A simple microfluidic 3D hydrodynamic flow focusing device has been developed and demonstrated quantitative determinations of quantum dot 525 with antibody (QD525-antibody) and hemagglutinin epitope tagged MAX (HA-MAX) protein concentrations. This device had a step depth cross junction structure at a hydrodynamic flow focusing point at which the analyte stream was flowed into a main detection channel and pinched not only horizontally but also vertically by two sheath streams. As a result, a triangular cross-sectional flow profile of the analyte stream was formed and the laser was focused on the top of the triangular shaped analyte stream. Since the detection volume was smaller than the radius of laser spot, a photon burst histogram showed Gaussian distribution, which was necessary for the quantitative analysis of protein concentration. By using this approach, a linear concentration curve of QD525-antibody down to 10 pM was demonstrated. In addition, the concentration of HA-MAX protein in HEK293 cell lysate was determined as 0.283 ± 0.015 nM. This approach requires for only 1 min determining protein concentration. As the best of our knowledge, this is the first time to determinate protein concentration by using single molecule detection techniques.  相似文献

The aim of the present study was to examine elbow joint extension power-load characteristics with and without a countermovement. Eight male participants performed maximal elbow extensions with and without a countermovement against different loads (0, 2.5, 5.0, 7.5, 10.0, 12.5, and 15.0 kg). Electromyographic activity of the lateral and long heads of the triceps brachii and the biceps brachii muscles was recorded. The average joint power in the concentric phase was significantly enhanced in the countermovement condition at all loads except for 0 kg. The optimal load for the maximal joint power was greater in the countermovement (7.5 kg) than in the no countermovement condition (5.0 kg). Electromyographic activity was unchanged over the intensities and conditions. Our results suggest that the optimal load for the maximal joint power depends on the type of action (i.e. with or without a counter-movement), and that the enhanced joint power in the countermovement condition is due primarily to the storage and utilization of elastic energy.  相似文献

This study looked into possible relationships between workplace organizational communication tactics, workers’ feelings of trust, and workers’ perceptions of procedural justice. Raw data was collected by questionnaires, and structural equation modeling (SEM) was used to reveal the relationships. The subjects in the study were Japanese (N = 417) and American (N = 591) workers, surveyed in a period running up to late May 2004. For both the Japanese and the Americans, perceived procedural justice was influenced positively by rational communication tactics, and negatively by hard communication tactics; and soft communication tactics did not relate to perceived procedural justice. In the case of Japanese workers, it was found that, after controlling for perceived procedural justice, soft, rational, and hard communication tactics affected trust in different ways: positively, positively, and negatively, respectively. Therefore, perceived procedural justice had a partially mediating effect on the relationships between rational and hard communication tactics and trust. In contrast, for the Americans, there were no direct relationships between communication tactics and trust; rather, there were indirect effects from rational and hard communication tactics, via perceived procedural justice as a full mediator. The influence of three communication tactics on trust was much stronger for Japanese workers than their American counterparts. On the other hand, the influence of hard and rational communication tactics on perceived procedural justice and that of perceived procedural justice on trust were much stronger for American workers than their Japanese counterparts. Based on the results, some academic and practical implications are presented.  相似文献

In microcirculation, red blood cells (RBCs) flowing through bifurcations may deform considerably due to combination of different phenomena that happen at the micro-scale level, such as: attraction effect, high shear, and extensional stress, all of which may influence the rheological properties and flow behavior of blood. Thus, it is important to investigate in detail the behavior of blood flow occurring at both bifurcations and confluences. In the present paper, by using a micro-PTV system, we investigated the variations of velocity profiles of two working fluids flowing through diverging and converging bifurcations, human red blood cells suspended in dextran 40 with about 14% of hematocrit level (14 Hct) and pure water seeded with fluorescent trace particles. All the measurements were performed in the center plane of rectangular microchannels using a constant flow rate of about 3.0 × 10−12 m3/s. Moreover, the experimental data was compared with numerical results obtained for Newtonian incompressible fluid. The behavior of RBCs was asymmetric at the divergent and convergent side of the geometry, whereas the velocities of tracer particles suspended in pure water were symmetric and well described by numerical simulation. The formation of a red cell-depleted zone immediately downstream of the apex of the converging bifurcation was observed and its effect on velocity profiles of RBCs flow has been investigated. Conversely, a cell-depleted region was not formed around the apex of the diverging bifurcation and as a result the adhesion of RBCs to the wall surface was enhanced in this region.  相似文献

This study compared 3- to 4-month-olds' recognition of previously unfamiliar faces learned in a moving or a static condition. Infants in the moving condition showed successful recognition with only 30 s familiarization, even when different images of a face were used in the familiarization and test phase (Experiment 1). In contrast, infants in the static condition showed successful recognition only when the familiarization duration was lengthened to 90 s and when the same image was used between the familiarization and test phase (Experiments 2 and 3). Furthermore, presentation of multiple static images of a face did not yield the same level of performance as the moving condition (Experiment 4). These results suggest that facial motion promotes young infants' recognition of unfamiliar faces.  相似文献

In this mixed methods research, the authors examine a unique type of small-scale event – a charity-affiliated sport event – and define and measure its social and charitable impacts as perceived by residents. Findings from interviews (N = 37) and surveys (N = 459) with residents indicated that the event’s social impacts can be defined by its capacity to develop social capital, enhance collective identity and pride, and promote sport, health, and well-being. Three types of charitable impacts also emerged, including empathy for cause, informational support, and tangible support. Of them, empathy for cause, which addresses a central social issue in the host community, had the strongest association with residents’ perceptions of social impacts. These results provide evidence of a variety of positive impacts that a charity-affiliated sport event has on a community, which can be used to bolster appeals for corporate sponsorship and government support to assist in event delivery.  相似文献

The dynamic behaviour of racetrack surfaces during hoof impact is a likely risk factor for racehorse injuries and fatalities. A track-testing device that simulates equine hoof impact was used to assess the effects of surface type (dirt, synthetic), surface depth (2, 3, 4 layers), test boundary area (929, 6,606, 11,381 cm2), harrowing, and impact angle (0°, 20° from vertical) on dynamic surface properties. Surfaces were tested within a laboratory track-in-a-box. Surface type and depth, boundary area, and harrowing significantly affected dynamic surface properties during impacts (P < 0.05). The synthetic surface was generally less stiff, and had 26–73% lower maximum impact forces and load rates, than the dirt surface. Harrowing decreased stiffness and differences between dirt and synthetic properties. Measured surface stiffness was larger for the smallest depth and boundary area, but most surface properties were not significantly different between the larger boundary areas. Surfaces were less stiff with angled impacts than vertical impacts.  相似文献

Extant U.S. research shows that when a persuader’s initial message is rebuffed, the next requesting message will tend to be ruder and more aggressive than the initial appeal. The robustness of these results has rarely been tested cross-culturally. Using conversational constraints theory, we further explicate implicit theories by investigating the perceived importance of constraints of re-requesting styles across two cultural-linguistic groups (i.e., Korean and American English speakers). Consistent with the “rebuff phenomenon,” results revealed that people rated the task constraint (“clarity”) as significantly more important, and the three face-related constraints (“concern for the other’s feelings,” “minimizing imposition,” and “avoiding negative evaluation”) as significantly less important for the second-attempt requesting than for the initial requesting. Some of these tendencies were more pronounced among American English speakers than among Korean speakers.  相似文献

We present a novel cell culture chip, namely, “inverting microwell array chip,” for cultivation of human induced pluripotent stem cells. The chip comprises a lower hydrogel microwell array and an upper polystyrene culture surface. We demonstrate the formation of uniform cellular aggregates in the microwell array, and after inversion, a culture with controlled aggregate size and geometrical arrangement on the polystyrene surface. Here, we report effects of cell concentrations on a cultivation sequence in the chip.  相似文献
