黄芪水提物对α-葡萄糖苷酶活性的抑制及其降血糖作用   总被引:1,自引:0,他引:1  
应用黄芪水提物与不同浓度的α-葡萄糖苷酶,不同浓度的底物PNP-glucoside进行反应,研究中药黄芪中α-葡萄糖苷酶抑制剂的抑制活性及其对正常小鼠血糖和糖耐量的影响.通过绘制酶量-反应速度曲线和双倒数曲线,确定其抑制剂类型,并对正常小鼠以40~160 mg.kg-1剂量灌胃后,观察其对血糖值和糖耐量的影响.结果表明,黄芪中α-葡萄糖苷酶抑制剂为可逆、非竞争性抑制;可降低正常小鼠血糖值,并且以80 mg.kg-1的剂量时血糖值最低,还可显著提高小鼠糖耐量.黄芪水提物中含有α-葡萄糖苷酶抑制剂成分,可用于糖尿病治疗.  相似文献

为探讨服用蛹虫草提取液和氨基酸对小鼠运动疲劳和抗缺氧的影响,建立训练鼠实验模型,灌服小鼠蛹虫草提取液和氨基酸,测定小鼠游泳力竭、爬竿、耐缺氧时间和运动训练血乳酸水平变化.结果显示;蛹虫草提取液复合灌喂训练模型小鼠,小鼠游泳力竭和爬竿及耐缺氧时间显著地长于生理盐水对照实验组小鼠(P〈0.05).蛹虫草提取液复合组与蛹虫草提取液单独灌喂组一样,游泳训练鼠的血乳酸水平变化显著地低于对照组和氨基酸处理组(P〈0.05).蛹虫草提取液和氨基酸复合对运动小鼠抗疲劳和运动生理机能的改善有积极作用.  相似文献

观察山楂、杭菊花、决明子(SHJ)煎剂高、中、低剂量代茶灌胃对小鼠动脉硬化(AS)模型的保护作用.用甲基硫氧嘧啶和动物猪油造小鼠动脉硬化模型,采用血象法分析检测小鼠血清总胆固醇(TC)、血清甘油三脂(TG)、血清高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)的含量,观察小鼠主动脉弓血管的形态学变化.研究结果表明:与正常对照组比较,模型组小鼠血清HDL-C含量显著减少,TG、TC、LDL-C显著增加(p〈0.05);与正常对照组比较,模型组血管壁增厚,脂肪堆积物增多.与模型组比较,山楂、杭菊花、决明子煎剂高、中剂量组小鼠血清HDL-C含量显著增加,TC、TG、LDL-C含量显著减少,对血管壁上脂肪的清除作用效果显著.结论:山楂、杭菊花、决明子煎剂代茶灌胃对小鼠高血脂模型具有保护作用,并能清除残留在血管壁上多余的脂肪堆积产物,加速脂肪物质代谢,预防动脉硬化的形成.  相似文献

目的:探讨生姜醇提物对糖尿病小鼠肾损害的保护作用。方法:将30只KM种雄性小鼠随机分为三组,正常对照组(C组,n=10)、模型组(DM组,n=10)及生姜醇提物干预组(ZGB组,n=10)。DM组及ZGB组小鼠尾静脉注射2%四氧嘧啶(90mg/kg)建立糖尿病肾损害模型。造模成功后,ZGB组给予生姜醇提物溶液0.5mL(300mg/kg·d)灌胃14d,DM组及C组给予生理盐水灌胃。14d后小鼠断头取血,测定血糖、血清肌酐、尿素氮含量,检测肾组织丙二醛(MDA)及超氧化物歧化酶(SOD)活力的变化,光镜下观察肾组织病理学改变。结果:与DM组比较,ZGB组血糖、血肌酐及尿素氮水平明显低于DM组(P〈0.01);肾组织MDA含量明显低于DM组(P〈0.01),而T—SOD活力则较DM组升高(P〈0.01)。光镜下ZGB组肾脏组织形态学的异常改变较模型组减轻。结论:生姜醇提物对糖尿病小鼠的肾损害有一定的保护作用。  相似文献

目的:探讨蒙医"蒜变剂"泻药对Ⅱ型糖尿病肾病小鼠微炎症的抑制及肾功能改善.方法:将实验动物随机分为正常组、Ⅱ型糖尿病肾病模型组、阳性对照组、低剂量治疗组、中剂量治疗组、高剂量治疗组,通过检测尿微量白蛋白和荧光探针标记技术观察各组动物肾小球滤过率情况,制备病理组织切片HE染色观察动物组织病理结构改变,运用透射电镜技术观察肾脏超微结构改变.结果:60mg/kg蒙医"蒜变剂"泻药可显著提高Ⅱ型糖尿病肾病小鼠的肾小球滤过率,修复肾小球基底膜功能,抑制肾间质纤维化,抑制肾小管上皮细胞足细胞广泛融合或丢失,降低电子致密物堆积.结论:蒙医"蒜变剂"泻药可有效抑制Ⅱ型糖尿病肾病小鼠微炎症.  相似文献

目的观察复方松针提取液对小白鼠肝损伤的保护作用.方法分别以四氯化碳和醋氨酚造成小鼠急性损伤模型,用赖氏法测定小鼠血清SALT活力并观察醋氨酚中毒对小鼠生存率的影响.结果复方松针提取液能降低小白鼠SALT水平并显著提高醋氨酚中毒小鼠的生存率.结论复方松针提取液具有明显的保肝作用。  相似文献

目的:观察香椿叶总黄酮对早期DN大鼠血糖值及血清中SOD、MDA水平的影响.方法:腹腔注射STZ60mg.kg^-1以造成糖尿病大鼠.注射1周后测定大鼠血糖,若血糖≥16.7mM者则纳入为糖尿病大鼠,并按血糖值范围随机分为模型组、氨基胍组(0.1g.kg^-1.d^-1)和香椿叶总黄酮低、中、高剂量组(O.1、0.2、0.4g.kg^-1.d^-1),同期另设一正常组,各组连续灌胃12周.于给药第6、12周测定大鼠血糖值,给药12周末测定大鼠血清SOD、MDA的含量.结果:香椿叶总黄酮能降低早期DN大鼠的血糖值,提高SOD活力,降低MDA水平.结论:香椿叶总黄酮具有降血糖作用,对早期DN引起的氧化应激损伤具有一定保护作用.  相似文献

研究小鼠禁食不同时间及注射胰岛素导致小鼠休克时血糖的变化,并观察酸性生理盐水稀释胰岛素对低血糖休克出现时间及血糖值有无影响.结果表明,小鼠分别禁食12﹑18﹑22﹑24h时的血糖值之间无明显差异(P〉0.05),在各组小鼠注射胰岛素后,血糖均明显下降(P〈0.05),但各组下降后的血糖值之间无明显差异(P〉0.05);不同剂量的胰岛素或是否用酸性生理盐水稀释胰岛素,注射后出现休克的时间及休克时各组小鼠血糖值之间无明显差异(P〉0.05).  相似文献

目的:观察竹节参的有效成分竹节参总皂苷(total saponins of Pallax japonicus,TSPJ)对小鼠免疫器官的影响;方法:灌胃TSPJ 14d,测定小鼠脾指数、胸腺指数,进行脾脏病理组织学切片的显微观察,了解TSPJ对小鼠免疫器官的影响;结果:与空白对照组比较,TSPJ对小鼠胸腺指数无明显影响,100mg·kg-1组,TSPJ对小鼠脾脏有抑制作用,100mg·kg-1组TSPJ小鼠脾脏生发中心不太明显,动脉周围淋巴鞘染色较淡;结论:TSPJ能影响小鼠免疫器官功能状态,这可能是其治疗风湿类疾病的机制之一.  相似文献

在71只小鼠上以200mg/kg剂量、闯日2次腹腔注射四氧嘧啶制备高血糖模型小鼠,得到35只血糖高于9.0mmol/L的小鼠。将高血糖模型小鼠按相似血糖水平分为对照组和2个实验组,血糖分别为:14.77±6.71(mmol/L,下同),14.53±5.40,14.40±6.18,组问无显著差异(P〉0.05)。对照组饲喂基础饲料,实验1和2组分别在基础饲料中添加10%的柑橘皮和10%的瓯柑橘皮,饲养1周后测定血糖,对照组为12.23+5.11,无显著变化(P〉0.05);柑橘组和瓯柑组下降到6.48±1.58和6.09±1.23(均P〈0.001),结果说明柑橘皮和瓯柑皮对小鼠有极显著的降糖作用。  相似文献

Psidium guajava (guava) leaf is a phytotherapic used in folk medicine to treat gastrointestinal and respiratory disturbances and is used as anti-inflammatory medicine. In nuclear medicine, blood constituents (BC) are labelled with technetium-99m (^99mTc) and used to image procedures. However, data have demonstrated that synthetic or natural drugs could modify the labelling of BC with ^99mTC. The aim of this work was to evaluate the effects of aqueous extract of guava leaves on the labelling of BC with ^99mTc. Blood samples of Wistar rats were incubated with different concentrations of guava extract and labelled with ^99mTc after the percentage of incorporated radioactivity (%ATI) in BC was determined. The results suggest that aqueous guava extract could present antioxidant action and/or alters the membrane structures involved in ion transport into cells, thus decreasing the radiolabelling of BC with ^99mTc. The data showed significant (P〈0.05) alteration of ATI in BC from blood incubated with guava extract.  相似文献

Objective

The present study aims at determining the stability of a popular type 2 diabetes rat model induced by a high-fat diet combined with a low-dose streptozotocin injection.

Methods

Wistar rats were fed with a high-fat diet for 8 weeks followed by a one-time injection of 25 or 35 mg/kg streptozotocin to induce type 2 diabetes. Then the diabetic rats were fed with regular diet/high-fat diet for 4 weeks. Changes in biochemical parameters were monitored during the 4 weeks.

Results

All the rats developed more severe dyslipidemia and hepatic dysfunction after streptozotocin injection. The features of 35 mg/kg streptozotocin rats more resembled type 1 diabetes with decreased body weight and blood insulin. Rats with 25 mg/kg streptozotocin followed by normal diet feeding showed normalized blood glucose level and pancreatic structure, indicating that normal diet might help recovery from certain symptoms of type 2 diabetes. In comparison, diabetic rats fed with high-fat diet presented decreased but relatively stable blood glucose level, and this was significantly higher than that of the control group (P>0.05).

Conclusions

This model easily recovers with normal diet feeding. A high-fat diet is suggested as the background diet in future pharmacological studies using this model.

目的:观察姜黄素对胰岛素抵抗(IR)大鼠糖脂代谢及血清脂肪细胞因子的影响,探讨姜黄素干预IR的作用机制。方法:采用高脂饲料喂养,建立IR大鼠模型。将IR大鼠随机分为3组:模型组、吡咯列酮组和姜黄素组。给药4周后测定空腹血糖、胰岛素、血脂、TN F-α、Leptin等指标。结果:模型组大鼠血清FIN S、TC、TG和LDL水平均较空白组显著升高,而ISI及HDL水平显著下降;血清TN F-α、Leptin及FFA水平也较空白组明显升高;用姜黄素干预后,大鼠ISI较模型组显著升高,血脂得到改善,FINS、TNF-α、Leptin及FFA水平均显著下降。结论:姜黄素可改善IR大鼠糖脂代谢紊乱,增加胰岛素的敏感性,其机制可能与其调节脂肪细胞因子TNF-α、Leptin的分泌有关。  相似文献

为了研究益智仁对束缚应激致大鼠海马神经元NMDA受体亚基NR2B表达的调节作用,将30只SD大鼠随机等分为正常对照组、模型对照组和益智仁治疗组。模型对照组、益智仁治疗组大鼠每日束缚1次,连续3周。益智仁治疗组束缚前给予益智仁水提取物灌胃。采用免疫组织化学方法,观察益智仁水提取物对束缚应激大鼠海马神经元NMDA受体亚基NR2B表达的调节作用。结果显示NR2B阳性产物呈深棕色或棕褐色,主要沉积在CA1区和CA3区锥体细胞的细胞膜周边。与正常对照组比较,模型对照组的NR2B阳性产物明显增多(P﹤0.01);益智仁治疗组与模型对照组比较,NR2B阳性产物明显减少(P﹤0.05)。实验结果提示益智仁能够降低束缚应激大鼠海马CA1区和CA3区NMDA受体亚基NR2B的表达。  相似文献

Objective: To investigate the potential and early effect of hypertonic saline resuscitation on T-lymphocyte subpopulations in rats with hemorrhagic shock. Methods: A model of rat with severe hemorrhagic shock was established in 18 Sprague-Dawley (SD) rats. The rats were randomly divided into Sham group, HTS group (hypertonic saline resuscitation group) and NS group (normal saline resuscitation group). Each group contained 6 rats. The CD4~ and CD8~ subpopulations of T-lymphocytes in peripheral blood were detected respectively before shock and after resuscitation by double antibody labelling and flow cytometry. Results: In the early stage after hemorrhagic shock, fluid resuscitation and emergency treatment, the CD4^+ lymphocytes of peripheral blood in HTS and NS groups markedly increased. Small volume resuscitation with HTS also induced peripheral CD8^+ lymphocytes to a certain extent, whereas NS resuscitation showed no effect in this respect. Consequently, compared with Sham and HTS groups, CD4^+/CD8^+ ratio of peripheral blood in NS group was obviously increased, and showed statistically differences. Conclusion: In this model of rat with severe hemorrhagic shock, small volume resuscitation with HTS is more effective than NS in reducing immunologic disorders and promoting a more balanced profile of T-lymphocyte subpopulations regulating network.  相似文献

[目的]〗研究知母多糖(Anemam,AM)对家兔血糖的影响.[方法]制备家兔高血糖模型,用不同剂量组的AM和D860后,测定对照组、AM组、D860组的血糖.[结果]AM1次灌胃2mg/kg,20mg/kg可显著降低四氧嘧啶引起的家兔血糖升高;AM1次灌胃20mg/kg对正常家兔血糖无明显影响,200mg/kg可降低正常家兔血糖.[结论]AM与D860同样具有实验性降低血糖作用.  相似文献

目的:研究肝龙胶囊对正常小鼠血糖及肾上腺素致高血糖模型小鼠的血糖浓度的影响。方法:采用葡萄糖氧化酶法测定正常小鼠的血糖浓度及肾上腺素诱导小鼠高血糖不同处理后的血糖浓度变化。结果:肝龙胶囊对正常小鼠血糖浓度无影响,而对肾上腺素高血糖模型小鼠有显著的降糖作用(P〈0.01,P〈0.05)。结论:肝龙胶囊能降低肾上腺素性高血糖小鼠的血糖浓度,对正常小鼠血糖浓度无影响。  相似文献

目的:观察脂肪肝大鼠在生化、病理、脂肪代谢、细胞因子、胰岛素及瘦素各方面指标的变化.方法:雄性Wistar大鼠22只,按体重层次随机分为2组,对照组(A)11只,脂肪肝组(B)11只.对照组饲以普通鼠饲料,脂肪肝组饲以高脂饲料.8周后,全部处死,取血及肝组织,测定血中AIT、AST、TG、CHO、INS、FBS、FFA、Ieptin、II-6、TNF.肝组织匀浆测定肝组织TC,TG,SOD、MDA,观察肝脏组织学变化.结果:脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素表达增高;细胞因子升高.结论:实验性高脂饮食脂肪肝大鼠存在瘦素抵抗及胰岛素抵抗.  相似文献

Objective

To investigate the effect of tea polyphenols on cardiac function in rats with diabetic cardiomyopathy, and the mechanism by which tea polyphenols regulate autophagy in diabetic cardiomyopathy.

Methods

Sixty Sprague-Dawley (SD) rats were randomly divided into six groups: a normal control group (NC), an obesity group (OB), a diabetic cardiomyopathy group (DCM), a tea polyphenol group (TP), an obesity tea polyphenol treatment group (OB-TP), and a diabetic cardiomyopathy tea polyphenol treatment group (DCM-TP). After successful modeling, serum glucose, cholesterol, and triglyceride levels were determined; cardiac structure and function were inspected by ultrasonic cardiography; myocardial pathology was examined by staining with hematoxylin-eosin; transmission electron microscopy was used to observe the morphology and quantity of autophagosomes; and expression levels of autophagy-related proteins LC3-II, SQSTM1/p62, and Beclin-1 were determined by Western blotting.

Results

Compared to the NC group, the OB group had normal blood glucose and a high level of blood lipids; both blood glucose and lipids were increased in the DCM group; ultrasonic cardiograms showed that the fraction shortening was reduced in the DCM group. However, these were improved significantly in the DCM-TP group. Hematoxylin-eosin staining showed disordered cardiomyocytes and hypertrophy in the DCM group; however, no differences were found among the remaining groups. Transmission electron microscopy revealed that the numbers of autophagosomes in the DCM and OB-TP groups were obviously increased compared to the NC and OB groups; the number of autophagosomes in the DCM-TP group was reduced. Western blotting showed that the expression of LC3-II/I and Beclin-1 increased obviously, whereas the expression of SQSTM1/p62 was decreased in the DCM and OB-TP groups (P<0.05).

Conclusions

Tea polyphenols had an effect on diabetic cardiomyopathy in rat cardiac function and may alter the levels of autophagy to improve glucose and lipid metabolism in diabetes.
