银杏叶总黄酮对人肝癌HepG2细胞增殖与凋亡的影响

摘    要:目的探讨银杏叶总黄酮对体外培养的人肝癌细胞HepG2增殖与凋亡的影响。方法将银杏叶总黄酮作用于体外培养的人肝癌细胞HepG2,MTT法检测其对HepG2细胞增殖的影响,缺口末端核苷标记(TUNNEL)法检测其对HepG2细胞凋亡的影响。结果银杏叶总黄酮对体外培养的人肝癌细胞HepG2的增殖效率下降,使凋亡细胞数增加(P〈0.01),且呈剂量依赖效应。结论银杏叶总黄酮对体外培养的人HepG2细胞增殖有抑制作用,并能诱导细胞凋亡。

Abstract:Objective To investigate the effect of Ginkgo biloba leaf flavonoids on proliferation and apoptosis of human liver cancer cell line HepG2 in vitro. Methods Human liver cancer cells line,HepG2 cultured in vitro,were treated with different concentrations of the flavonoids,and then the proliferation and apoptosis of the cells was detected by MTT and TUNNEL assay. Results MTT assay showed that Ginkgo biloba leaf flavonoids inhibited the proliferation of HepG2 cells line in a dose-dependent manner( P 〈0.01). And the apoptotic rate of the cells was increased ( P 〈 0.01 ) . Conclusion Ginkgo biloba leaf flavonoids could inhibit the cell proliferation and promote apoptosis of human hepatoma cell line HepG2.
