黄豆籽苗胺氧化酶的制备及其特性研究
摘    要:本文报告了黄豆籽苗胺氧化酶(SSAO)的制备方法及其酶学特性。每100 g 豆芽可制备5ml 黄色酶液,以腐胺为底物比活性为20μmol/60min/mg 蛋白质。酶液经聚丙烯酰胺凝胶电泳可分成5个组分,其电泳位置相当于血清白蛋白、α和β球蛋白,经 Sephadex G-100凝胶层析分离可得到2个组分,98%的酶活性分布在第一组分中。酶液经4℃保存,活性渐降、7天降9%,19天降22%、80天降64%。如在-20℃保存,活性下降更快,7天降28%,19天降42%,80天降51%。用腐胺作底物的米氏常数(K_m)为0.26mmolL~(-1)。当酶量为0.10ml,作用时间为5分钟时,最大线性范围达200nmol;如果及应时间增至15分钟最大线性范围降为100nmol。本酶的最适 pH 为8—9。尿素、肌酐、组氨酸精氨酸和赖氨酸对本酶无干扰作用.

The Preparation of Soybean Seedling Amine Oxidase and the Studying of its Enzymatic Characteristics

Abstract:This paper reports on the preparation of soybean seedling amine oxidase and the studying of its enzymatic characteristics.5ml yellowish enzyme solu- tin is obtained from 100g soybean seedling.The specific activity of enzyme with the putrescine as a substrate is 20 micromole per 60 min per mg protein. This enzyme solution were separated into 5 sections by polyacrylamide gel electrophoresis (PAGE).2 sections are obtained by chromatography with Sephadex G-100.98% of enzyme present in the first section.The activity of enzyme stored at 4℃ decreasd 9% after 7 days,22% after 19 days and 64% after 80 days.With putrescine as substrate the K_m of enzyme is 0.26 mmol per liter.The limit linearity is 200 nmol.The optimum pH is 8—9.Urea. creatinine,histidine,arginine and lysine do not inetrference with the action of the enzyme.
