针刺对运动疲劳大鼠骨骼肌线粒体形态和游离Ca2+的影响

摘    要:目的:从肌细胞线粒体损伤的角度,探讨针刺抗运动性疲劳的效应与机制。方法:将30只SD大鼠分为针灸组、空白组和运动疲劳组,针刺组取足三里、阳陵泉、内关和肾俞穴,每日一次,留针20min。待针刺组针灸治疗1 h后,运动疲劳组和针灸组大鼠均进行中等运动强度的水平跑台运动,连续14d后取材,在电镜下观察肌细胞线粒体形态、检测骨骼肌MDA含量和SOD活性、测试肌细胞线粒体钙离子含量和膜电位。结果:与运动疲劳组相比,针刺组骨骼肌线粒体形态更趋于正常,其MDA含量低、SOD活性高,且能提高线粒体膜电位和钙离子含量。结论:针刺能有效的减轻运动性疲劳状态下骨骼肌线粒体的损伤,改善线粒体功能紊乱的情况,以解除活性氧自由基的毒性,提高运动机能。

Abstract:Objective: To observe the effects and mechanism of acupuncture on the structure of rat skeletal muscle with exercise-induced fatigue.Method: Thirty SD rats were divided into acupuncture group,control group and model group.Acupuncture group were treated Zusanli and Yanglingquan,Neiguan,Shenshu points once a day,retained the needle for 20 minutes.One hour later,the rats in the model group and acupuncture group received medium-intensity treadmill running to induce exercise-induced fatigue.The training lasts for 14 days.The muscle fibers,mitochondria and nucleus of damage were observed under the electron microscope,and MDA content,SOD activity,mitochondria membrane potential and calcium ion content of rat skeletal muscle were measured.Result: Compared with the model group,acupuncture could reduce the damage of the skeletal muscle and its mitochondria and nucleus,and could decrease MDA of skeletal muscle cells,activate SOD activity,and help solve the problems of steady imbalance of Ca2+ and reduction of mitochondria membrane potential in skeletal muscle cells.Conclusion: Acupuncture group could gain effects of anti-fatigue by alleviating damage of the skeletal muscle and improved exercise ability.
