黑曲霉Aspergillus nigerS菌株所产β-葡糖苷酶 的纯化和酶学性质

摘    要:利用黑曲霉Aspergillus niger S菌株, 通过液体发酵的方法, 得到了一种可同时高效水解大豆异黄酮糖苷成大豆异黄酮,水解白藜芦醇苷成白藜芦醇的糖基水解酶.该酶经硫酸铵分级沉淀、离子交换层析及分子排阻色谱纯化得单一条带, 经SDS聚丙烯酰胺凝胶电泳测得该酶的表观分子量约为165ku 1u=1.660540×10-27kg。 ,比通常所报道的传统β-葡糖苷酶都大,该酶的最适反应温度为50℃; 最适pH值为5~10.

Abstract:A high activity glucosidase, which can hydrolyze glycosides, was isolated from Aspergillus niger S strain. The enzyme was purified to one spot after ammonium sulfate precipitation, ion exchange &; size exclusion chromatography and its molecular weight was about 165ku, which was quite differ from other reported glucosidase. The optimum temperature of this enzyme was 50℃ &; the optimum pH was pH 5~10. Based on the HPLC result, this enzyme can transform glycone to aglycone of isoflavones &; resveratrol with high specific activity.
