目的探讨大肠癌组织中CD105和Ki-67的表达及其意义。方法采用免疫组织化学PV-9000法检测50例大肠癌组织和10例正常大肠黏膜组织中CD105和Ki-67的表达,检测肿瘤内微血管密度(MVD)以及计算增殖指数(PI)。结果Ki-67和CD105在大肠癌组织中呈高表达(51.34±24.44,16.44±6.33),与正常黏膜组织中的表达(21.10±8.66,4.00±1.25)相比较,差异有极显著的统计学意义(P〈0.01)。Ki-67的表达与大肠癌的分化程度、临床分期和淋巴结的转移有关,有显著统计学意义(P〈0.05)。MVD与大肠癌的分化程度、临床分期有关(P〈0.05),但与淋巴结转移无关(P〉0.05)。Ki-67在大肠癌高微血管密度组的表达(68.28±16.91)明显高于低微血管密度组(30.59±13.90),差异非常显著(P〈0.01),Ki钾的表达与MYD呈正相关(r=0.28,P〈0.01))。结论CD105和Ki-67在大肠腺癌的发生发展中起着重要作用,并且同时检测两者的表达对大肠腺癌的抗血管治疗和判断其预后有一定应用价值。  相似文献

目的:分析探讨乳腺癌组织中CD105的表达与ER、PR、C-erbB-2的表达及同侧腋淋巴结转移的关系.方法:应用免疫组织化学二步法,检测60例乳腺癌组织和20例乳腺良性病变组织(纤维腺病和纤维腺瘤)中的CD105、ER、PR、C-erbB-2的表达,用MVD值(微血管密度)表示CD105的表达量;用阳性细胞比例表示ER、PR、C-erbB-2的表达程度.结果:(1)在60例乳腺癌和20例乳腺良性病变组织中,CD105的表达量(MVD值)在乳腺癌组明显高于良性病变组(P〈0.01);在乳腺良性病变组织——纤维腺病和纤维腺瘤之间的CD105的表达量(MVD值)无显著性差异(P〉0.05).(2)CD105的表达量(MVD值)与淋巴结转移(P〈0.05)呈正相关;而与年龄、肿瘤大小、分化程度、ER、PR、C-erbB-2无相关性(P〉0.05).结论:(1)乳腺癌中CD105的表达量(MVD值)可作为一种反映血管新生程度的指标;血管新生与乳腺癌的演进有密切关系;(2)乳腺癌中CD105的表达量(MVD值)对判断乳腺癌的预后有重要临床意义.  相似文献

目的:探讨卵巢上皮癌组织中环氧合酶-2(COX-2)表达及其与微血管密度(MVD)的关系。方法:应用免疫组织化学技术检测65例卵巢上皮癌组织和18例正常卵巢组织中COX-2的表达,并以抗CD34抗体标记微血管内皮细胞测定MVD。结果:卵巢上皮癌组织COX-2表达阳性的比例为73.8%,显著高于正常卵巢组织COX-2表达阳性的比例11.1%(P<0.01)。COX-2阳性表达的卵巢上皮癌组织中的MVD(57.8±18.6)明显高于阴性表达者(37.4±11.2)(P<0.01)。结论:COX-2在卵巢上皮癌组织中高表达,COX-2可促进肿瘤血管生成。  相似文献

目的:探讨甲状腺乳头状癌(PTC)组织中基质金属蛋白酶9(MMP-9)和E-钙黏附素(E-cadherin)表达与肿瘤侵袭转移的关系.方法:应用微波-链霉菌素-生物素(微波-LSAB)法检测69例PTC组织MMP-9和E-cadherin蛋白的表达.结果:MMP-9和E-cadherin阳性率分别为68.1%(47/69)和30.4%(21/69),MMP-9阳性表达与PTC的侵袭转移呈正相关(P<0.05);E-cadherin阳性表达与PTC的侵袭转移呈负相关(P<0.05).结论:MMP-9高表达和E-cadherin低表达可能参与PTC的发生发展过程,尤其与侵袭转移密切相关,联合检测对于PTC预后的判断具有临床指导意义.  相似文献

目的:检测CD44v6mRNA和HSP70mRNA在胃癌组织中的表达,探讨CD44v6mRNA和HSP70mRNA在胃癌发生发展中的作用.方法:利用核酸原位杂交技术对39例胃癌组织进行检测.结果:CD44v6mRNA和HSP70mRNA在胃癌组织表达的阳性率都为61.54%(24/39),CD44v6mRNA和HSP70mRNA在高分化管状腺癌与低分化管状腺癌中的表达都有显著性差异(P<0.05),但HSP70mRNA在胃癌组织的表达与性别、年龄、浸润深度及淋巴结有无转移都无关(P>0.05),而CD44v6mRNA与淋巴结有无转移有关(P<0.05).结论:CD44v6mRNA和HSP70mRNA在胃癌组织都有较高的表达,两者均可作为评估胃癌预后的指标.  相似文献

目的探讨介入化疗对直肠癌血管内皮生长因子表达(VEGF)及微血管计数(MVD)的影响.方法对34例直肠癌患者介入化疗前后的肿瘤组织,用CD34对VEGF表达进行测定,并进行MVD计数.结果本组患者介入化疗前及后4至5周VEGF阳性表达率分别为61.7%(21/34)、32.4%(11/34),MVD计数明显降低,差异均有显著性意义(P<0.01);结论介入化疗能降低直肠癌组织VEGF的表达,减少MVD计数,提示介入化疗可能调节直肠癌的分化程度,减少术后转移.  相似文献

目的:探讨CD15s和E-钙黏蛋白(E-cadherin)在肝细胞癌组织中的表达及其与肿瘤侵袭转移和患者预后的关系。方法:应用免疫组织化学方法,检测120例肝细胞癌组织中CD15s和E-cadherin蛋白表达,结合随访资料进行分析。结果:CD15s和E-cadherin在肝细胞癌中阳性表达率分别为39.2%和44.2%。CD15s阳性表达的肝细胞癌转移率高(P〈0.05),分化程度及患者5年生存率低(P〈0.05) E-cadherin阳性表达的肝细胞癌转移率低(P〈0.05),分化程度及患者5年生存率高(P〈0.05)。CD15s表达与E-cadherin表达呈显著负相关(P〈0.05)。结论:CD15s和E-cadherin表达与肝细胞癌转移和患者生存期密切相关,联合检测CD15s和E-cadherin蛋白表达可作为判断肝细胞癌预后有价值的参考指标。  相似文献

目的:检测环氧化酶-2(COX-2)及CD34在宫颈鳞癌组织中的表达,并探讨二者在宫颈鳞癌形成及进展中的意义.方法:应用二步法免疫组化检测系统检测94例宫颈鳞癌组织、26例宫颈上皮内瘤变(CIN)及5例正常宫颈上皮组织标本中COX-2及CD34的表达.结果:COX-2在CIN和鳞癌组织中阳性率分别为53.85%,77.66%.随着分化程度的降低,COX-2表达阳性率显著提高,在中、低分化鳞癌中的表达显著高于高分化鳞癌(P<0.01);有淋巴结转移组明显高于无淋巴结转移组(P<0.01).MVD表达随着病理学分级的增高而明显增高,在Ⅲ级为(46.21±9.07);MVD在有淋巴结转移组(42.27±12.36)显著高于无转移组(26.58±7.15)(P<0.01).COX-2和CD34在正常宫颈上皮组织中均呈阴性表达;两者表达与患者年龄及临床分期均无关(P>0.05).结论:人宫颈鳞癌组织COX-2表达增强及MVD增高,COX-2可能通过促进肿瘤血管生成而促进宫颈鳞癌的进展,在其发生、进展中可能具有重要作用.  相似文献

探讨RhoC和基质金属蛋白酶-9(MMP-9)蛋白在结直肠癌发生发展中的作用。选取新鲜结直肠癌标本作为实验组,正常新鲜结肠粘膜组织作为对照组,应用流式细胞术分别检测RhoC和MMP-9的表达,检测其在不同临床病理特征之间的关系。结果表明,RhoC和MMP-9在结直肠癌组织中高表达(P<0.05),且其表达均与Dukes分期、浸润深度及炎细胞浸润相关(P<0.05),MMP-9的表达与结直肠癌的淋巴结转移密切相关(P<0.05),RhoC和MMP-9在结直肠癌中的表达呈正相关(r=0.3430,P=0.0420)。流式细胞术联合检测RhoC和MMP-9蛋白的表达有望成为判断预后的重要指标。  相似文献

This study aimed to explore the molecular mechanism in tumor invasion and metastasis. The expression of matrix metalloproteinase-2, −9 (MMP-2, MMP-9), tissue inhibitor-1 of matrix metalloproteinase (TIMP-1), cell adhesion molecule 44 variant 6 (CD44v6), HER2/neu and p53 was investigated in 154 patients with head and neck squamous cell carcinoma (SCC) by ABC and ImmunoMax immunohistochemical method. Their clinical relevance and correlation were analysed. The expression of MMP-2, MMP-9, TIMP-1, CD44v6, HER2/neu and p53 was found in cancer cells in 87.01%, 85.71%, 68.18%, 98.05%, 55.19% and 50.65% cases respectively. Linear regression and correlation analysis revealed that there was close positive relationship (P<0.05) between the expression of MMP-2 and MMP-9, TIMP-1 and CD44v6, HER2/neu and MMP-9, MMP-2 and p53. Up-regulation of MMP-2 was accompanied by advanced T stage (P<0.01). There was also a trend of MMP-2 expression being related with tumor metastasis. Increased expression of HER2/neu was found in patients with tumor recurrence (P<0.05). The expression of TIMP-1 was higher in laryngeal cancer than that in pharyngeal cancer, and higher in keratinizing and non-keratinizing SCC than that in basaloid SCC (P<0.05). These findings suggested that MMP-2 and MMP-9, HER2/neu and MMP-9, MMP-2 and p53 had a coordinate function in aggression of tumor; that MMP-2 had a more important function than MMP-9 in tumor invasion and metastasis; and that HER2/neu might serve as a biomarker for poor prognosis in HNSCC. Methods: Tumor and corresponding normal mucosal tissue, collected from 39 gastric carcinoma patients, were examined by immunohistochemistry and RT-PCR for the expression of CD97 and CD55. Results: CD97stalk was strongly stained on scattered tumor cells or small tumor cell clusters at the invasion front of gastric carcinomas. The expression of CD97stalk was frequently observed in tumors of stage Ⅰ and T1 gastric carcinoma patients. The expression of CD97stalk between Stage Ⅰ and Stage Ⅱ, Ⅲ, Ⅳ specimens showed significant difference (P<0.05), between T1 and T2, T3, T4 specimens also showed significant difference (P<0.05). Specimens with tumor invasion depth limited in mucosa of T 1 specimens showed higher positive CD55 expression than specimens with the same tumor invasion depth in T2, T3, T4 specimens, the expression of CD55 between T1 and T2, T3, T4 specimens was significantly different (P<0.05).There was strong correlation between the distribution patterns of CD97stalk and CD55 on tumor tissues (r=0.73, P<0.05). Signet ring cell carcinomas frequently contained strong CD97stalk and CD55-staining. Conclusions: Our results suggest that CD97stalk is probably involved in the growth, invasion and aggressiveness of gastric carcinomas by binding its cellular ligand CD55. CD97stalk and CD55 could be useful as molecular markers for prognosis and therapy of gastric carcinoma patients.  相似文献

目的:检测胃癌组织中DNA损伤修复酶(O6-甲基鸟嘌呤-DNA甲基转移酶O6-methylguanine DNA methyltransferase, MGMT)的表达变化,探讨其表达和胃癌临床病理特征之间的关系,进一步用于指导临床化疗方案的制定.方法:采用免疫组化方法(SABC法)检测55例胃癌标本中MGMT的表达情况.结果:MGMT在胃癌组织中的阳性表达率为43.64%(24/55),其阳性表达与年龄、病理类型有关,而与性别、分化程度、侵犯深度、淋巴结转移的相关性不明显.结论:MGMT对胃癌化学治疗方案的制定有指导作用.  相似文献

目的:探讨HSP_(27)在消化道癌中的表达状况和意义。方法:收集食道癌、胃癌和大肠癌标本168例,用SP免疫组织化学方法染色。结果:HSP在食道癌、胃癌和大肠癌中的表达率分别为66.04%、37.04%和34.43%;在食道癌中高分化组阳性率(75%)明显高于低分化组(38.46%)。HSP表达与性别、年龄、癌侵润深度、淋巴有无转移的相关性不显著。结论:HSP在食道、胃、大肠癌中表达率依次下降,HSP表达率与癌分化程度有一定相关性。  相似文献

本研究应用较灵敏的SP免疫组织化学方法,对80例胃癌组织进行bcl-2、c-myc基因蛋白的检测,并对两种基因与临床病理的关系进行了研究。结果发现,bcl-2基因蛋白在胃癌的阳性率为46.3%。高、中分化腺癌阳性率为59.4%,显著高于低、未分化腺癌(37.5%);其表达与Merkel分型有关,乳头状腺癌显著高于低分化腺癌;在lauren分型中,肠型胃癌为59.4%,显著高于弥漫型(37.5%);在Ming'S分型中,膨胀型(52.3%)显著高于浸润型胃癌(23.5%)。在伴淋巴结转移的胃癌中,bcl-2的表达率(38.6%)显著低于不伴淋巴结转移者(65.2%)。c-myc基因蛋白在胃癌的阳性率为67.5%,其表达率在低、未分化腺癌中为72.5%,显著高于粘液腺癌(25.0%);而粘液结节型腺癌(25%)明显低于粘液细胞型腺癌(81.8%)和管状腺癌(71.4%)。在膨胀型胃癌的表达率(73.0%)显著高于浸润型(47.1%)。c-myc的表达率与淋巴结转移明显关系。bcl-2、c-myc基因表达与患者性别、年龄、肿瘤发生部位、浸润深度无明显相关性。  相似文献

目的 :探讨基质金属蛋白酶 (matrixmetalloprotein .ase - 2MMP - 2 )在膀胱癌和阴茎癌中的表达及意义。方法 :采用免疫组化法对MMP - 2在 4 2例膀胱癌和 4 5例阴茎癌中的表达进行研究。结果 :MMP - 2在膀胱癌中的阳性表达率为 2 8.5 7% (12 / 4 2 ) ,在阴茎癌中的阳性表达率为 5 7.78% (2 6 / 4 5 ) ;在膀胱移行细胞癌和腺癌中的阳性率分别为 2 3.6 8% (9/ 38)、75 .0 0 % (3/ 4 ) ,两者比较有显著性差异 (P <0 .0 1)。在阴茎癌高分化组阳性率为 4 5 .16 % ,中分化组阳性率为 85 .7% ,两组比较有显著性差异 (P <0 .0 5 )。结论 :MMP - 2在移行上皮癌与鳞状上皮癌中均有表达 ,其在鳞癌中的表达高于移行细胞癌 ,腺癌细胞的高表达提示腺癌细胞的浸润性高于移行细胞癌。  相似文献

目的:探讨热休克蛋白70(HSP70)在人乳腺癌和胃癌中的表达及其意义。方法:应用免疫组织化学的方法对64例乳腺癌和55例胃癌组织中的HSP70进行检测。结果:(1)HSP70在乳腺癌和胃癌组织中的表达分别为78.13%.61.82%。(2)乳腺癌中HSP70的表达与乳腺癌的病理类型.淋巴结转移情况及年龄未发现显著差异(P〉0.05)。(3)胃癌中HSP70的表达与肿瘤浸润深度、淋巴结转移有密切关系(P〈0.05)。结论:HSP70在乳腺癌和胃癌中均呈较高水平表达.其中在胃癌中的表达与肿瘤浸润深度、恶性程度有密切关系,HSP70在乳腺癌和胃癌的发生发展中起重要作用。HSP70可能成为乳腺癌,胃癌的诊断指标之一。  相似文献
