摘    要:以壳聚糖和海藻酸钠为载体原料,制备了载胰岛素的缓释剂,进行了缓释性能研究,利用正交实验方法,优化了制备工艺条件。结果表明,最佳制备工艺为:壳聚糖与海藻酸钠的质量配比为1:2,加胰岛素量为28mL(浓度为1.0mg/mL),混合溶液(担载胰岛素后)反应时间为30min;采用最佳工艺条件制备的缓释剂在模拟人工胃液中溶出4h时溶液中胰岛素浓度为26.6μg/m L。

Abstract:Chitosan and the sodium alginate used as carrier materials, insulin slow- release formulation was prepared, the release function was studied and the preparation conditions was optimized using the orthogonal experimental method. The findings show that the optimal preparation process is as follows: the mass ratio of chitosan and sodium alginate as 1 : 2, plus the amount of insulin 28mL (concentration of 1.0mg/mL) ; reaction time of mixed solution (supported by insulin) is 30min; insulin concentration is 26.6 g/mL in 4h dissolution solution of the slow- release formulation being in the artificial gastric juice.
