Oxygen and glucose supply is one of the important factors for the growth and viability of the cells in cultivation of tissues, e.g., spheroid, multilayered cells, and three-dimensional tissue construct. In this study, we used finite element methods to simulate the flow profile as well as oxygen and glucose supply to the multilayered cells in a microwell array chip for static and perfusion cultures. The simulation results indicated that oxygen supply is more crucial than glucose supply in both static and perfusion cultures, and that the oxygen supply through the wall of the perfusion culture chip is important in perfusion cultures. Glucose concentrations decline with time in static cultures, whereas they can be maintained at a constant level over time in perfusion cultures. The simulation of perfusion cultures indicated that the important parameters for glucose supply are the flow rate of the perfusion medium and the length of the cell culture chamber. In a perfusion culture chip made of oxygen-permeable materials, e.g., polydimethylsiloxane, oxygen is hardly supplied via the perfusion medium, but mainly supplied through the walls of the perfusion culture chip. The simulation of perfusion cultures indicated that the important parameters for oxygen supply are the thickness of the flow channel and the oxygen permeability of the walls of the channel, i.e., the type of material and the thickness of the wall.  相似文献

The present study examined the extent to which scores on the Dispositional Flow Scale-2 (DFS-2) could differentiate individuals who frequently experience flow characteristics in physical activity from those who do not. A total of 993 participants completed the Japanese version of the DFS-2. Latent class factor analysis (LCFA), which combines the strengths of both latent class analysis and factor analysis, was conducted on the DFS-2 responses. Six classes were identified through a series of LCFAs and the patterns of the item-average scores for the nine flow attributes were found to be parallel among these classes. The top two and bottom two classes (19.3% and 13.4% of the whole sample) were considered the groups who experience flow characteristics frequently and seldom, respectively. These results indicated that individuals who often experience flow attributes in physical activity could be differentiated from those who do not based on their DFS-2 scores.  相似文献

In rapid interceptive actions such as hitting a baseball, cricket ball or tennis ball, ball speed varies between trials, and players have to compensate the time lag by controlling the moment of movement onset and movement duration. Previous studies have found that these two variables can flexibly co-vary and are robustly influenced by target speed (i.e. velocity-coupling effect: faster movement for faster target). However, some studies reported an interpersonal variability in the timing control strategy and the relationship between the strategy and temporal accuracy in rapid interception is unclear. We used a baseball-simulated rapid interceptive task to assess this issue. Under relatively easy time constraints, there was a large interpersonal variability, and participants were distinctively divided into two groups: those who mainly modulated their movement duration and those who mainly controlled their movement onset. When the time constraint became severe, the second strategy shifted to the first strategy in most of the second group participants. In the both cases, being able to mainly control movement onset resulted in higher temporal accuracy. These results suggest that minimising the velocity-coupling effect is an important factor to achieve high temporal accuracy in rapid interception.  相似文献

When extinction is delayed very long, the superior resistance to extinction of the random schedule group relative to the alternating schedule group disappears (partial reinforcement delayed extinction effect, PRDE). Two experiments assessed the effects of reinforcement/nonreinforcement on Trial 1 on the PRDE. Following extended partial reinforcement acquisition training in a runway, rats received extinction training after a short (1-day) or long (23-day) retention interval. The schedules used in Experiment 1 were: a single-alternation (SA) schedule beginning each day with a rewarded (r) trial, for Group r-SA; an SA schedule beginning with a nonrewarded (n) trial, for Group n-SA; and a random (Rd) schedule, for Group Rd. The schedules and group names used in Experiment 2 were r-SA, Rd, and r-Rd. The results were that (1) rats given r-SA schedules yielded considerable resistance under delayed extinction, (2) those given Rd and r-Rd schedules showed a decline in resistance to extinction over a long retention interval, (3) those given the n-SA schedule showed relatively low resistance at both retention intervals, although retention deficit was not greater than in the case of the Rd schedule, and thus, (4) the PRDE was found in both experiments, although only weakly in Experiment 1. The results indicated that a regularly alternating reward pattern was a more important determinant than was type of reward on Trial 1 for the PRDE. The PRDE due to differential retention deficits among schedules is discussed on the basis of dual-process associative sequential mechanisms and cognitive rule-encoding mechanisms.  相似文献

Spheroid culture is a preferable cell culture approach for some cell types, including hepatocytes, as this type of culture often allows maintenance of organ-specific functions. In this study, we describe a spheroid microarray chip (SM chip) that allows stable immobilization of hepatocyte spheroids in microwells and that can be used to evaluate drug metabolism with high efficiency. The SM chip consists of 300-μm-diameter cylindrical wells with chemically modified bottom faces that form a 100-μm-diameter cell adhesion region surrounded by a nonadhesion region. Primary hepatocytes seeded onto this chip spontaneously formed spheroids of uniform diameter on the cell adhesion region in each microwell and these could be used for cytochrome P-450 fluorescence assays. A row of microwells could also be connected to a microchannel for simultaneous detection of different cytochrome P-450 enzyme activities on a single chip. The miniaturized features of this SM chip reduce the numbers of cells and the amounts of reagents required for assays. The detection of four cytochrome P-450 enzyme activities was demonstrated following induction by 3-methylcholantlene, with a sensitivity significantly higher than that in conventional monolayer culture. This microfabricated chip could therefore serve as a novel culture platform for various cell-based assays, including those used in drug screening, basic biological studies, and tissue engineering applications.  相似文献

Ski jumping flight posture was analyzed for achieving large flight distance on the basis of high-speed video images of the initial 40 m part of 120-m ski jumping flight. The time variations of the forward leaning angle and the ski angle of attack were measured from the video images, and the aerodynamic forces were calculated from the kinematic data derived from the images. Some correlations were investigated between the initial-speed corrected flight distance and such parameters as the angles of jumper, the initial transition time and the aerodynamic force coefficients. The result indicated that small body angle of attack was a key for large flight distance in the initial phase of flight because of small drag force, and that the most distinctive fault of beginners was too large body angle of attack and ski angle of attack leading to aerodynamic stall. Too small drag force does not give an optimal condition for large flight distance because the lift force is also too small. The ratio of the lift to the drag was larger than 0.95 for advanced jumpers.  相似文献

We investigated whether pigeons are able to discriminate color photographs of male and female pigeons, using a categorical discrimination procedure. In Experiments 1 and 2B, 10 out of 14 pigeons learned the discrimination. Of these, 5 pigeons showed transfer to novel stimuli, demonstrating the categorical nature of the trained discrimination. Experiment 3 showed that the discriminative behavior was based primarily on the body, as opposed to the head and the neck region. In 1 out of 3 pigeons, the discriminative behavior was maintained by the black-and-white photographs. The results suggest that some pigeons have the ability to discriminate the sex of conspecifics without behavioral cues.  相似文献

This study evaluated baseball pitching accuracy using a variety of parameters to quantify pitching errors and analysed the validity of the accuracy measurements by comparing the outcomes of two small groups of pitchers. Several professional (n = 5) and high school (n = 8) pitchers threw 30 pitches each, including 20 fastballs and 10 breaking balls. To assess pitching accuracy, pitch locations relative to the catcher’s mitt (as the target) were evaluated with various parameters, including major/minor radius, an area of 95% confidence ellipse, absolute error, constant error and pitch location trajectory. Compared to the high school pitchers, the professional pitchers exhibited shorter major and minor radii in their 20 fastball pitches (p < 0.05), more accurate control in the lateral direction (p < 0.05), and shorter pitch location trajectories (p < 0.05). The evaluation methods presented in this study can objectively assess pitching accuracy and may thus provide useful coaching feedback with visual information.  相似文献
