间歇性负重游泳训练对大鼠杏仁基底外侧核(BLA)c-fos蛋白表达的影响

摘    要:目的:观察间歇性负重游泳训练后大鼠杏仁基底外侧核(BLA)内c-fos蛋白表达的变化,探讨间歇性游泳训练对c-fos蛋白表达的诱导机制。方法:建立大鼠间歇性负重游泳训练模型,用ABC免疫组织化学方法检测训练后即刻、0.51、h2、h4、h大鼠BLA内c-fos蛋白表达状况,并通过图像分析系统和统计学软件进行图像和数据分析。结果:对照组大鼠BLA内c-fos阳性神经元少量散在分布;模型组明显增多,且随着时间的增长呈上升趋势(0.5 h<1 h<2 h),2 h达到峰值,与对照组相比具有非常显著性差异(P<0.01);然后逐渐下降。结论:间歇性负重游泳训练可引起大鼠BLA中c-fos蛋白表达上调,且具有一定的时效性,其机制可能与间歇运动能引起神经细胞内钙离子浓度变化有关。

Abstract:Objective To investigate the mechanism of the expression of cfos protein by observing the changing tendency of c-fos protein in basolateral nucleus of rat amygdale(BLA) after intermittent swimming exercise.Methods: To establish the intermittent swimming exercise models,and detect the expression of c-fos protein in BLA at the moments of 0 h,0.5 h,1 h,2 h,4 h after exercise by immunohistochemical SABC method,then analyze the image and data by image analyzing system and statistic software.Results: Several Fos neurons of control group distribute in the BLA dispersively while the swimming groups express significantly higher,and increases progressively with time(0.5 h<1 h<2 h),the peak of Fos neurons happened at 2 h,and appears significant difference as compared with the control group(P<0.01);then descends regularly.Conclusion: Intermittent swimming exercise raises the expression of c-fos protein in BLA,and has a certain timeliness.The mechanism has deal with the change of the intracellular calcium concentration at nerve cells.
