该实验通过测定冠心宁注射液中的丹参酚酸类成分和糖类成分,为评价冠心宁注射液的质量品质提供理论依据.采用紫外可见分光光度法检测,进行方法学考察,测定10批冠心宁注射液的丹参酚酸和总糖的含量.结果表明,NaNO2-Al（NO3）3显色法用于测定丹参酚酸的含量;苯酚-硫酸法用于测定总糖的含量;10批冠心宁注射液中丹参酚酸和总糖的平均含量分别为93.27mg.10 ml-1、969.87 mg.10 ml-1,分别占固含物的7.66%、79.60%.因此,该方法简便快速、准确可靠,可有效评价冠心宁注射液中80%以上的大类成分的含量. 相似文献

It is well known that ethacrynic acid (EA) can potentiate the ototoxicity of aminoglycoside antibiotics (AmAn) such as kanamycin
(KM), if they were applied at the same time. Currently, to create the model of EA-KM-induced cochlear lesion in rats, adult
rats received a single injection of EA (75 mg/kg, intravenous injection), or followed immediately by KM (500 mg/kg, intramuscular
injection). The hearing function was assessed by auditory brainstem response (ABR) measurement in response to click and/or
tone bursts at 4, 8, 12, 16, 20, 24, and 32 kHz. The static microcirculation status in the stria vascularis after a single
EA injection was evaluated with eosin staining. The pathological changes in cochlear and vestibular hair cells were also quantified
after co-administration of EA and KM. After a single EA injection, blood flow in vessels supplying the stria vascularis rapidly
diminished. However, the blood supply to the cochlear lateral wall partially recovered 5 h after EA treatment. Threshold changes
in ABR were basically parallel to the microcirculation changes in stria vascularis after single EA treatment. Importantly,
disposable co-administration of EA and KM resulted in a permanent hearing loss and severe damage to the cochlear hair cells,
but spared the vestibular hair cells. Since the cochlear lateral wall is the important part of the blood-cochlea barrier,
EA-induced anoxic damage to the epithelium of stria vascularis may enhance the entry of KM to the cochlea. Thus, experimental
animal model of selective cochlear damage with normal vestibular systems can be reliably created through co-administration

This study was carried out to elucidate the nephroprotective effects from a mixture of 8 L-amino acids and the possible mechanism
of protection by this amino acid mixture. Acute renal failure model was induced by an intravenous administration of 10 mg/kg
cisplatin to male Sprague-Dawley rats. A mixture of 8 L-amino acids or 0.9% saline was infused at a rate of 2 ml/h for 3 h,
starting with a 2 ml bolus injection before cisplatin administration. Amino acids showed no acute effect on renal morphology.
The infusion of a mixture of 8 L-amino acids increased GFR by 85% in control rats. The abnormalities of urine sodium and potassium
excretion caused by cisplatin were markedly attenuated by the administration of the amino acid mixture. With the infusion
of this amino acid mixture, cisplatin-induced abnormal state 4 respiration returned to control levels and the depressed state
3 respiration, respiratory control ratio and carbonyl cyanide p-trifluoromethoxyphenyl hydrazone-uncoupled respiration were
ameliorated remarkably. A mixture of 8 L-amino acids showed marked nephroprotection against cisplatin-induced acute renal
failure in rats and might function through augmentation of the cisplatin-injured renal mitochondrial electron transport-oxidative
phosphorylation sequence, probably via stabilizing the membrane (including inner mitochondrial membrane) protein tertiary
structure. In addition, this amino acid mixture remarkably increased GFR and decreased urine sodium excretion in rats. 相似文献

A high-performance liquid chromatographic (HPLC) method with ultraviolet (UV) detector had been developed for simultaneous quantification of danshensu, protocatechuie aldehyde, caffeic acid, salvianolic acid D, rosmarinic acid, salvianolic acid B and salvianolic acid A in Danshen injection. According to the UV spectra of these components, three detection wavelengths have been selected as follows: 280 nm for danshensu and protocatechuic aldehyde, 326 nm for caffeic acid, salvianolic acid D and rosmarinic acid, 286 nm for salvianolic acid B and salvianolic acid A. The limit of detection (LOD) was improved to be in the range of 0.008～0.160 μg/ml. Moreover, excellent linear behavior over the investigated concentration range was observed, with R>0.999 for all the analytes. 相似文献

目的:研究海藻多糖(Polysaechrides from Spirulina Platensis,PSP)对小鼠肝损伤动物模型与肝匀浆脂质过氧化模型的抗氧化作用。方法:采用腹腔注射CCL4的方法,制造小鼠肝损伤动物模型,金氏法测定血清谷氨酸丙酮酸转氨酶(SGPT)和肝脏的过氧化脂质(Lipoper-oxides,LPO)含量;Fenton反应诱导小鼠肝匀浆脂质过氧化动物模型,采用硫代巴比妥酸(TBA)法测定肝匀浆LPO。结果:在体外,海藻多糖可剂量依赖性地抑制Fenton反应导致的CCL4毒化小鼠肝匀浆中LPO的生成,其IC50为1.6×10-4。在体内,灌胃250mg/kg的海藻多糖组可明显降低CCL4毒化小鼠血清SGPT活性(P<0.01)。结论:海藻多糖对体内外的肝组织有一定程度的抗氧化作用。 相似文献

Taste aversions were conditioned by exposing subjects to a 1.0% saccharin solution 30 min after an injection of lithium chloride. The aversion learning was disrupted if subjects had also received an additional lithium injection some time earlier (Experiments 1–3). This interference effect of US preexposure was a decreasing function of the preexposure interval, beyond the optimal interval (105 min) for observing the phenomenon (Experiment 1), and was directly related to the dose of the preexposure injection (Experiment 2). No interference with conditioning occurred at short (e.g., 30-min) preexposure intervals (Experiment 1), probably because under these circumstances the preexposure injection itself conditioned a strong aversion (Experiment 4). At moderate (105-min) but not at short (30-min) preexposure intervals, the interference with aversions learned as a result of taste exposure following drug injection was comparable to the interference with learning in a more conventional forward conditioning procedure (Experiments 3 and 4). These findings are similar to previously documented effects of proximal CS- and US-preexposure and are consistent with recent stimulus rehearsal and opponent-process theories. 相似文献

为了探讨光敏剂瘤内注射法在临床上的应用,本实验将高浓度血卟啉衍生物（HpD）直接注入荷瘤小鼠的肿瘤内,利用组织涂片法,通过荧光显微镜观察．结果,注射后1～5天内,局部注射组肿瘤内发橘红色荧光的细胞的百分比明显高于腹腔注射组,以1～4天为著．局部注射组的肝、脾、肾组织中发荧光细胞的百分比低于腹腔注射组．根据实验结果．肿瘤内注射可以提高瘤内HpD的浓度,而减少正常组织中HpD的含量,减轻毒副作用． 相似文献
