| 植物组织培养实验中存在的问题及其解决办法 |

| 摘 要： | 植物组织培养操作过程中的经验积累和技术的掌握是很重要的。在实验过程中常常出现下列问题:培养基母液配置时容易出现钼酸钠难溶解,母液的贮存过程中产生沉淀或有结晶析出,培养基高压蒸气灭菌后出现灭菌不彻底,组织培养中出现褐化及玻璃化现象。通过改进母液的配制方法、培养基高压灭菌时采用二次排气法,在培养基中,添加有机物、抑制剂等可解决实验出现的问题。

| Abstract: | Experiences and skills are crucial in the tissue culture. The existing issues are: infusible sodium molybdate inthe culture medium solution; separation by crystallization and precipitation from the culture medium solution; inexhaus-tive sterilization after high pressure steam sterilization; browning and vitrification in the culture. The issues can be solvedby modifying the solution, adopting twice exhausting air in steam sterilization process, and adding organic materials and(or) inhibitors in the culture medium. |
