以籼稻的成熟胚为实验材料,经过成熟胚愈伤组织的诱导、继代培养,探索研究不同浓度激素和潮霉素对愈伤组织生长的影响以及农杆菌对转化效率的影响.结果显示,最适宜愈伤组织分化的6-BA浓度为2.0 mg·L-1,分化率达82.3%.潮霉素最佳浓度为30 mg·L-1时可以完全抑制愈伤组织的分化.该实验以杂交水稻的亲本籼稻不育系15S为材料,建立了农杆菌转化的体系,以期获得转PEPC基因的籼稻亲本,为杂种优势利用和生理育种的结合提供依据. 相似文献

以金边瑞香茎段为外植体,探讨了不同培养基对愈伤组织诱导、愈伤组织分化的影响。结果表明:1)金边瑞香茎段愈伤组织诱导以培养基MS 2,4-D2.0 mg.L-1 6-BA0.2 mg.L-1效果最佳;2)愈伤组织增殖宜采用培养基MS 2,4-D2.0 mg.L-1,并且增殖培养不宜超过1代;3)愈伤组织分化成苗宜采用培养基1/2MS 6-BA2.0 mg.L-1 NAA0.2 mg.L-1,对愈伤组织进行脱水处理能提高分化率。 相似文献

以自交亲和芸芥7-5-1-1为研究材料,对影响芸芥花药愈伤组织诱导的主要因素进行了初步研究.结果表明:MS+2.0 mg/L 2,4-D+1.0 mg/L 6-BA+0.5 mg/L NAA是诱导芸芥花药愈伤组织适宜的培养基;芸芥花药在离体培养前最佳低温(4℃)预处理时间为2d,诱导率可高达46.2%;在诱导期间始终处于弱光或完全黑暗条件下的花药愈伤组织诱导率最高;单核期花药的愈伤组织诱导率最高,与单核前期、双核期花药的愈伤组织诱导率相比,差异达到极显著水平. 相似文献

以一株无患子优树嫩叶为外植体,采用多因素正交试验筛选其愈伤组织诱导、体胚分化及植株再生的适宜培养基组成。结果表明：6-BA对无患子叶片愈伤组织诱导的影响显著,在添加1.0 mg·L^-16-BA和0.1 mg·L^-1NAA的MS培养基上培养21 d,愈伤组织诱导率达92.5%;愈伤组织在添加1.0 mg·L^-16-BA、0.5 mg·L^-12,4-D和1.0 mg·L^-1NAA的MS培养基上经3次继代,可分化出体胚,分化率为33.4%;初分化体胚在MS＋1.0 mgo L-1 6-BA＋0.1 mg·L^-1NAA＋60 g·L^-1蔗糖的培养基上培养21 d后可转变为成熟体胚,成熟率45%;成熟体胚在添加1.0mg·L^-16-BA和0.5 mg·L^-1IBA的1/2 MS培养基上可萌发生根,培养30 d后生根率为68.2%。 相似文献
