动脉粥样硬化等多种心血管疾病,表现为血管内皮功能紊乱,并伴有非对称性二甲基精氨酸 (ADMA,内源性一氧化氮合酶抑制物)血浓度的升高,ADMA可能是一种重要的致动脉粥样硬化因子。 ADMA通过抑制一氧化氮合酶(NOS)、致炎与诱导氧化应激等机制影响内皮细胞、平滑肌细胞、单核细胞及血小板等多种细胞功能,从而促进动脉粥样硬化发生与发展。 相似文献

文章以清洁级纯系雄性SD大鼠为实验模型,应用组织病理学、RT—PCR技术,研究高脂饮食诱发动脉粥样硬化过程中血管内膜细胞大电导钙激活的钾通道(large conductance Ca—activated K+ channels,BKCa)、中电导钙激活的钾通道(Intermediate condutance Ca—activated K+ channels,IKCa)基因表达的动态改变情况,从基因表达水平探讨高脂饮食诱发动脉粥样硬化的作用机制。同时指出,IKCa可能为动脉粥样硬化的早期启动因子,此观点有助于动脉粥样硬化的早期检测与防治。 相似文献

目的:近年来发现内皮抑素可以抑制组织纤维化,但是具体机制不详。本文旨在研究内皮抑素抑制纤维化的作用机制。创新点:首次阐明了内皮抑素对成纤维细胞PDGFRβ(血小板衍生生长因子受体β)/ERK(细胞外调节蛋白激酶)信号通路的影响及其与纤维化的关系。方法:体外培养人成纤维细胞,采用血小板衍生生长因子-BB(PDGF-BB)或转化生长因子-β1(TGF-β1)建立细胞纤维化模型,进一步应用内皮抑素处理,通过检测胶原及肌成纤维细胞表面标志等了解内皮抑素对细胞纤维化和表型转化的抑制作用。进而检测内皮抑素对成纤维细胞PDGFRβ/ERK信号通路的影响,并分析其可能与抗纤维化作用存在的关联。结论:内皮抑素通过调控PDGFRβ/ERK信号通路抑制瘢痕增生,是多靶点的、具有较好的抗纤维化临床应用前景的药物。 相似文献

目的:探讨辛伐他汀对外源性制备的糖基化终末产物损伤大鼠离体胸主动脉环内皮依赖性舒张功能的影响及其机制。方法:按文献方法制备糖基化终末产物,采用外源性糖基化终末产物(AGE-BSA)孵育大鼠离体胸主动脉环90min诱导血管内皮功能的损伤,并观察辛伐他汀、超氧化物歧化酶和L-精氨酸对糖基化终末产物致的血管内皮依赖性舒张功能损害的影响。结果:外源性糖基化终末产物孵育大鼠离体胸主动脉环90min,明显抑制乙酰胆碱诱导的内皮依赖性血管舒张反应(endothelium-dependent relaxation,EDR)。但对硝普钠诱导的内皮非依赖性血管舒张反应没有影响。辛伐汀(0.05,0.1,0.2μmol/L)与AGE-BSA共同孵育血管环90min,浓度依耐性地改善AGE-BSA对血管内皮依赖性舒张功能的损害。氧自由基清除剂超氧化物歧化酶(superoxide dismutase,SOD,200U/mL)也可逆转AGE—BSA对内皮依赖性血管舒张反应的损害,而L-精氨酸(L-arginine,L-Arg,3mmol/L)却只有部分保护作用。而且,辛伐他汀也能取消SOD抑制剂二乙基二硫氨甲酸酯(diethyldithiocarbamate,DETC;10μmol/L).诱导产生内源性氧自由基所致的血管内皮依赖性反应的抑制。结论:辛伐他汀能取消AGE-BSA对大鼠离体胸主动脉环血管内皮依赖性舒张反应的抑制。该保护作用可能与其抗氧化作用有关。 相似文献

This paper discusses research examining the attitudes and behaviours of researching women in academia and considers the effect
of these factors on successful researching outcomes. The results of this exploratory research highlight in particular, a number
of interesting environmental influencers which contribute to enhancing successful work outcomes for academic women researchers.
Specifically, personal factors such as, marital status, partner support, age, cultural background and level of organisation
(in life) coupled with, research defined factors such as incentive for conducting the research and the existence of research
partnerships and/or groups are identified as significant performance influencers. These dimensions appear to facilitate the
level of research productivity for women academics based on key performance indicators such as journal/conference paper submissions
and successful research funding applications. The potential benefits of this exploratory research are that any correlation
between specific self-supporting attitudes or behaviours of successful women academics and effective research outcomes could

宫颈癌早期是以手术为主的综合治疗,中晚期是以放疗为主的综合治疗。尽管有多种治疗方法可供选择,具有复发高风险因素如巨块型、盆腔淋巴结转移、脉管淋巴转移等情况的宫颈癌,预后仍较差。了解评价宫颈癌预后的生物学指标,对宫颈癌患者治疗方案的选择、化疗及放疗的剂量调整等至关重要。本文从宫颈癌肿瘤内环境指标、与宫颈癌细胞生物学特性相关指标、离体标本相关指标、分子功能影像学方法四方面评价宫颈癌预后生物学指标的最新进展。 相似文献

Research on factors that shape recruitment and retention in undergraduate science majors currently is highly fragmented and in need of an integrative research framework. Such a framework should incorporate analyses of the various levels of organization that characterize academic communities (i.e., the broad institutional level, the departmental level, and the student level), and should also provide ways to study the interactions occurring within and between these structural levels. We propose that academic communities are analogous to ecosystems, and that the research paradigms of modern community ecology can provide the necessary framework, as well as new and innovative approaches to a very complex area. This article also presents the results of a pilot study that demonstrates the promise of this approach at the student level. We administered a questionnaire based on expectancy-value theory to undergraduates enrolled in introductory biology courses. Itself an integrative approach, expectancy-value theory views achievement-related behavior as a joint function of the person's expectancy of success in the behavior and the subjective value placed on such success. Our results indicated: (a) significant gender differences in the underlying factor structures of expectations and values related to the discipline of biology, (b) expectancy-value factors significantly distinguished biology majors from nonmajors, and (c) expectancy-value factors significantly predicted students' intent to enroll in future biology courses. We explore the expectancy-value framework as an operationally integrative framework in our ecological model for studying academic communities, especially in the context of assessing the underrepresentation of women and minorities in the sciences. Introductory tertiary level science classes (i.e., at the university or post-compulsory school level) including those for biology face increasing diversity in intake. Previous research has indicated university level teachers assume a certain level of prior knowledge which may or may not be possessed by such students. This report focuses on the use of concept mapping in introductory biology tutorial classes. ABSTRACTScience education today has become increasingly focused on research into complex natural, social and technological systems. In this study, we examined the development of high-school biology students’ systems understanding of the human body, in a three-year longitudinal study. The development of the students’ system understanding was evaluated using the Components Mechanisms Phenomena (CMP) framework for conceptual representation. We coded and analysed the repertory grid personal constructs of 67 high-school biology students at 4 points throughout the study. Our data analysis builds on the assumption that systems understanding entails a perception of all the system categories, including structures within the system (its Components), specific processes and interactions at the macro and micro levels (Mechanisms), and the Phenomena that present the macro scale of processes and patterns within a system. Our findings suggest that as the learning process progressed, the systems understanding of our students became more advanced, moving forward within each of the major CMP categories. Moreover, there was an increase in the mechanism complexity presented by the students, manifested by more students describing mechanisms at the molecular level. Thus, the ‘mechanism’ category and the micro level are critical components that enable students to understand system-level phenomena such as homeostasis. 相似文献
