Microcystis aeruginosa FACHB 905 was used as a model organism to study the effects of small water clusters(SWCs) on the growth and microcystin(MC) production of toxic cyanobacteria.The results showed that SWCs were able to stimulate the growth of Microcystis aeruginosa,which resulted in increased cell numbers and higher specific growth rates after a 30-d treatment.The cell morphology indicated that Microcystis aeruginosa was in a better state of growth,and it was more prone to divide in SWCs than in normal water clusters.The SWCs treatment up-regulated MC synthesis and exudation in 10 d in Microcystis aeruginosa,and the intra-cellular MC content decreased after the 20th day subsequently.Moreover,the cellular photosynthetic pigment contents were temporarily stimulated by SWCs.A possible reason is that SWCs stimulated the growth by promoting photosynthesis,whereas the increased MC production was relevant to pigment contents.  相似文献

6-aminopenicillanic acid (6-APA) crystals obtained under different physical and chemical conditions of the solutions may present different habits. The habits of diamond-shaped plates are desirable compared with other habits of 6-APA crystals. To obtain ideal 6-APA crystals, the effects of the mixed solvents and additives on 6-APA crystal habits were investigated. Ethanol or acetone was used as the organic solvent, and impurities existing in the ,6-APA purification process were used as the additives. 6-APA growth habits were changed when the concentrations of ethanol, acetone or phenyl acetic acid were increased to exceed their critical concentration. The observed results show that the dominant face on 6-APA crystals was identified to be , {020}, but the overall habit was controlled by the relative growth rates of the : 101 and {002} faces. Crystal growth rates and habits can be appreciably changed by specific adsorption of additives on crystal faces. In some cases solvent molecules can act in a similar way and may be regarded as bulk additives. The effects of additives and organic solvents on 6-APA crystal habits were the results of adsorption effect,which fitted the experimental results quite well.  相似文献

Oil contaminated soil was collected from Huangpu River-Yangtze River estuary wetland, with the aim of isolating oil-degrading microorganisms and evaluating their ability to degrade diesel. Three bacterial strains were discovered and identified by sequencing their 16S rDNA genes, two were Pseudomonas and one was Alcaligcnes. The proper growth conditions of each bacterium were measured and presented for diesel biodegradation. Biodegradation assays revealed that the degradation rates of three bacterial strains were 42.5%, 14.6% and 15.9% in 7 d respectively. They all play an important role on the nalkanes within the range of C16-C25 components of diesel. The results indicated that the oil-degraders can adapt to degrade diesel. The bacterial strains can be used in wetland diesel pollution control.  相似文献

The treatment of relapsing primary nephrotic syndrome in children   总被引:2,自引:0,他引:2  
Objective: To explore better therapy and reduce the rate of re-relapse of primary nephritic syndrome in children who had been treated with corticosteroids but relapsed. Methods: Eighty relapsers were enrolled from Jan. 1994 to Apr. 2000, who were randomly divided into two groups. The treatment group (n=39) had been treated with tripterysium glucosides for three months, with the control group (n=41) members were treated with cyclophosphmide (CTX) by intermission intravenous pulse, with total dose of CTX not being more than 150 mg/kg. Prednisone, meanwhile, was given to both groups. The total treatment period of prednisone was prolonged by 12-18 months. Results: After following up for 3-7 years, the re-relapse rates of both groups were observed. The re-relapse rate of the treatment group was 28.2% to 29.3% in the CTX-controlled group. The re-relapse rates between two groups were almost similar, and with no observed significant difference (P>0.05). The side effect of tripterysium glucosides was less than that of CTX. Conclusion: For the treatment of relapsing nephritic syndrome in children, the combination of tripterysium glucosides and prolonged corticosteroid therapy is as effective as the regimen of CTX plus prolonged use of prednisone.  相似文献

The aim of this study was to compare differences in imaging quality at different injection rates of gadobutrol(Gadovist? 1.0M) for dynamic supra-aortic imaging by contrast-enhanced magnetic resonance angiography(CEM RA). In 6 healthy volunteers,dynamic CEM RA examinations were performed twice at a 7 d interval with a gadobutrol dose of 0.1 mmol given to 1 kg body mass. For the two examinations,the contrast medium was injected at rates of 1.5 mL/s and 2.0 mL/s,followed by 20 mL normal saline injected at the same rate. Using a GE Signa 3.0T M R apparatus and the time-resolved imaging of contrast kinetics(TRICKS) technique,scanning was performed continuously for 15 phases at a time resolution of about 2.4 s. Vascular reconstruction was undertaken by multiplane reconstruction and maximum intensity projection techniques. Two radiologists experienced in CEM RA diagnosis subjectively evaluated the contrast effect,vascular detail,and pseudo-shadow of all images,and graded them via a 5-point scale. With the two drug injection rates,the image quality,peak signal-to-noise ratio(SNR) and contrast-to-noise ratio(CNR) were compared for imaging of the infra-aortic arch,brachiocephalic trunk,subclavian artery(left/right),common carotid artery(left/right),internal carotid artery(left/right),and external carotid artery(left/right). Characteristics of the time-signal-intensity(T-S-I) curves were also analyzed at the two injection rates. All studies were performed successfully,without any complications. There was good image quality with both gadobutrol injection rates of 1.5 mL/s and 2.0 mL/s,and subjective scores for image quality,SNR and CNR were not significantly different between the two rates(P 0.05). The time-to-peak(TTP) from the hand dorsal vein injection site to the carotid artery ranged from 16.0 s to 27.1 s(mean 21.7 s ±3.4 s) in the 6 subjects,and was also not significantly different between the two drug injection rates(P 0.05). We conclude that dynamic CEM RA examination of the supra-aortic arch with gadobutrol injection at a rate of 1.5 mL/s was feasible,and satisfactorily reflected vascular shape and dynamic blood flow information.  相似文献

Objective: To estimate the oxidative stress and oxidative damage induced by abnormal free radical reactions in IgA nephropathy (IgAN) patients' bodies. Methods: Seventy-two IgA N patients (IgANP) and 72 healthy adult volunteers (HAV) were enrolled in a random control study design, in which the levels of nitric oxide (NO) in plasma, lipoperoxide (LPO) in plasma and in erythrocytes, and vitamin C (VC), vitamin E (VE) and β-carotene (β-CAR) in plasma as well as the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in erythrocytes were determined with spectrophotometric mothods. Results: Compared with the HAV group, the averages of NO in plasma, and LPO in plasma and in erythrocytes in the IgANP group were significantly increased (P<0.0001), while those ofVC, VE and β-CAR in plasma as well as those of SOD, CAT and GPX in erythrocytes in the IgANP group were significantly decreased (P<0.0001). Linear correlation analysis showed that with the increase of the values of NO, and LPO in plasma and in erythrocytes, and with the decrease of those ofVC, VE, β-CAR,SOD, CAT and GPX in the IgAN patients, the degree of histological damage of tubulointerstitial regions was increased gradually (P<0.0001); and that with the prolongation of the duration of disease the values of NO, and LPO in plasma and erythrocytes were increased gradually, while those of VC, VE, β-CAR, SOD, CAT and GPX were decreased gradually (P<0.005). The discriminatory correct rates of the above biochemical parameters reflecting oxidative damage of the IgAN patients were 73.8%-92.5%, and the correct rates for the HAV were 70.0%-91.3% when independent discriminant analysis was used; and the correct rate for the IgAN patients was increased to 98.8%, the correct rate for the HAV was increased to 100% when stepwise discriminant analysis was used. The above biochemical parameters' reliability coefficient (alpha) were used to estimate the oxidative damage of the IgAN patients as 0.8145, the standardized item alpha=0.9730, F=53273.5681, P<0.0001. Conclusions: A series of free radical chain reactions caused serious pathological aggravation in the IgANP' bodies, thus resulting in oxidative damage in their bodies. In treating IgANP, therefore, it is necessary that suitable dose antioxidants should be supplemented to them so as to alleviate the oxidative damage in their bodies.  相似文献

A bacterial strain WY047 was isolated from fermented grains and the bacterium was identified as Bacillus amyloliquefaciens, based on morphological, biochemical, and physiological tests, and analysis of 16S rRNA and gyr A sequences. The culture supernatant of WY047 demonstrated inhibition ofa wide spectrum ofbacteria(Gram positive and Gram negative) and fungi. Nine pairs ofprimers were designed and six genes( bmyD, fenA, hag, ituA, mrsA, and tasA) ofantimicrobial substances were detected by PCR, one ofwhich was isolated by 80% ammonium sulfate precipitation, D201 resin anion-exchange chromatography, and Sephadex G-75 filtration column. The purified peptide was estimated to be 35,207 Da and identified as flagellin by MALDI-TOF mass spectrometry. Another four antimicrobial substances were extracted with methanol and identified as iturin A, fengycin, bacillomycin D, and mersacidin through the liquid chromatography–mass spectrometry(LC–MS) method. The sixth possible peptide encoded by tas A could not be isolated in this study; however, the broader spectrum suggested huge application prospects.  相似文献

The protected tetrapeptide, N-o-Ns-N ( Me ) -Val-N ( Me ) -Val-N ( Me ) -Val- N ( Me ) -Phe- OtBu, was prepared from L-valine and L-phenylalanine. Ted-butyl acetate and HClO4 were used to protect carbonyl group, o-nitrobenzenesulfonyl chloride and triethyl amine were used to protect amino group, and N-alkylation was finished with iodomethane. Then the protected amino acid was turned into acid chloride which was taken as coupling reagent. After 14 steps, such as protection, alkylation, deprotection and coupling, the protected tetrapeptide was obtained with a yield of 26.9%. The structures of intermediates and target compound were identified with NMR spectra and high resolution mass spectra.  相似文献

The study aimed to assess and compare the values prevalent among the students and teachers of Universities in Bangladesh, Japan, USA and Germany. The sample consisted of 480 students and 236 teachers. The sample included 120 undergraduate students Japan; 120 undergraduate students from Bangladesh; 120 undergraduate students from USA, and 120 undergraduate students from Germany. The faculty sample included 60 teachers from Japan, 60 teachers from Bangladesh; 60 teachers from USA, and 56 teachers from Germany. To identify the value preferences of the individuals a list of 10 values, pro-social, achievement, power over others, security, self direction, otherworldliness, fatalism, narcissism, inner directed, and conservative, based on previous values studies by Singh and Parek were prepared. The first five values were identified in previous studies as functional (Singh, 1975) and other five were identified as dysfunctional (Parek, 1988) in the context of national development of Asian cultures. The results revealed that Bangladeshi students held stronger preferences for values identified as functional values than for those identified as dysfunctional. Japanese students indicated stronger preferences for the values identified as functional values except narcissism. The American students indicated a preference for three of the five values identified as functional but also ranked narcissism and other worldliness as third and fifth preferred values respectively. German student showed a preference for functional values, except narcissism which they also ranked as third. Bangladeshi teachers' preferences for functional values were higher than dysfunctional values. Japanese teachers indicated a preference for functional values except narcissism. American teachers preferred functional values except other worldliness. German teachers' value preferences were also functional, except for narcissism which they ranked as fifth. Students and teachers in the four countries sampled indicate preferences for values identified as functional with few exceptions. This research suggests that value preferences among university students and teachers are more similar than different, suggesting a homogenizing effect (Boli, 2005) on human values.  相似文献

Objective: The aim of this retrospective investigation was to explore the influence of femoral osteoporosis on short-term curative effects ofcementless hip arthroplasty and to evaluate the femoral metaphyseal bone mineral density (BMD) for femoral osteoporosis in order to guide prosthesis choice and rehabilitation. Methods: We performed 127 total arthroplasty operations between June 1999 to February 2003 and investigated 49 cementless hip replacements with the Metalcancellous cementless Lubeck Ⅱ system being used in all hips. There were twenty men and twenty-nine women whose mean age at the time of the operation was 60 years (range, 52-81 years). The patients were divided into osteoporosis or normal groups according to the femoral metaphyseal BMD measured preoperatively. The average duration of follow-up was 30 months (range, 8-52 months). We evaluated all of the patients from a clinical standpoint with use of a standard-terminology questionnaire with respect to the short-term curative effects and patients' satisfaction. Hip pain status and functional ability were important indicators of treatment efficacy. Results: Harris hip score and patients' satisfaction in femoral osteoporosis patients who underwent noncemented hip arthroplasty were lower (P=0.004, P=0.03) while the incidence of thigh pain was higher (P=0.03) than the patients with non-osteoporosis. Conclusion: The higher incidence of pain, as well as the decrease in function experienced by the patients in osteoporosis group, supports the case that cementless arthroplasty is not a better choice for those patients and that we had better select prosthesis based on the femoral metaphyseal BMD.  相似文献

Objective: To identify the mutations of iduronate-2-sulfatase (IDS) gene, to reveal its mutation features, and to establish a basis for genetic counseling and prenatal gene diagnosis of Hunter syndrome. Methods: Urine glycosaminoglycans (GAGs) assay, PCR and DNA sequencing were performed to detect mutation of IDS gene of the patient and his parents. Results: The result showed that the patient was: DS( ), HS( ), KS(-), CS(-), and that both of his parents were negative. A frame-shift deletion mutation (1062 del 16) was identified in exon 7 of the patient's IDS gene. His parents' genotypes were normal. Conclusion:The patient's mutation was not inherited by his parents but a novel one. The mutation probably altered the primary structure and tertiary structure of IDS enzyme protein remarkably and lowered the activity of IDS enzyme greatly. Therefore it is supposed to be the direct cause of the disorder.  相似文献

Atrazine is a selective herbicide used in agricultural fields to control the emergence of broadleaf and grassy weeds. The persistence of this herbicide is influenced by the metabolic action of habituated native microorganisms. This study provides information on the occurrence of atrazine mineralizing bacterial strains with faster metabolizing ability. The enrichment cultures were tested for the biodegradation of atrazine by high-performance liquid chromatography(HPLC) and mass spectrometry. Nine cultures JS01.Deg01 to JS09.Deg01 were identified as the degrader of atrazine in the enrichment culture. The three isolates JS04.Deg01, JS07.Deg01, and JS08.Deg01 were identified as efficient atrazine metabolizers. Isolates JS04.Deg01 and JS07.Deg01 produced hydroxyatrazine(HA) N-isopropylammelide and cyanuric acid by dealkylation reaction. The isolate JS08.Deg01 generated deethylatrazine(DEA), deisopropylatrazine(DIA), and cyanuric acid by N-dealkylation in the upper degradation pathway and later it incorporated cyanuric acid in their biomass by the lower degradation pathway. The optimum pH for degrading atrazine by JS08.Deg01 was 7.0 and 16S rDNA phylogenetic typing identified it as Enterobacter cloacae strain JS08.Deg01. The highest atrazine mineralization was observed in case of isolate JS08.Deg01, where an ample amount of trzD mRNA was quantified at 72 h of incubation with atrazine. Atrazine bioremediating isolate E. cloacae strain JS08.Deg01 could be the better environmental remediator of agricultural soils and the crop fields contaminated with atrazine could be the source of the efficient biodegrading microbial strains for the environmental cleanup process.  相似文献

A 24-membered ring macrolide compound,macrolactin A has potential applications in pharmaceuticals for its anti-infectious and antiviral activity.In this study,macrolactin A was produced by a marine bacterium,which was identified as Bacillus subtilis by 16S ribosomal RNA(rRNA) sequence analysis.Electrospray ionization mass spectrometry(ESI/MS) and nuclear magnetic resonance(NMR) spectroscopy analyses were used to characterize this compound.To improve the production,response surface methodology(RSM) involving Box-Behnken design(BBD) was employed.Faeces bombycis,the main by-product in sericulture,was used as a nitrogen source in fermentation.The interactions between three significant factors,F.bombycis,soluble starch,and(NH4)2SO4 were investigated.A quadratic model was constructed to fit the production and the factors.Optimum medium composition was obtained by analysis of the model.When cultivated in the optimum medium,the production of macrolactin A was increased to 851 mg/L,2.7 times as compared to the original.This study is also useful to find another way in utilizing F.bombycis.  相似文献

This study investigated characteristics of bifurcation and critical buckling load by shape imperfection of space truss,which were sensitive to initial conditions.The critical point and buckling load were computed by the analysis of the eigenvalues and determinants of the tangential stiffness matrix.The two-free-nodes example and star dome were selected for the case study in order to examine the nodal buckling and global buckling by the sensitivity to the eigen buckling mode and the analyses of the influence,and characteristics of the parameters as defined by the load ratio of the center node and surrounding node,as well as rise-span ratio were performed.The sensitivity to the imperfection of the initial shape of the two-free-nodes example,which occurs due to snapping at the critical point,resulted in bifurcation before the limit point due to the buckling mode,and the buckling load was reduced by the increase in the amount of imperfection.The two sensitive buckling patterns of the numerical model are established by investigating the displaced position of the free nodes,and the asymmetric eigenmode greatly influenced the behavior of the imperfection shape whether it was at limit point or bifurcation.Furthermore,the sensitive mode of the two-free-nodes example was similar to the in-extensional basis mechanism of a simplified model.The star dome,which was used to examine the influence among several nodes,indicated that the influence of nodal buckling was greater than that of global buckling as the rise-span ratio was higher.Besides,global buckling is occurred with reaching bifurcation point as the value of load ratio was higher,and the buckling load level was about 50%-70% of load level at limit point.  相似文献

Transition metal-doping could effectively extend the light response range of TiO _2 photocatalysts from the ultraviolet(UV)to the visible region.Co-doped brookite titanium dioxide(Co–TiO_2)photocatalysts were synthesized via the hydrothermal method with titanium tetrachloride as the raw material and cobalt chloride hexahydrate as the dopant.The prepared Co–TiO_2 photocatalysts were characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM),transmission electron microscopy(TEM),Raman spectroscopy,X-ray photoelectron spectroscopy(XPS)and UV–Vis diffuse reflectance spectroscopy(UV–Vis DRS).The photocatalytic activities of Co–TiO _2 photocatalysts were evaluated by photocatalytic degradation of isopropanol alcohol(IPA),a typical volatile organic compound(VOC),under visible light.The influences ofdifferent Co doping rates,initial concentrations of IPA gas and the amounts of photocatalyst addition were also studied.At the same time,the enhancement mechanism ofcobalt ions as a trap for photogenerated holes was discussed.Thus,we found the optimum doping rate,initial concentration of IPA gas and amount of photocatalyst to add.The results show that the mesoporous Co–TiO _2 photocatalysts possess smaller size particles,larger specific surface area,lower forbidden bandgap energy(Eg)and better photocatalytic activity than pure brookite TiO _2.When the doping of Co was 7% by mass,the initial concentration ofIPA gas was 1.0×10~(-6 )mol/L and the addition of Co–TiO_2 photocatalysts was 50 mg,the best photocatalytic activity was achieved.Furthermore,the degradation rate ofIPA was up to 91%,which shows great potential for waste water treatment.  相似文献

Due to the importance of biological safety in drinking water quality and the disadvantages which exist in traditional methods of detecting typical microorganisms such as Cryptosporidium and Giardia, it is necessary to develop an alternative. Particle counts is a qualitative measurement of the amount of dissolved solids in water. The removal rate of particle counts was previously used as an indicator of the effectiveness of a biological activated carbon (BAC) filter in removing Cryptosporidium and Giardia. The particle counts in a BAC filter effluent over one operational period and the effects of BAC filter construction and operational parameters were investigated with a 10 m^3/h pilot plant. The results indicated that the maximum particle count in backwash remnant water was as high as 1296 count/ml and it needed about 1.5 h to reduce from the maximum to less than 50 count/ml. During the standard filtration period, particle counts stay constant at less than 50 count/ml for 5 d except when influenced by sand filter backwash remnant water. The removal rates of particle counts in the BAC filter are related to characteristics of the carbon. For example, a columned carbon and a sand bed removed 33.3% and 8.5% of particles, respectively, while the particle counts in effluent from a cracked BAC filter was higher than that of the influent. There is no significant difference among particle removal rates with different filtration rates. High post-ozone dosage (〉2 mg/L) plays an important role in particle count removal; when the dosage was 3 mg/L, the removal rates by carbon layers and sand beds decreased by 17.5% and increased by 9.5%, respectively, compared with a 2 mg/L dosage.  相似文献

The protein encoded by CC chemokine receptor 7 (CCR7) is a member of the G protein-coupled receptor family. This receptor was identified as a gene induced by the Epstein-Barr virus (EBV), and is thought to be a mediator of EBV effects on B lymphocytes. This receptor is expressed in various lymphoid tissues and activates B and T lymphocytes. It has been shown to control the migration of memory T cells to inflamed tissues, as well as stimulate dendritic cell maturation. To map the CCR7 gene in chicken chromosome, a 6 000 rads chicken-hamster radiation hybrid panel (ChickRH6) was used. PCR of samples from ChickRH6 revealed that the location of CCR7 gene is linked to the maker SEQ0347 (6 cR away) with LOD score of 16.6 and that the marker SEQ0347 is located on chromosome 27 at 27 cR of RH (radiation hydrid) map. We compared the corresponding human mRNA sequence with the predicted coding sequence of chicken CCR7 gene, and found that the assembled contig shared a high percentage of similarity with that of the human gene.  相似文献

Phytoremediation effectiveness and remediation costs are driving factors of this project. Full utilization of plant resources after their being used for phytoremediation is an unsolved problem. GC/MS technique was used to investigate the volatiles of the flowers from Elsholtzia argyi (PFE 1 : Purple Flower Elsholtzia) and their variation (WFE: White Flower Elsholtzia),naturally growing in Pb/Zn mined area, and Elsholtzia argyi (PFE2: Purple Flower Elsholtzia), naturally growing in Jiuxi uncontaminated agriculture soil. Seventeen compounds constituting 86.88% of total essential oils were identified in PFE1, with 2,6-octadienoic acid,3,7-dimethyl-methyl ester being the main constituent (63.30%). Sixteen compounds accounting for 95.32% of total essential oils were identified in WFE, with caryophyllene being the main component (55.02%). Compared to PFE1, PFE2 contained lower level of 2,6-octadienoic acid,3,7-dimethyl-methyl ester (31.76%), which was the main constituent in the total essential oils of PFE2. Caryophyllene is the main ingredient of flavor. Elsholtzia ketone was identified in all the three Elsholtzia plants. It can be concluded that the selected Elsholtzia argyi plants can be exploited on their versatile uses as fragrances and antiseptics due to the perfume ingredient and antibacterial components existing in their essential oils.  相似文献
