选择性分离放线菌的土壤处理方法研究

摘    要:为了提高从土壤中分离放线菌的效率,解决土壤中杂菌对放线菌分离的污染问题.采用涂布平板法研究了土壤分散和预处理对放线菌的作用,结果表明采集的土样在28℃下风干10d后,用1%胆酸钠、Na+离子交换树脂和PEG作为化学分散剂制备土壤悬液(10-1),在40℃下,加6%酵母膏+0.05%SDS(5mmol/L磷酸缓冲液)用韦林氏搅拌器和振荡机振荡1h,平板分离放线菌的效果最佳.

Abstract:In order to imprve the efficiency of isolateing actinomycete from soil soil and solve the problem of contam intation when actinomycete were isolated,the effect of pretreatment and dispersion by plate paint isolation methods.The results showed that soil samples collected from air-dried at 28 ℃ for 10d,with 1% sodium cholate,Na+ ion exchange resin and PEG as a chemical dispersant to prepare the soil suspensions(10-1),At 40 ℃,plus 6% yeast extract +0.05% SDS(5mmol / L phosphate buffer) with Weilin Shi mixer and local oscillator oscillation 1h,the effect of isolated actinomycete is best.
