绿色木霉纤维素酶系分泌特性及酶解条件的研究 总被引：3，自引：0，他引：3
采用正交实验法分析出绿色木霉3.3711纤维酶系各组分量适酶解条件组合分别为（pH5.0、45℃）（C1酶），（pH3.5、55℃）（Cx酶）和（pH4.5、65℃）（βG酶）。分析比较了分别使用纤维素偻和麦麸为单一或混合碳源时在产酶过程中各组分酶的最高酶活，比酶活及其形成时间和分泌特性。结果表明，同种碳源不同浓度所产酶系中C1比酶活性相对稳定，C1和Cx酶活分别形成一个和两个高峰，βG酶均为一个高峰。混合碳源使各组分酶活高峰期相对一致，高峰期稳定时间延长和酶量增加。 相似文献

利用木聚糖作为唯一碳源的基础盐培养,在50℃培养条件下,从新乡周围土样及腐殖质中筛选到1株产耐热β-木聚糖酶细菌,初步鉴定为巨大芽孢杆菌（Bacillus megaterium FLH-2）.该菌株在40~55℃生长良好,最适生长温度50℃.木聚糖酶粗酶性质研究表明：该酶最适pH和最适温度分别为6.0和65℃;在pH 5.0~9.0及温度30~75℃之间酶活相对稳定,分别处理30 min,相对酶活仍保持在80%以上. 相似文献

从温泉、堆肥等环境采集样品,以滤纸条为唯一碳源,用微晶纤维素粉和羧甲基纤维素钠(CMC-Na)-刚果红培养基进行分离和筛选出几株高温纤维素分解菌.进行测定这些菌株的Cx、滤纸酶活性试验,两种酶的作用温度相对较宽,Cx酶的最适酶活性温度约70℃,而滤纸酶活性的最适温度因菌株差异有所差异. 相似文献

利用杏鲍菇(pleurotus eyngii)降解啤酒糟液态发酵生产木聚糖酶,运用单因素与正交试验研究了啤酒糟、玉米芯与麸皮的不同组分、氮源、pH值、培养时间、转速、装瓶量等对产木聚糖酶活力的影响,并对其粗酶的酶学性质进行了研究。结果表明:碳源为3%啤酒糟、1%麸皮和1%玉米芯,氮源为0.5%酵母膏,pH值为5,培养时间为8d,转速为160 r/m in,接种量为10%,装瓶量为100 mL/250mL时,酶活力最高。其粗酶液的最适反应温度为50℃,最适反应pH为6,在pH4~6的范围内酶活性较稳定。但粗酶液的热稳定较差,当温度升至60℃时酶活力损失65%以上。 相似文献

本文报告了黄豆籽苗胺氧化酶(SSAO)的制备方法及其酶学特性。每100 g 豆芽可制备5ml 黄色酶液,以腐胺为底物比活性为20μmol/60min/mg 蛋白质。酶液经聚丙烯酰胺凝胶电泳可分成5个组分,其电泳位置相当于血清白蛋白、α和β球蛋白,经 Sephadex G-100凝胶层析分离可得到2个组分,98%的酶活性分布在第一组分中。酶液经4℃保存,活性渐降、7天降9%,19天降22%、80天降64%。如在-20℃保存,活性下降更快,7天降28%,19天降42%,80天降51%。用腐胺作底物的米氏常数(K_m)为0.26mmolL~(-1)。当酶量为0.10ml,作用时间为5分钟时,最大线性范围达200nmol;如果及应时间增至15分钟最大线性范围降为100nmol。本酶的最适 pH 为8—9。尿素、肌酐、组氨酸精氨酸和赖氨酸对本酶无干扰作用. 相似文献

本文对一株分离自黄海深海海底的产纤维素酶海洋适冷茵进行了产酶条件研究。结果显示。在接近自然海水盐度、温度20℃，自然pH条件下，茵体生长和产酶较好。所产纤维素酶为诱导酶，主要作用底物为CMC-Na和微晶纤维素。以1％球磨Avicel 1％蔗糖构成的复合碳源产酶活力最高；最适氮源为1％蛋白胨 1％酵母膏。 相似文献

目的：研究纤维素分解菌的分离及产酶条件优化.方法：以含菌秸秆、腐木叶和玉米地土壤为原料,经富集、初筛培养后,根据水解圈直径/培养天数进行纤维素分解菌的复筛培养,测定CMC酶活和FPA滤纸酶活,并进行了产酶条件的单因素优化实验.结果：采集的样品中共分离到4个菌株,均为细菌,其中H-3的羧甲基纤维素（CMC）酶活和滤纸（FPA）酶活最高,分别是0.5401U.m l-1、0.2923U.m l-1.结论：H-3菌分解纤维素的能力最强,最佳产酶条件为温度30℃,pH4.5.不含尿素,葡萄糖和纤维素含量为0.4%和0.6%. 相似文献

通过筛选从土壤和腐朽树木中得到了4株纤维素酶高产菌株MF1、MF4、TF3和TF9,经初步鉴定分别是绿色木霉、青霉属、康氏木霉和黑曲霉.通过对TF3发酵产酶的影响因素研究表明,其发酵产酶的最佳碳源是麸皮-纤维素粉复合物,最佳氮源是硫酸铵,最适发酵温度范围是30～32℃,最适初始pH范围是6～7. 相似文献
