目的:研究与胰岛素抵抗有关的三个基因的多态性GPR10(G-protein-coupled re-ceptor 10)(-62 G/A)、FOXC2(Forkhead box C2)(-512 C/T)、PGC1(Peroxisome proliferatoractivated receptorγcoactivator-1)(Thr394Thr)与2型糖尿病相关性.方法:利用等位基因专一性PCR的方法,对来自上海地区的正常人和肥胖的2型糖尿病患者进行基因分型,并通过X2比较基因型频率及等位基因频率在两组人群中是否存在显著性差异.结果:GPR10基因-62G/A多态性基因型频率在糖尿病人群和正常人群中的分布分别为:糖尿病人AA 14、AG 39、GG 34;正常人AA 2、AG 44、GG 42,具有显著性差异,等位基因频率分布为:糖尿病人A 67 G107、正常人A48、G 128,也有显著性差异.结论:GPR10基因-62G/A多态性可能与2型糖尿病相关,但该结果仍有待于进一步扩大样本量进行验证.  相似文献

目的:通过检测张家口地区45例健康女性和34例卵巢癌患者CYP1A1基因Ile/Val位点多态性,探讨Ile/Val位点多态性与卵巢癌发生的关系.方法:采用等位基因特异性扩增和PCR-RFLP技术,检测CYP1A1基因Ile/Val基因多态性.结果:健康女性Ile、Val等位基因频率为68.9%,31.1%;Ile/Ile,Ile/Val,Val/Val基因型分布频率为44.4%,48.9%,6.7%.卵巢癌患者Ile及Val等位基因的频率分别为63.2%,36.8%;其Ile/Ile,Ile/Val,Val/Val基因型分布频率分别为38.2%,50%,11.8%.结论:张家口地区健康人群和卵巢癌患者CYP1A1基因MspI位点均呈多态性分布,两者基因型分布频率存在显着性差异,说明卵巢癌发病率可能与CYP1A1基因型有关.  相似文献

目的:探讨胰岛素样生长因子1(insulin-like growth factor1,IGF1)基因单核苷酸多态性(SNP)与埃及水牛生长性状的关联性。创新点:发现IGF1基因多态性对埃及水牛的生长性状、血液生化指标和基因表达有显著影响,并为埃及水牛的选育提供重要的分子遗传标记。方法:以200头埃及水牛为试验对象,对其IGF1基因的多态性进行检测,并分析其与生长性状、血液生理生化指标和基因表达的相关性。结论:在水牛IGF1基因多态性分析中,发现两个新的SNP位点(G64A和G280A)分别分布在外显子1和外显子4的非编码区。不同基因型的统计分析表明,G64A和G280A位点的GG基因型水牛个体分别在3~6月龄和6~9月龄的体重(BW)和平均日增重(ADG)均显著高于其他基因型(P0.05)。这两个SNPs位点的组合基因型产生了三种单倍体GG/GG、AG/AG和AA/AA。在3~12月龄的水牛个体中,单倍体基因型与BW和ADG存在显著关联(P0.0001)。纯合的GG/GG单倍体基因型水牛生长性能优于其他水牛。两个SNP位点与半腱肌中IGF1和IGF1R的mRNA水平以及IGF1血清浓度水平相关。此外,GG/GG单倍体水牛表现出较高的mRNA和血清浓度水平。综上所述,这两种SNP位点G64A和G280A可作为埃及水牛生长性状选育的重要分子遗传标记。  相似文献

采用PCR-RFLP方法分析了太湖猪(二花脸)群体中生长激素(growth hormone,GH)、心脏脂肪酸结合蛋白(heart fatty acid-binding protein,H-FABP)和肌细胞生成素(Myogenin,MyoG)基因的多态性.结果表明:在检测的二花脸群体中,GH基因经DraⅠ、MspⅠ酶切无多态性,ApaⅠ酶切出现2个等位基因、3种基因型,基因型频率呈AA>AB>BB的趋势,等位基因A的基因频率大于等位基因B.MyoG基因在PCR1/MspⅠ位点上,基因型AB占据一定的优势;在PCR2/MspⅠ位点上,等位基因A占据绝对的优势,基因型频率趋势为AA>AB>BB.H-FABP基因的HinfⅠ位点上,等位基因h的基因频率有一定的优势.经χ2适合性检验表明,GH、MyoG、H-FABP3个基因在太湖猪(二花脸)群体中的分布均符合Hardy-Weinberg平衡(P>0.05).  相似文献

目的:研究猪PIT-1基因多态性及其对生长性状的遗传效应,旨在寻找改良猪生长性能的分子标记。方法:采用PCR-RFLP方法检测猪PIT-1基因的单核苷酸多态性(SNP),并分析基因多态性与生长性状的关联性。结果:猪PIT-1基因的Rsa I酶切位点存在多态性。其中,杜洛克猪未检测到AA型,BB基因型频率高于AB型,等位基因B频率高于等位基因A,SNP处于低度多态(PIC0.25);淮猪新品系未检测到BB型,AA基因型频率高于AB型,等位基因A频率高于等位基因B,SNP处于低度多态(PIC0.25);杜洛克猪×淮猪新品系未检测到BB型,AB基因型频率高于AA型,等位基因A频率高于等位基因B,SNP处于中度多态(0.25PIC0.5)。关联分析表明,PIT-1基因型对猪平均日增重和饲料转化率的遗传效应都未达到显著水平(P0.05)。结论:杜洛克猪、淮猪新品系、杜洛克猪×淮猪新品系PIT-1基因Rsa I酶位点都存在多态性,但PIT-1基因对猪生长性状没有显著影响。  相似文献

Sedum alfredii Hance has been identified as zinc (Zn) and cadmium (Cd) co-hyperaccumulator. In this paper the relationships of Zn or Cd hyperaccumulation to the generation and the role of H2O2 in Sedum alfredii H. were examined, The results show that Zn and Cd contents in the shoots of Sedum alfredii H. treated with 1000 μmol/L Zn2 and/or 200 μmol/L Cd2 increased linearly within 15 d. Contents of total S, glutathione (GSH) and H2O2 in shoots also increased within 15 d, and then decreased. Total S and GSH contents in shoots were higher under Cd2 treatment than under Zn2 treatment. However, reverse trends of H2O2 content in shoots were obtained, in which much higher H2O2 content was observed in Zn2 -treated shoots than in Cd2 -treated shoots. Similarly, the microscopic imaging of H2O2 accumulation in leaves using H2O2 probe technique showed that much higher H2O2 accumulation was observed in the Zn2 -treated leaf than in the Cd2 -treated one. These results suggest that there are different responses in the generation of H2O2 upon exposure to Zn2 and Cd2 for the hyperaccumulator Sedum alfredii H. And this is the first report that the generation of H2O2 may play an important role in Zn hyperaccumulation in the leaves. Oar results also imply that GSH may play an important role in the detoxification of dissociated Zn/Cd and the generation of H2O2.  相似文献

Objective: To test the influence of homocysteine on the production and activation of matrix metalloproteinase-2 (MMP-2) and tissue inhibitors of matrix metalloproteinase-2 (TIMP-2) and on cell migration of cultured rat vascular smooth muscle cells (VSMCs). Also, to explore whether rosuvastatin can alter the abnormal secretion and activation of MMP-2 and TIMP-2 and migration of VSMCs induced by homocysteine. Methods: Rat VSMCs were incubated with different concentrations of homocysteine (50–5 000 μmol/L). Western blotting and gelatin zymography were used to investigate the expressions and activities of MMP-2 and TIMP-2 in VSMCs in culture medium when induced with homocysteine for 24, 48, and 72 h. Transwell chambers were employed to test the migratory ability of VSMCs when incubated with homocysteine for 48 h. Different concentrations of rosuvastatin (10−9–10−5 mol/L) were added when VSMCs were induced with 1 000 μmol/L homocysteine. The expressions and activities of MMP-2 and TIMP-2 were examined after incubating for 24, 48, and 72 h, and the migration of VSMCs was also examined after incubating for 48 h. Results: Homocysteine (50–1 000 μmol/L) increased the production and activation of MMP-2 and expression of TIMP-2 in a dose-dependent manner. However, when incubated with 5 000 μmol/L homocysteine, the expression of MMP-2 was up-regulated, but its activity was down-regulated. Increased homocysteine-induced production and activation of MMP-2 were reduced by rosuvastatin in a dose-dependent manner whereas secretion of TIMP-2 was not significantly altered by rosuvastatin. Homocysteine (50–5 000 μmol/L) stimulated the migration of VSMCs in a dose-dependent manner, but this effect was eliminated by rosuvastatin. Conclusions: Homocysteine (50–1 000 μmol/L) significantly increased the production and activation of MMP-2, the expression of TIMP-2, and the migration of VSMCs in a dose-dependent manner. Additional extracellular rosuvastatin can decrease the excessive expression and activation of MMP-2 and abnormal migration of VSMCs induced by homocysteine.  相似文献

Objective: To evaluate the effects oftramadol on the proinflammatory responses in a rat model of incisional pain by investigating its effects on nociceptive thresholds and serum interleukin-6 (IL-6) and IL-2 levels. Methods: Forty-two male Sprague-Dawley (SD) rats scheduled for plantar incision were randomly divided into 7 groups (n=6 in each group). Rats in Group 1 receiving general anesthesia with no incision were served as control; At 30 min before skin incision, Groups 2~5 were given 5 ml normal saline or 1, 10, and 20 mg/kg tramadol, respectively, intraperitoneally (i.p.); Group 6 received 10 mg/kg tramadol after operation; Group 7 received 10 mg/kg tramadol before incision, followed by 200 μg/kg naloxone after operation. Mechanical allodynia was measured by electronic von Frey filament to evaluate the nociceptive thresholds 1 h before incision, and 1 h and 2 h after operation. Serum IL-6 and IL-2 levels were measured by enzyme-linked immunosorbent assay (ELISA) 2 h after operation. Results: Mechanical thresholds decreased significantly and serum IL-6 level increased significantly after operation in Group 2 compared with control (P<0.01), and these changes were reversed respectively by tramadol in a dose-dependent manner (P<0.05 and P<0.01, respectively). IL-2 level remained unchanged after operation in Group 2, but decreased in Group 3 (P<0.05), then gradually returned to the normal level in Groups 4 and 5. The intraperitoneally injected tramadol (10 and 20 mg/kg) produced a potent and dose-dependent antinocicptive effect on the lesioned paw. The antinocicptive effects of tramadol were partially an-tagonized by naloxone (200 μg/kg), suggesting an additional non-opioid mechanism. Conclusion: The results suggest that tramadol could be a good choice for the treatment of pain under the conditions that immunosuppression may be particularly contraindicated.  相似文献

Objective

Numerous studies examining the relationship between human epidermal growth factor receptor 2 (HER-2) overexpression and survival in patients with colorectal cancer (CRC) have yielded controversial results. We therefore performed a meta-analysis more precisely to estimate its prognostic value.

Methods

Published studies investigating the effect of HER-2 overexpression on CRC survival were identified; the hazard ratios (HRs) and their corresponding 95% confidence intervals (95% CIs) were pooled in terms of disease-specific or overall survival.

Results

Eleven studies were included in the meta-analysis. The pooled data showed that HER-2 overexpression was negatively related to CRC survival (HR=1.10, 95% CI: 0.77–1.44). Subgroup analyses regarding test method and study quality also demonstrated little association between HER-2 overexpression and CRC survival (HR=0.89, 95% CI: 0.50–1.29; HR=0.90, 95% CI: 0.43–1.37, respectively).

Conclusions

Regardless of several limitations, our study suggested that HER-2 overexpression probably had little impact on CRC survival.  相似文献

目的:研究环氧合酶-2(COX-2)对卵巢癌细胞迁移和耐药性的影响及其机制。创新点:本研究发现COX-2对卵巢癌发生发展有一定的促进作用,可以通过上皮间质转化(EMT)途径促进卵巢癌细胞的迁移和顺铂(CDDP)耐药。其抑制剂塞来昔布(CXB)能起到协同抗癌的效果。方法:CCK-8检测CXB和CDDP对SKOV3和ES2细胞的毒性作用。划痕实验评估COX-2对卵巢癌细胞迁移的作用。蛋白质免疫印迹(western blot)和聚合酶链式反应(PCR)检测EMT相关基因和蛋白的表达水平。结论:COX-2可以通过EMT促进卵巢癌细胞迁移和CDDP耐药;CXB可以起到抑制作用,与CDDP协同抗癌。COX-2可以作为卵巢癌治疗的一个潜在靶点。  相似文献
