摘    要:为了更快更清晰地观察显微镜下洋葱鳞茎内表皮的细胞骨架结构,对实验教材常用方法进行优化.通过采用单因子试验设计研究了不同固定荆、不同染色时间、不同Ca2+处理浓度和处理时间等对洋葱鳞茎内表皮细胞骨架观察的影响.结果表明,用l%CaCl2溶液浸泡2 h,再用甲醇-冰醋酸固定液固定30 min,尔后用考马斯亮蓝R250染色2...

Abstract:In order to observe the cytoskeleton of onion inner-epidermal cell with microscope more quickly and clearly,this article optimize the usual method of the experimental directions.We used the unifactor experiment method to design the different fixatives,different time for coloration,different concentration and time of Ca2+ to deal!with onion inner-epidermal cell,and then compared the effects of observation for cytoskeleton.The results showed that the cytoskeleton of onion inner-epidermal cell became more clearer through soaking in 1%CaCl2 for 2h and then fixing in!methanol-glacial acetic acid for 30 min,and staining by coomassie brilliant blue R250 for 20 min.
