不同负荷游泳运动对大鼠海马食欲素A及其受体1表达的影响

摘    要:目的:观察不同负荷游泳运动对大鼠海马OXA及OX1R表达的影响,探讨不同负荷游泳运动对大鼠空间学习和记忆能力的影响机制。方法:将30只雄性大鼠随机分为3组(n=10):对照组(C组)、中等负荷运动组(M组)、大负荷运动组(O组)。C组不做任何运动,M组做中等负荷游泳运动8周,O组做大负荷游泳运动8周。采用Morris水迷宫、免疫荧光、Real-Time PCR和Western Blot实验技术分别对大鼠的空间学习记忆能力及海马OXA和OX1R的表达水平进行评估。结果:1)在Morris水迷宫实验定位航行训练期间,各组大鼠逃避潜伏期均呈逐渐下降趋势,第3天M组平均逃避潜伏期显著低于C组及O组(P<0.05),其余几天均无显著差异(P>0.05);在定位航行实验中,M组穿越原平台所在区域的次数显著高于C组及O组(P<0.05,P<0.01);2)M组OXA mRNA表达水平显著高于C组(P<0.05),OX1R mRNA表达水平显著低于C组(P<0.05)。而C组与O组OXA mRNA及OX1R mRNA的表达水平无显著差异(P>0.05)。免疫荧光实验表明,M组OXA蛋白表达水平显著低于C组(P<0.05)。Western Blot实验揭示,M组OX1R蛋白表达水平显著低于C组(P<0.05)。结论:1)中等负荷游泳运动可能通过降低OXA及OX1R的表达水平改善大鼠的空间学习记忆能力;2)大负荷游泳运动对海马OXA及OX1R的表达水平没有显著影响。

Abstract:Objective:To observe the effect of swimming with different loads on the expression of OXA and OX1R in the hippocampus of rats,and to explore the influence mechanism of swimming with different loads on the spatial learning and memory ability of rats.Methods:Thirty male rats were randomly divided into three groups(n=10):Control Group(C),Medium load Group(M),Heavy load Group(O).C group didn’t do any exercise.M group did medium load exercise for 8 weeks.O group did over load exercise for 8 weeks.Then the Morris Water Maze test,Immunofluorescence,Western Blot and Real-Time PCR technology were adopted to estimate rats’spatial learning and memory and the expression levels of OXA and OX1R in hippocampus respectively.Results:1)In the process of navigation training,all animals’escape latencies gradually shortened.On the third day,the average escape latency of Group M was significantly lower than that of Group C and Group O(P<0.05),and there was no significant difference in other days;in the navigation experiment,Group M was significantly higher than Group C and Group O for the times of going through the area of the original platform(P<0.05,P<0.01).2)The OXA mRNA expression level of M group was significantly higher than C group(P<0.05).The OX1R mRNA expression level of M group was significantly lower than C group(P<0.05).But the OXA mRNA and OX1R mRNA expression had no difference compared with C and O group(P>0.05).Immunofluorescence experiments showed that,the OXA protein expression level was significantly decreased in the hippocampus than group C(P<0.05).Western blot analysis revealed that OX1R protein expression level in group M was significantly lower than group C(P<0.05).Conclusion:Medium load swimming can improve the spatial learning and memory of rats by lowering the expression of OXA and OX1R.Heavy load swimming had no significant effect on hippocampal OXA and OX1R expression levels.
