振动训练对废用性肌萎缩大鼠比目鱼肌IGF-1及Akt-1表达的影响

摘    要:目的研究振动训练延缓废用性肌肉萎缩(SMDA)进程的分子机制。方法 SD雄性大鼠32只,随机均分为4组:正常对照组(A)、模型对照组(B)、每日振动训练组(C)和隔日振动训练组(D)。RT-PCR测比目鱼肌中IGF-1及Akt-1 mRNA的表达;Western-blotting测Akt-1和磷酸化Akt(pAkt-Thr308)的蛋白含量。结果 (1)与A组相比,B组、C组及D组右肢比目鱼肌湿重、肌肉湿重/体质量均显著下降(P<0.05)。(2)与A组比较,B组比目鱼肌IGF-1 mRNA、Akt-1 mRNA的表达和Akt-1蛋白含量均有显著性下降(P<0.05);与B组比较,C组和D组比目鱼肌IGF-1 mRNA、Akt-1 mRNA的表达和Akt-1蛋白含量均有显著性升高(P<0.05)。(3)与A组比较,B组和C组的Akt-1磷酸化水平均有下降,D组大鼠萎缩肌肉内磷酸化蛋白的表达水平显著下降(P<0.05)。结论振动训练可能通过IGF-1/PI3k/Akt1信号传导通路延缓SMDA,且不同间隔振动训练会对SMDA产生不同影响,其中每天振动训练干预的效果要优于隔天振动训练干预。

Abstract:Objective To study the molecular mechanism of vibration training delay muscular dystrophy process.Methods:32 SD male rats were randomly divided into four groups: normal control group(A),model control group(B),daily vibration training group(C) and every other day vibration training group(D);measure soleus IGF-1 mRNA and Akt-1 mRNA expression through the method of RT-PCR,measure soleus Akt-1 and Akt(pAkt-Thr308) protein content through the method of Western-blotting.Results:(1) compared with group A,group B,C and D right limbs soleus muscles wet weight,wet weight/weight indexes dropped significantly(P<0.05).(2) compared with group A,group B,C and D soleus IGF-1 mRNA、Akt-1 mRNA and Akt-1 protein content all declined significantly(P<0.05);compared with group B,group C and D soleus IGF-1 Mrna,Akt-1 mRNA and Akt-1 protein content all increased significantly(P<0.05).(3) Compared with group A,group B and group C phosphorylation Akt-1 expression level down,group D phosphorylation Akt-1expression level droped significantly(P<0.05).Conclusion:Vibration training effects SMDA may pass IGF-1/PI3k/Akt1 signaling pathways,and different intervals vibration training intervention produce different effects to SMDA,which dayly vibration training intervention is superior to every other day vibration training intervention.
