| 摘 要： | 设计DTT还原胰岛素,HPLC再现A、B链开链反应过程的生物化学实验。筛选出最适还原反应条件,最佳HPLC分离条件。通过质谱(MALDI-TOF-MS)分析确定了产物的结构。实验操作简便,结果稳定,再现性好,通过本实验学生能更好地理解和掌握HPLC的分析原理、操作方法及其在生命科学研究中的重要意义。

| Abstract: | This article designed an experiment where insulin was first reduced and unfolded by DTT,and then HPLC was used to represent the A and B chains.It developed the most probable conditions for the reduction reaction,found the best HPLC isolation method,and made a confirmation of each sample using MALDI-TOF-MS.The experiment is easy to perform,the result is invariable,and in addition,it has a desired reproducibility.It is helpful for college students to have a better understanding of the HPLC principles,to master the ways to operate it,and the importance of its role in life science research. |
