三七总皂苷对力竭运动后大鼠大脑皮质胶质纤维酸性蛋白表达的影响

摘    要:目的:研究三七总皂苷(Panax Notoginseng Saponins)对一次性力竭游泳运动后大鼠大脑皮质星形胶质细胞胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响。方法:实验动物随机分为空白对照组、生理盐水组和三七总皂苷组。适应性喂养1周后,运动前30 min分别腹腔注射生理盐水和三七总皂苷,进行一次性力竭运动,分别在力竭运动后即刻和24 h灌注取材,采用免疫组织化学检测大脑皮质GFAP蛋白的表达,并记录游泳时间。结果:GFAP的阳性表达位于星形胶质细胞的细胞浆和细胞膜。生理盐水组在力竭即刻出现GFAP的表达增强,显著高于空白对照组和三七皂甙组(P<0.05),24 h后生理盐水组仍见GFAP的表达,而三七总皂苷组仅见少量GFAP的表达,二者表达未见显著差异(P>0.05);三七皂苷组与生理盐水组相比延长了大鼠的游泳时间(P<0.05)。结论:一次性力竭运动的即刻引起大鼠大脑皮质星形胶质细胞GFAP表达增强,提示与运动性中枢性疲劳的发生有关,三七总皂苷可以抑制力竭运动后星形胶质细胞GFAP的高表达,提示三七总皂苷能够延缓运动性中枢疲劳的发生,促进疲劳的消除。

Abstract:Objective:To investigate the influences of panax notoginseng saponins on glial fibrillary acidic protein(GFAP) in rats cerebral cortex after exhaustive swimming.Methods: Rats are selected and divided randomly into control group,physiological saline solution group(PSS group) and panax notoginseng saponins group(PNS group).All the rats are fed adaptively for 1 week.Rats in PSS group and PNS group are injected(IP) physiological saline solution and physiological saline solution respectively 30 min before swimming,and then forced to one-time exhaustive swimming.Materials are drawn from rats in the instant time and 24h later with 4% paraformaldehyde by filling.Cerebral cortex is gained to perform FAPimmunohistochemical staining.We recorded the swimming time.Results: The positive expression of GFAP locates in cell plasm and cell membrance.The expression of GFAP in PSS group in the instant time is stronger than that in PNS group and control group(P<0.05);24hrs later there is no difference of between PSS and PNS.The rats in PNS group swim longer than that in PSS group(P<0.05).Conclusion: One-time exhaustive swimming can cause special high expression of GFAP in cerebral cortex immediately which indicates that it has relation with occurrence of central fatigue.Moreover,panax notoginseng saponins can control the high expression of GFAP after exhaustive swimming,which indicates that panax notoginseng saponins can postpone the occurrence of central fatigue and release the central fatigue.
