| 摘 要： | 目的 :通过ENU诱发子代大鼠脑肿瘤的模型 ,对子代鼠脑肿瘤发生前、后p2 1基因的变化进行连续地观察 ,探讨抑癌基因 p2 1与胶质瘤发生、发展的关系。方法 :( 1)通过对孕18d大鼠腹腔注射亚硝基乙基脲 (ENU ,75mg/kg)的方法 ,建立子代鼠脑肿瘤模型 ;( 2 )应用免疫组化的方法 ,对子代鼠 90d、12 0d和 15 0d鼠脑 p2 1蛋白表达进行观察。 结果 :p2 1蛋白的表达减弱或消失 ,见于子代鼠出生后 90d、12 0d和 15 0d ,特别是 90d组 ,且 15 0d组中肿瘤内与瘤周组织之间有统计学差异。结论 :异常 p2 1蛋白的表达存在于肿瘤发生前、发生早期及发生后各阶段 ,进一步说明p2 1WAF1/CIP1发生突变、其表达减弱或消失是脑胶质瘤起源的重要原因。p2 1基因的功能失常可能是导致基因链活动紊乱的因素之一 ,而肿瘤的形成还可能需要其它因素参与。

| Abstract: | Objective: To study the relationship between p21 WAF1/CIP1 gene and glioma, we made a series of observation on p21 protein expression of the ENU-induced brain tumor in rats. Methods: (1) The animal model was established by exposing pregnant rats to carcinogen with intraperitoneally injecting one dose of N- Nitroso- ethylurea ( ENU, 75mg/kg) ; (2) The p21 protein was examined by immunohistochemistry stain in the brain slices of offspring rats on their 90, 120, and 150 day after their birth. Results: The reduction or disappearance of p21 protein expression was found in all the groups, especially in the 90-day groups. There were statistically significant differences between intra-tumor tissues and tumor surrounding tissues. Conclusion: As this experiment provided basis of different tumor developing stages, the results of this experiment showed that the reduction or disappearance of p21 protein expression happened in the pretumor period, the early stage and the peak period of tumor incidence. It suggested that disfunction of p21 is significant factor in tumorigenesis and may be one of the factors related to the disfunction of gene chains. |
