本文用免疫组织化学PAP法染色,观察胃癌组与对照组之间胃窦粘膜幽门腺非病变区5—HT样免疫反应细胞的变化。结果:5—HT样免疫反应细胞在癌症组明显增加,与对照组相比有显著性差异,提示5—HT样免疫反应细胞可能参与胃癌期间的调制。  相似文献

应用真彩色医学图象分析仪,对15例DU、GU和Con组患者的胃窦粘膜肉眼下非病变区30张切片,随机选择,经免疫组化双重染色的G和D细胞300个,进行形态定量分析。每个细胞检测12个参数,各项参数都行统计学t检验两两相比处理。在三组中,G细胞的周长、面积、长径、短径、比表面、平均截距、平均体积、平均表面积、圆球度、平均直径和D细胞的长径、短径、形状因子、平均轴比的显著差异率均在60%以上,这些结果,建立了溃疡病非病变区胃窦粘膜幽门腺G和D细胞的体视学形态参数数据库,并为它们参与内分泌调节提供了客观依据。  相似文献

应用真彩色图象分析仪,对15例胃溃疡(GU)、十二指肠溃疡(DU)和浅表性胃炎作对照组(Con)患者,非病变区胃窦粘膜的151个β—内啡肽细胞(β—EP 细胞)形态计量学参数及45个测试单位面积(5000μm~2)的细胞数进行检测,收集13项参数行统计学 t 检验。呈高度显著性差异的参数分别有三组间的周长(C)、面积(A)、长径(LD)、短径(SD)和平均体积(V);DU 与 Con,GU 与 Con 组的比表面(δ),平均截距(τ)、平均面积(?)和平均直径((?)c);DU 与 GU、或与 Con 组的细胞均数(N)。三组中各参数的显著性差异率 C、A、LD、SD、(?)达100%,δ、τ、(?)、(?)c、N 为60%以上。本文首次应用真彩色图象分析仪,建立了 DU、GU 和 Con 组非病变区胃窦粘膜幽门腺β—内啡肽细胞的二维和三维体视学、形态计量学参数及细胞均数数据,并为其参与内分泌调节提供了客观依据。  相似文献

用雄性昆明种小鼠45只,分为实验性溃疡组和对照组,后者又分为盐水组和空白组。在实验性胃溃疡术后3、6、9和20d,腹腔注射秋水仙碱3小时后,取胃窦组织。用免疫组织化学和苏木精染色。观察胃窦粘膜G细胞的变化。正常胃窦粘膜幽门腺分裂指数5.93±1.23;G细胞百分率2.76±0.45;G细胞分裂指数Ⅰ0.85±0.18;G细胞分裂指数Ⅱ0.023±0.01。幽门腺细胞分裂指数、G细胞百分率及其分裂指数Ⅱ,在术后6、9和20d与盐水组相比,呈高度显著性差异;G细胞分裂指数Ⅰ,术后9d与空白组、术后20d与盐水组相比,均呈显著性差异。本文讨论了以上变化的意义。  相似文献

目的观察胃窦粘膜幽门腺内分泌细胞中5-HT和胃泌素共存与否.方法应用已树脂封片的成人胃窦粘膜5-HT细胞再行双重PAP法显示G细胞的方法,在同一张切片上进行5-HT和胃泌素的双重染色.结果5-HT细胞多散在于粘膜的中下1/3,呈棕褐色,形态多样,常见有突起的细胞.G细胞呈带状分布于粘膜的中1/3,大部分G细胞为蓝色,呈圆形或锥体形.有时可见5-HT细胞和G细胞相比邻,有趣的是可见少量细胞为蓝黄混合色.结论在胃窦粘膜幽门腺内分泌细胞中,可见5-HT与胃泌素的共存.  相似文献

胃溃疡(Gu)、十二指肠溃疡(Du)和对照组(Con)各5例,取肉眼下非病变区胃窦粘膜,用免疫组织化学PAP双重染色法,在同一张切片上显示5-HT细胞和G细胞。结果5-HT细胞多散在于粘膜的中下1/3,呈棕褐色,形状多样,常见有突起的细胞。G细胞呈带状分布于粘膜的中1/3,大部分G细胞为蓝色,个别细胞为蓝黄混合色。它们多为圆形或锥体形。有时可见5-HT细胞和G细胞相毗邻。5-HT细胞数和G细胞数在Gu组分别为:4.17±1.21、7.87±3.13;Du组:5.45±1.96、9.21±5.72;Con组:3.21±1.64、6.19±1.72。5-HT细胞数和G细胞数在Gu组和Du组都增加。且都以Du组为最多,经相关分析二者呈正相关关系。Gu和Du组的5-HT细胞数和G细胞数与对照组相比均有高度显著性差异。Du组的5-HT细胞数和G细胞数与Gu组相比较,亦有高度显著性差异。提示5-HT细胞和G细胞可能参与了消化性溃疡病的调节,特别与Du的关系更为密切。5-HT细胞和G细胞之间的关系有待于进一步研究。  相似文献

采用放射免疫法测定15例活动期十二指肠溃疡(Du组 n=15)及36例浅表性胃炎患者(Con组n=36)血浆及胃窦粘膜β-内啡肽含量;应用免疫组织化学方法、图象分析和免疫金标电镜观察胃窦粘膜β-内啡肽阳性细胞灰度变化以及超微结构特点。结果发现,Du患者胃窦粘膜β-EP含量为62.80±19.81ng/g,高于Con组,37.42±12.64ng/g,有显著性差异(P<0.01);β-EP阳性细胞灰度密度增大为0.22±0.04μm-2,与Con组0.14±0.04μm-2相比,差异显著,(P<0.01);细胞内内质网和线粒体丰富,基质密度增高,5nm胶体金标记的颗粒散在于细胞基部。结果提示,β-EP阳性细胞的分泌活动增强,提示其可能参与十二指肠溃疡的调节。  相似文献

目的探讨幽门螺杆菌(Hp)与反流性食管炎(RE)的关系.方法对303例内镜诊断RE的患者取胃窦活检,采用Warthin-Starry嗜银染色检测Hp阳性率,并设慢性胃炎对照组进行比较.结果RE组与对照组Hp的阳性率分别为33%、56.82%,两组比较Hp阳性率有明显差异(P<0.01).RE组I、II、III级Hp的阳性率分别为51.37%、16%、14.54%;I、II级比较差异有显著性(P<0.05),I、III级比较差异有显著性(P<0.05),II、III级比较差异无显著性(P>0.05).结论RE组Hp的阳性率低于对照组;随着Hp的阳性率下降,RE炎症程度逐渐加重.  相似文献

本文应用免疫组织化学PAP法,对硫脉致甲状腺功能低下(甲低)小鼠胃窦粘膜5—羟色胺及生长抑素样免疫反应细胞,进行免疫组织化学染色。结果发现:甲低时胃窦部5—羟色胺细胞数目减少(P<0.01),染色变浅,形态无明显变化。而生长抑素免疫反应细胞数目有增加趋势。此结果提示:甲状腺素可影响胃窦粘膜EC细胞中5—羟色胺的合成。  相似文献

Objective: To investigate the distribution of H. pylori antigens in the gastric mucosa in patients with H. pylori infection, and the relationship between the distribution and gastric cancer. Methods: Of 112 patients confirmed by pathological study to have chronic superficial gastritis, precancerous changes (chronic atrophic gastritis, intestinal metaplasia or atypical hyperplasia) and gastric cancer, 28 were H. pylori negative and 84 were H. pylori positive. H. pylori antigens in the gastric mucosa were detected by immunohistochemistry. Results: The H. pylori positive group, comprised 12 of 22 (50.0%) in the chronic superficial gastritis group, 22 of 25 (88.0%) in the precancerous changes group and 13 of 35 (37.1%) in the gastric cancer group. The positive rates of H. pylori antigens in the cytoplasm progressively increased, respectively at 0.0% (0/12),63.6% (14/22) and 84.6% (11/13) for the same groups (χ2=19.76, P=0.000); H.pylori antigens were located in the mucus layer and above the neck of the mucosal gland in 9 of 12 (75.0%) cases with chronic superficial gastritis, at the neck of the mucosal gland and the isthmus in 12 of 22 (54.5%) cases with precancerous changes, below the isthmus in 9 of 13 (69.2%) cases with gastric cancer (x2=25.30, P=0.000). In the H. pylori negative group, no H.pylori antigen was observed. Conclusion: With the progression of chronic superficial gastritis→precancerous changes→gastric cancer, H. pylori antigens progressively migrated from the outer part to the inner part of the cell, and from the superficial to the deep gastric mucosa.  相似文献

目的 观察大鼠实验性胃溃疡期间大脑皮层生长抑素免疫反应(SS-ir)和SSmRNA神经元的变化。方法 免疫细化PAP法;地高辛标记反意SScRNA探针mRNA原位杂交组化法。结果 正常大鼠SS-ir神经元广泛分布于大脑皮层。溃疡术后1d,大脑皮层纹状皮质区各层SS-ir神经元减少,但可见大量串珠样SS强阳性纤维,溃疡组SS-ir神经元数与盐水组相比无差异;术后4d,溃疡组大脑皮层纹状皮质区各层SS-ir神经元密集、数量明显增多,与盐水组、正常组相比均有差异(P<0.05,P<0.01或P<0.001)。术后10和23d,以上核区SS-ir神经元数仍高于盐水组和正常组(P<0.01,P<0.001)。术后4d,溃疡组大脑皮层SSmRNA神经元数量明显多于盐水组(P<0.001)。结论 大鼠实验性胃溃疡时期,大脑皮层SS-ir和SSmRNA神经元可能参与了胃溃疡自愈过程的调节。  相似文献

取胃镜活检标本93例,SP免疫组化法检测bcl-2、c-myc基因蛋白。结果表明bcl-2基因蛋白表达率在伴有上皮异型增生(GED)和肠化生(IM)的慢性浅表性胃炎(CSG)、慢性萎缩性胃炎(CAG)明显高于不伴GED、IM组(P<0.05-0.01)。c-myc表达则差异不明显P1>0.05。GC及伴有GED、IM癌旁粘膜与不伴GED、IM的癌旁粘膜比较,bcl-2与c-myc的表达差异均非常显著(P<0.01)。bcl-2、c-myc共同表达阳性率在CSG、CAG、癌旁粘膜中伴有GED和IM组及胃癌组分别为60.0%、34.8%、54.5%、36.8%。而无GED、IM组均无阳性表达(P<0.05-0.01)。bcl-2、c-myc阳性表达与患者年龄、性别无关(P>0.05)。本文对它们表达的意义进行了讨论。  相似文献

This study was aimed to investigate the level of sEGF in CAG with different therapies of either Chinese traditional medicine or western medicine. Patients were divided into spleen-reinforcingⅠ, spleen-reinforcing Ⅱ,dampness-resolving and western medicine group by Differentiation Syndrome. Healthy volunteers acted as controls. The sEGF was investigated before and after treatment. The level of sEGF in CAG was higher than that in control group(P<0.01). In patients of spleen-reinforcingⅠgroup and spleen-reinforcing Ⅱgroup, the symptoms and pathological manifestations were improved dramatically (P<0.01) and sEGF dropped markedly after treatment(P<0.01 and P<0.05).The level of sEGF in the dampness-resolving group increased after treatment (P<0.05). But the level of sEGF in the western-medicine group dropped a little after treatment (P>0.05). The inflammation of gastric mucosa may cause the elevation of sEGF in CAG reflectively. After being effectively treated with Chinese traditional medicine, the symptoms of CAG improved simultaneously with the return of sEGF. SEGF is a sensitive index to prognosis of CAG.  相似文献

Objectives: To explore the mechanism of development and aggressiveness in gastric carcinomas by investigating the expression and role of CD97 and its cellular ligand CD55 in gastric carcinomas. Methods: Tumor and corresponding normal mucosal tissue, collected from 39 gastric carcinoma patients, were examined by immunohistochemistry and RT-PCR for the expression of CD97 and CD55. Results: CD97stalk was strongly stained on scattered tumor cells or small tumor cell clusters at the invasion front of gastric carcinomas. The expression of CD97stalk was frequently observed in tumors of stage Ⅰ and T1 gastric carcinoma patients. The expression of CD97stalk between Stage Ⅰ and Stage Ⅱ, Ⅲ, Ⅳ specimens showed significant difference (P<0.05), between T1 and T2, T3, T4 specimens also showed significant difference (P<0.05). Specimens with tumor invasion depth limited in mucosa of T 1 specimens showed higher positive CD55 expression than specimens with the same tumor invasion depth in T2, T3, T4 specimens, the expression of CD55 between T1 and T2, T3, T4 specimens was significantly different (P<0.05).There was strong correlation between the distribution patterns of CD97stalk and CD55 on tumor tissues (r=0.73, P<0.05). Signet ring cell carcinomas frequently contained strong CD97stalk and CD55-staining. Conclusions: Our results suggest that CD97stalk is probably involved in the growth, invasion and aggressiveness of gastric carcinomas by binding its cellular ligand CD55. CD97stalk and CD55 could be useful as molecular markers for prognosis and therapy of gastric carcinoma patients.  相似文献
