贲门癌患者外周血Th1/Th2细胞因子检测的临床意义 总被引：2，自引：0，他引：2
检测贲门癌患者外周血CD3^＋T细胞分泌细胞因子的水平变化，分析Th1和Th2细胞的细胞因子免疫活动，为肿瘤的免疫治疗提供实验依据,采用刺激物刺激细胞，增加细胞内细胞因子的表达，再加入荧光标记的特异性抗细胞因子单克隆抗体，特异性抗原抗体结合，最后应用流式细胞仪分析特异性细胞因子表达水平．用酶联免疫吸附法（ELISA）检测相应的细胞因子的方法进行研究．结果：贲门癌患者Th1型细胞因子干扰素-γ（IFN-γ）、白细胞介素-2（IL2）、白细胞介素-12（IL-12）表达水平较正常对照组显著降低，Th2型细胞因子白细胞介素-4（IL-4）和白细胞介素-10（IL-10）表达水平较正常对照组升高，差异有统计学意义．肿瘤坏死因子-α（TNF-α）表达水平较正常对照组升高，差异有显著性．贲门癌患者体内Th2型细胞因子模式占优势状态，上述结果可能是肿瘤细胞发生免疫逃逸，从而导致肿瘤的发生或者转移的原因之一． 相似文献

目的：探讨流式微球阵列术（Cytometric Bead Array,CBA）检测Th1/Th2细胞因子在V型狼疮性肾炎（LN）患者血清中的表达及意义。方法：用CBA法检测20例V型LN患者和20例正常人血清中Th1（TNFα、IFNγ）和Th2（IL-4和IL-5）类细胞因子表达情况。结果：V型LN组Th1类细胞因子TNFα和IFNγ表达均高于正常对照组,而正常对照组中IL-4和IL-5则高于狼疮肾患者,两组中IL-2表达无差异。结论：Th1和Th2类细胞因子平衡/紊乱在狼疮肾患者肾组织损害中可能起一定作用。CBA法检测Th1/Th2细胞因子是一种高效、简便和敏感度高的方法。 相似文献

目的:通过测定女性吸食海洛因患者外周血中T细胞亚群Th1/Th2细胞因子和血清中性激素的变化,从分子水平探讨吸食海洛因对女性免疫功能和性激素的影响。方法:取吸毒组和正常对照组清晨空腹静脉血3mL,应用双抗体夹心ABC-ELISA法测定Th1/Th2细胞因子,采用放免法检测血清雌激素(E2)、孕激素(P)。结果:吸毒组30例患者Th1细胞因子(IL-2:1.45±0.48μg/L,TNF-β:0.37±0.14μg/L,INF-γ:2.99±0.55μg/L)和Th2细胞因子(IL-4:23.68±11.22μg/L,IL-10:12.11±5.13μg/L)低于32例健康组Th1细胞因子(IL-2:4.09±0.79μg/L,TNF-β:0.91±0.35μg/L,INF-γ:5.69±0.91μg/L)、Th2细胞因子(IL-4:54.24±17.21μg/L,IL-10:22.34±7.26μg/L)水平(P<0.01);吸毒组患者血清雌激素与孕激素水平显著低于健康组(吸毒组E2:38.71±18.18ng/L,P:0.79±0.53μg/L;健康组分别为79.98±34.42ng/L和2.10±0.86μg/L)(P<0.01)。结论:海洛因的摄入可导致青年女性性腺激素水平下降和免疫水平全面下降。 相似文献
