摘    要:利用抗HGH单克隆抗体建立了HGH的RIA和IRMA。方法学鉴定表明,这两种测定方法的灵敏度高(RIA为1.2ng/ml,IRMA为2ng/ml)、特异性强(与HPL及HPRL均无交叉反应)、重复性好。同时,还将这两种方法与HGH常规抗血清RIA进行了比较。

Abstract:A mixture of 3 monoclonal antibodies(McAbs)against human growth hor- mone(HGH)has been employed successfully to establish a radioimmunoassay (RIA)for measurement of serum HGH levels. A new one-step, two-site im- munoradiometricassay(IRMA)has been developed by using McAb-1 as the solid- phase antibody and McAb-4 as the labeled antibody. There are two non-over- lapping antigenic sites for McAb-1 and McAb-4. The results indicate that the two immunoassays are highly sensitive (RIA. 1.2ng/ml; IRMA: 2ng/ml), strongly specific(no cross-reactivity with human placental lactogen and human prolactin)and excellent repeated, and these McAbs are useful in RIA and IRMA of HGH in clinical diagnosis. Finally, the two methods are compared with the RIA using polyclonal antibodies.
