摘    要:目的:探索不同运动方式对生长期大鼠TGFβ/BMP信号通路相关分子表达及骨生长的影响。方法:将生长期大鼠随机分成4组:对照组(C)、游泳组(S)、跑台组(R)和下跳组(D),进行6周的不同方式的运动,利用RT-PCR、west-bloting检测生长板TGFβ/BMP信号通路的m RNA和蛋白表达,利用染色技术对生长板不同层的软骨细胞进行测量和计数。结果:胫骨、股骨长度R组和D组都非常显著性增加(P<0.01);生长板的肥大层宽度只有D组非常显著性增加(P<0.01);静息层占生长板总宽度百分比R组和D组的非常显著性下降(P<0.01);生长板的肥大层占生长板总宽度百分比R组和D组都非常显著性增加(P<0.01);生长板的肥大软骨细胞数量R组的显著性增加(P<0.05),D组的非常显著性增加(P<0.01)。BMP2 mRNA的表达水平R组和D组都非常显著性上调(P<0.01),D组与R组相比,也显著性上调(P<0.05);Smad1 mRNA的表达水平只有D组显著性上调(P<0.05);Smad5 mRNA的表达水平R组和D组都显著性上调(P<0.05);Smad8 m RNA的表达水平R组和D组都非常显著性上调(P<0.01);TGFβ1 mRNA的表达水平R组和D组都非常显著性下降(P<0.01);Smad2 mRNA的表达水平与只有D组显著性下降(P<0.05)。蛋白检测发现,BMP2蛋白的表达水平R组和D组都非常显著性增加(P<0.01);Smad1蛋白的表达水平R组和D组显著性增加(P<0.05),与S组相比,R组和D组非常显著性增加(P<0.01);TGFβ1蛋白的表达水平R组和D组的非常显著性降低(P<0.01)。结论:跑台运动和跳跃运动通过TGFβ/BMP信号通路促进生长期大鼠生长板软骨细胞的增殖和肥大,促进胫骨和股骨的增长,且效果优于游泳运动。

Abstract:Objective:To explore the effects of different exercise modes on the expression of related molecules of TGFβ/BMP signaling pathway and bone growth in growing rats.Methods:Growing rats were randomly divided into four groups:Control group(C),swimming group(S),running group(R)and down jump group(D),which were exercised in different ways for six weeks.The expression of mRNA and protein in TGFβ/BMP signaling pathway was detected by RT-PCR and western blotting.The chondrocytes in different layers of growth plate were measured and counted by section staining.Results:The length of tibia and femur increased significantly in group R and D(P<0.01);the width of hypertrophic layer of growth plate increased significantly only in group D(P<0.01);the percentage of resting layer to total width of growth plate in group R and D decreased significantly(P<0.01);and the percentage of hypertrophic layer to total width of growth plate in group R and D was very significant(P<0.01).The mast chondrocyte of growth plate increased significantly in group R(P<0.05)and group D(P<0.01).In this study,BMP2 expression level significantly increased in both R and D groups(P<0.01),and also in D group compared with R group(P<0.05);Smad1 expression level significantly increased only in D group(P<0.05);Smad5 mRNA expression level in R group and D group significantly increased(P<0.05);Smad8 expression level in R group and D group significantly increased(P<0.05);The expression level of TGFβ1 in group R and D decreased significantly(P<0.01),while the expression level of Smad2 in group D decreased significantly(P<0.05).The protein level of BMP2 protein increased significantly in group R and D(P<0.01);the expression level of Smad1 protein increased significantly in group R and D(P<0.05).Compared with group S,the expression level of TGFβ1 protein increased significantly in group R and group D(P<0.01);the expression level of TGFβ1 protein decreased significantly in group R and group D(P<0.01).Conclusion:Treadmill exercise and jumping exercise can promote the proliferation and hypertrophy of growth plate chondrocyte and the growth of tibia and femur in growing rats through TGFβ/BMP signaling pathway,and the effect is better than swimming exercise.
