鸡γ-干扰素的克隆和序列分析

摘    要:根据GenBank发表的鸡γ-干扰素核苷酸序列,使用primer 5设计一对特异性引物,通过RT-PCR技术从ConA诱导培养的鸡脾脏淋巴细胞中克隆出鸡γ-干扰素基因并对其进行测序,测序结果表明,鸡γ-干扰素基因全长495bp,具有一个完整的开放阅读框,编码164个氨基酸,与国外发表的序列比较,两序列间同源性为100%.计算机软件对鸡γ-干扰素编码的氨基酸序列进行了抗原性分析,结果表明鸡γ-干扰素具有良好的免疫原性.

Abstract:Based on chicken interferon-gamma(chicken IFN-γ) cDNA sequence published by GenBank,a pair of special primers were designed by computer software.A 495bp DNA fragment was amplified by RT-PCR using total RNA template extracted from chicken spleen lymphocyte that stimulate 24 hour by 1 milligram per milliliter concentration CoA.In order to identify this gene, the nucleotide was sequenced and compared with existing data in the GenBank published by Weining,K.C.The result indicated this chicken IFN-γ gene included an open reading frame(ORF) which encoded 164 amino acids residues,and had a 100% homology with chicken IFN-γ gene in GenBank,DNAstar soft analyze antigenicity of the chicken IFN-γ protein.The result shows the chicken IFN-γ protein has beter immunogenic.
