摘    要:目的:观察一次性离心运动及针刺对大鼠股四头肌MyoD与Myogenin的影响,探讨针刺在骨骼肌损伤修复中的作用。方法:96只雄性SD大鼠随机编为安静(C)、安静针刺(CA)、运动(E)与运动针刺4组(EA),E与EA组完成一次性跑台运动(坡度为-16°);采用实时荧光定量PCR与免疫印迹法检测MyoD和Myogenin mRNA与蛋白表达。结果:CA组MyoD mRNA表达120 h显著高于C组(P0.05),蛋白表达24 h、48 h显著高于C组(P0.05);CA组Myogenin mRNA表达48 h、120 h显著高于C组(P0.05),蛋白表达48 h后均显著高于C组(P0.05)。EA组MyoD mRNA表达12 h至72 h显著高于E组(P0.05),蛋白表达12 h、24 h显著高于E组(P0.05);EA组Myogenin mRNA表达均显著高于C组(P0.05),12 h、48 h显著高于E组(P0.05);蛋白表达12 h、120 h显著高于E组(P0.05)。结论:针刺正常骨骼肌、一次性离心运动及运动后针刺均能上调MyoD、Myogenin mRNA与蛋白表达,且针刺能快速增高运动后骨骼肌MyoD和Myogenin的蛋白表达。

Abstract:Abstract: Purpose: This study examined the effects of acute eccentric exercise and acupuncture on MyoD and Myogenin of quadriceps muscle in rats, and investigated the effect of acupuncture on injury repair in skeletal muscle. Methods: Ninety-six male SD rats were divided into four groups: control (C), control plus acupuncture (CA), exercise (E), exercise plus acupuncture (EA) groups. E and EA groups underwent a single bout of eccentric exercise on a treadmill with -16o. mRNA and protein expression of MyoD and Myogenin were detected by real-time quantitative PCR and Western blot. Results: The mRNA expression of MyoD in CA group was significantly higher than that in C group at 120h (P < 0.05), and its protein expression in CA group was significantly higher than that in C goup at 24h and 48h (P < 0.05). Compared with C group, mRNA expression of Myogenin in CA group increased significantly at 48h and 120h (P < 0.05), and protein expresson in CA group increasd significantly at 48h (P < 0.05). Compared with E group, mRNA expression of MyoD in EA group was significantly higher from 12h to 72h (P < 0.05), and protein expresson in EA group was significantly higher at 12h and 24h (P<0.05). mRNA expression of Myogenin in EA group was higher than that in C group (P < 0.05), and higher than in E group at 12h and 48h (P < 0.05). Protein expression of Myogenin in EA group was higher than that in E group at 12h and 120h (P < 0.05). Conclusion: A single bout of eccentric exercise, acupuncture before and after exercise can up-regulate mRNA and protein expression of MyoD and Myogenin. Acupuncture after exercise can quickly raise MyoD and Myogenin protein expression in skeletal muscle.
