大鼠力竭运动后恢复过程中胃肠5-羟色胺免疫活性细胞的变化

摘    要:目的:探讨大鼠力竭运动后恢复过程中胃肠5-羟色胺免疫活性细胞(5-HTIR细胞)的变化.方法:以力竭游泳大鼠为模型,取恢复过程中的胃窦和十二指肠,用免疫组织化学SP法检测5-HT,用图像分析系统测定胃肠5-HTIR细胞数及平均灰度.结果:1)胃窦5-HTIR细胞的数量分别在力竭游泳后1h和24h时出现了两个峰值(P<0.05),阳性细胞平均灰度也相应下降,但只有恢复24h时的平均灰度有统计学意义(P<0.05).恢复48h时,实验组阳性细胞的数量和平均灰度已接近正常水平.2)十二指肠5-HTIR细胞的数量分别在力竭游泳后1h和24h时也出现了两个峰值,阳性细胞的平均灰度明显下降(P<0.05).恢复48h时,实验组阳性细胞数量仍明显低于对照组(P<0.05),但平均灰度已接近正常水平.结论:力竭运动后恢复过程中胃肠5-HTIR细胞处于活跃的合成和分泌状态,可能通过5-HT的合成和释放来参与应激反应,参与机体生理功能的调节.

Abstract:Objective:To investigate the changes of 5-HT immunoreactive cells (5-HTIR cells) in rat gastrointestinal tract during the recovery course following exhaustive swimming exercise. Method: Rats were divided into two groups: exhaustive exercise and resting control. Sinuses ventriculi and duodenum were taken from each group to detect the expression of 5-HT by immunohistochemistry, and followed with analysis by cell image analysis system. Result (1) The number of 5-HTIR cells of sinuses ventriculi reached peaks at recovery 1h and 24h after exhaustive swimming; the average gray level of 5-HT decreased accordingly, but only the value at recovery 24h had statistical significance (P<0.05) compared with that of the control group. At recovery 48h, the 5-HTIR cell numbers and the average gray level of 5-HT in experimental groups returned to normal levels. (2) similar to sinuses ventriculi tissue, the number of 5-HTIR cells of duodenum also reached peaks at recovery 1h and 24h after exhaustive swimming (P<0.05), the average gray level of 5-HTIR cells decreased significantly (P<0.05). At recovery 48h, the number of 5-HTIR cells had obvious difference compared with control group (P<0.05), but the average gray level of 5-HT had approached the normal level. Conclusion:Gastrointestinal tract 5-HTIR cells could take part in stress reaction and regulate physiological function by synthesis and release of 5-HT.
