扩增体系,同时对19个X-STR基因座进行扩增,用毛细管电泳进行基因扫描和分型。系统分析和评价这些X-STR基因座常用的各种法医学参数及应用价值;并对7组连锁的基因座组成的单倍型进行分析。基于分子方差分析的方法对新疆维吾尔族和其他11个民族共有的X-STR基因座进行比较研究,探寻这些群体在等位基因频率分布上的差异。结论:研究19个X-STR基因座共发现238个等位基因,相应的基因频率分布在0.0021~0.5644;女性累积个体识别力为0.999 999 999 999 999 999 998 091,男性为0.999 999 999 999 966,累积非父排除率为0.999 999 986 35。分子方差分析的结果显示新疆维吾尔族人群与格陵兰和索马里人群差异最大,有8个基因座存在差异;与马来西亚和关中汉族人群差异最小,仅3个基因座存在差异。结果表明:这19个X-STR基因座多态性高、且具有较高的累积个体识别力,可很好地应用于法医学及群体遗传学研究,也为新疆维吾尔族人群的遗传背景的研究提供基础资料。 相似文献

采用PCR-RFLP方法分析了太湖猪(二花脸)群体中生长激素(growth hormone,GH)、心脏脂肪酸结合蛋白(heart fatty acid-binding protein,H-FABP)和肌细胞生成素(Myogenin,MyoG)基因的多态性.结果表明:在检测的二花脸群体中,GH基因经DraⅠ、MspⅠ酶切无多态性,ApaⅠ酶切出现2个等位基因、3种基因型,基因型频率呈AA>AB>BB的趋势,等位基因A的基因频率大于等位基因B.MyoG基因在PCR1/MspⅠ位点上,基因型AB占据一定的优势;在PCR2/MspⅠ位点上,等位基因A占据绝对的优势,基因型频率趋势为AA>AB>BB.H-FABP基因的HinfⅠ位点上,等位基因h的基因频率有一定的优势.经χ2适合性检验表明,GH、MyoG、H-FABP3个基因在太湖猪(二花脸)群体中的分布均符合Hardy-Weinberg平衡(P>0.05). 相似文献

目的:分析常染色体19个短串联重复序列(STR)位点在新疆蒙古族群体中的遗传多态性分布,为其法医学应用提供数据支持。创新点:丰富了新疆蒙古族群体的常染色体STR遗传数据。方法:从新疆地区采集1133个无血缘关系的蒙古族健康个体,应用一个包含常染色体19个STR位点的复合扩增体系对这些样本进行基因分型检测。同时,基于15个共有的STR位点,采用多维尺度分析、遗传距离(DA)和遗传分化系数(FST)的方法探讨了新疆蒙古族群体和既往报道的20个群体间的群体遗传关系。结论:研究的19个STR位点在新疆蒙古族群体中表现出高度的多态性,获得的累积非父排除概率和累计个体识别率分别为0.999 999 992 163和0.999 999 999 999 999 999 999 995 67,表明这19个STR位点可很好地应用于新疆蒙古族群体的个体识别和亲缘关系鉴识研究。群体遗传分析的结果显示:相比其他洲际群体,新疆蒙古族群体和维吾尔族、锡伯族以及其他中国群体具有更近的遗传距离。 相似文献

目的:探讨LDB3(LIM domain binding 3)基因多态性与中国汉族特发性扩张型心肌病发病的相关性,并分析LDB3基因多态性对患者临床表现和植入型心律转复除颤器(ICD)植入的影响。创新点:首次明确LDB3基因多态性与中国汉族特发性扩张型心肌病发病密切相关,发现了三个与脑钠肽、舒张压、左室射血分数相关的多态性位点和一个与植入心律转复除颤器相关的多态性位点。方法:本研究纳入我院159例中国汉族特发性扩张型心肌病患者和247例无亲缘关系健康对照并收集基线临床资料。提取外周血DNA后,聚合酶链式反应扩增LDB3基因的14个外显子,经Sanger一代测序,获得基因突变位点。通过关联分析,研究基因多态性与特发性扩张型心肌病发病之间的相关性。根据连锁不平衡系数和基因分布频率进行单倍体分析,研究不同单倍体分型对特发性扩张型心肌病发病的影响。结论:在LDB3基因多态性分析中,发现9个多态性位点符合哈代-温伯格平衡。其中rs4468255位点多态性与中国汉族特发性扩张型心肌病发病密切相关。rs11812601、rs56165849和rs3740346位点多态性与患者舒张压、左室射血分数存在显著关联(P0.05)。且携带rs4468255多态性的患者更倾向于安装植入型心律转复除颤器。 相似文献

应用聚合酶链反应(PCR)研究了103例冠心病患者和100例正常人载脂蛋白B(apoB)基因上四个位点的遗传多态性及其与冠心病的关联。结果显示:(1)apoB基因上xbaI和EcoRI酶切位点的少见x~ 和E~-等位基因,VNTR的3＇VNTR-B等位基因不同程度地与冠心病有关联。(2)xbaI酶切位点的多态性与Ins/Del多态性及VNTR多态性呈联锁不平衡关系,多个突变点共位于同一染色体上时冠心病的危险性明显增加。(3)基因单体型对冠心病的危险性具有独立判别意义。综上结果提示,apoB基因可能是冠心病多基因中的一个易感基因。 相似文献
