This study examined the effects of dietary α-tocopheryl acetate supplementation on antioxidant enzyme activities and fillet quality in commercial-size Sparus macrocephalus. Three hundred fish [main initial weight (350±12) g] were divided into three groups (E250, E500 and El000) and reared in 9 cages. The fish were fed for 8 weeks with three diets containing different levels of dietary α-tocopheryl acetate (289, 553, 1069 mg/kg). Over the experimental period, fish were fed to satiation and reached a final mean weight of (465±28) g without significant body weight difference and proximate composition difference. Fillet α-tocopherol was significantly (P〈0.05) different between groups, reaching levels of 14.2, 22.1, 30.9 pg/mg fillet for groups E250, E500 and El000, respectively. Total serum superoxide dismutase (SOD) activity increased significantly (P〈0.05) in fish fed the diets high in α-tocopheryl acetate, but serum glutathione peroxidase (GPX) activity was unaffected. In storage on ice, fillets of fish fed the diets high in α-tocopheryl acetate exhibited significantly lower (P〈0.05) levels of oxidation. These results suggested that increased dietary α-tocopheryl acetate could increase its flesh deposition, increase the activity of SOD and prevent lipid peroxidation ofSparus macrocephalus fillets in retail storage on ice.  相似文献

The study was conducted to investigate fasting effects on flesh composition and antioxidant defenses of market-size Sparus macrocephalus. Two hundred fish (main initial weight 580 g) were divided into two groups (control and fasted) and reared in 6 cages. After two weeks of adaptation, group I fasted for 28 d; group II was fed normally as a control. In 3, 7, 14, 21 and 28 d, 6 fish per group were sampled for proximate flesh composition, liver antioxidant enzyme activities and malondialdehyde flesh content analyses. In fasted fish, the reduction of lipid content in muscle occurred after day 3, and, compared to controls, the content of protein decreased from day 14, the activities of liver antioxidative enzymes superoxide dismutase (SOD) and glutathione peroxidase (GPX) increased from day 3, and flesh malondialdehyde levels increased from day 21. Flesh fat reduction shows that fasting may be used as a technique to reduce flesh lipid content in Sparus macrocephalus. However, considering flesh protein loss and the subsequent oxidative stress, the fasting technique should be used with precautions.  相似文献

Gestational diabetes mellitus(GDM) is characterized by glycemia and insulin disorders.Bile acids(BAs) have emerged as vital signaling molecules in glucose metabolic regulation.BA change in GDM is still unclear,which exerts great significance to illustrate the change of BAs in GDM.GDM patients and normal pregnant women were enrolled during the oral glucose tolerance test(OGTT) screening period.Fasting serums were sampled for the measurement of BAs.BA metabolism profiles were analyzed in both pregnant women with GDM and those with normal glucose tolerance(NGT).Delivery characteristics,delivery gestational age,and infant birthweight were extracted from medical records.GDM patients presented distinctive features compared with NGT patients,including higher body mass index(BMI),elevated serum glucose concentration,raised insulin(both fasting and OGTT),and increased hemoglobin A1 c(HbA1c) levels.Higher homeostasis model assessment of insulin resistance(HOMA-IR) and decreased β-cell compensation(i.e.,oral disposition index(DIo)) were also prevalent in this group.Total BAs(TBAs) remained stable,but glycodeoxycholic acid(GDCA) and taurodeoxycholic acid(TDCA) levels declined significantly in GDM.GDCA was inversely correlated with HOMA-IR and positively correlated with DIo.No obvious differences in clinical outcome between the GDM and NGT groups were observed.However,GDM patients with high HOMA-IR and low DIo tended to have a higher cesarean delivery rate and younger delivery gestational age.In conclusion,GDCA provides a valuable biomarker to evaluate HOMA-IR and DIo, and decreased GDCA levels predict poorer clinical outcomes for GDM.  相似文献

Objective: To investigate the potential and early effect of hypertonic saline resuscitation on T-lymphocyte subpopulations in rats with hemorrhagic shock. Methods: A model of rat with severe hemorrhagic shock was established in 18 Sprague-Dawley (SD) rats. The rats were randomly divided into Sham group, HTS group (hypertonic saline resuscitation group) and NS group (normal saline resuscitation group). Each group contained 6 rats. The CD4~ and CD8~ subpopulations of T-lymphocytes in peripheral blood were detected respectively before shock and after resuscitation by double antibody labelling and flow cytometry. Results: In the early stage after hemorrhagic shock, fluid resuscitation and emergency treatment, the CD4^+ lymphocytes of peripheral blood in HTS and NS groups markedly increased. Small volume resuscitation with HTS also induced peripheral CD8^+ lymphocytes to a certain extent, whereas NS resuscitation showed no effect in this respect. Consequently, compared with Sham and HTS groups, CD4^+/CD8^+ ratio of peripheral blood in NS group was obviously increased, and showed statistically differences. Conclusion: In this model of rat with severe hemorrhagic shock, small volume resuscitation with HTS is more effective than NS in reducing immunologic disorders and promoting a more balanced profile of T-lymphocyte subpopulations regulating network.  相似文献

Objectives:To investigate the intestinal microflora status related to ischemia/reperfusion(I/R)liver injury and explorethe possible mechanism.Methods:Specific pathogen free grade Sprague-Dawley rats were randomized into three groups:Controlgroup(n=8),sham group(n=6)and I/R group(n=10).Rats in the control group did not receive any treatment,rats in the I/R groupwere subjected to 20 min of liver ischemia,and rats in the sham group were only subjected to sham operation.Twenty-two hourslater,the rats were sacrificed and liver enzymes and malondialdehyde(MDA),superoxide dismutase(SOD),serum endotoxin,intestinal bacterial counts,intestinal mucosal histology,bacterial translocation to mesenteric lymph nodes,liver,spleen,andkidney were studied.Results:Ischemia/reperfusion increased liver enzymes,MDA,decreased SOD,and was associated withplasma endotoxin elevation in the I/R group campared to those in the sham group.Intestinal Bifidobacteria and Lactobacillidecreased and intestinal Enterobacterium and Enterococcus  相似文献

Objective: Our aim was to investigate clinical and laboratory characteristics of osteoarthritic patients who had amyloid deposition in their knee joints. Methods: Synovial membranes were obtained from 36 patients with knee osteoarthritis (OA) who underwent joint replacement surgery. From this sample, the diagnosis of amyloid was deter- mined by Congo red staining, which demonstrated apple-green birefringence under a polarized microscope. All syn- ovial membranes were immunohistochemically characterized for the expressions of amyloid immunoglobulin light chain (AL-K and AL-,k), serum amyloid-A (SAA), amyloidogenic transthyretin (ATTR), and amyloidogenic 152- microglobulin (A152M). Matrix-assisted laser desorption-ionizaton/time of flight mass spectrometry (MALDI-TOF MS) was used to analyze transthyretin (TTR) isoforms in the serum of each patient. Results: Nine cases (25%) were found to be amyloid-positive. Immunohistochemicaliy, eight cases (88.9%) had ATTR deposition, and one sample (11.1%) was shown to be AL-K-positive. MALDI-TOF MS identified that the TTR in the serum of the patients was unmodified wild-type TTR, TTR-Cys-S-S-Cys, and TTR-Cys-S-S-CysGly. The age at surgery and the disease duration were sig- nificantly higher in the ATTR-positive group than in the ATTR-negative group. Knee score and function score were significantly lower in the ATTR-positive group than in the ATTR-negative group. Conclusions: Amyloid deposition in synovial membranes of OA patients was found to be ATTR and AL-K. TTR in the serum of the patients was unmodified wild-type TTR together with two isoforms. The high age at surgery, long disease duration, and a deteriorated knee function were associated with ATTR amyloid deposition in the osteoarthritic knee joints.  相似文献

Objective: To observe the effect ofangiotensin Ⅱ (Ang Ⅱ) on expression of gap junction channel protein connexin 43 (Cx43) in the proliferation process of vascular smooth muscle cells (VSMCs) during the early stage of arteriosclerosis. Methods: Thirty-two adult male rabbits were randomly divided into 4 groups. Rabbits in Group A were fed common diet while others in Groups B, C, and D were fed high-cholesterol diet. Losartan (10 mg/(kg.d)) and ramipril (0.5 mg/(kg-d)) were added in the diet of Groups C and D, respectively. The animals were sacrificed after 8 weeks and abdominal aortas were removed and dissected. The expression of Cx43 was assayed using RT-PCR and Western Blotting analysis. Results: Cx43 was increased markedly in both protein and mRNA level in Groups B, C, and D fed high-cholesterol diet compared with that in control group (P〈0.01). Cx43 level in losartan or ramipril treated groups was higher than that in control group (P〈0.01, P〈0.05), but lower than that in high-cholesterol diet groups (P〈0.05, P〈0.01). Conclusion: Cx43 level was upregulated in VSMCs during early atherosclerosis. Losartan and ramipril can inhibit the expression of Cx43.  相似文献

Background: Electrochemical therapy (ECT) has been used to treat unresectable hepatic tumor. In order to improve its efficacy, we combined ECT with hyperthermia induced by electrothermal needle (ETN) (ETECT). The aim of this study is to investigate the destructive effect of ETECT on normal rat liver. Methods: Twenty rats were randomized into 4 treatment groups (n=5 in each group): control, ECT alone, hyperthermia alone and ETECT. Following the treatment, sections of the livers were histologically examined by light microscopy and the destructive volumes were measured with micrometer. Results: We found that the destructive volumes in ETECT group were the largest (P〈0.01). In ETECT group coagulative necrosis was found in both anode and cathode areas, around which transition zones existed. The transition zones can only be seen when coulomb was increased in ECT group. Conclusion: ETECT was demonstrated to enhance the destructive effect of ECT. This study provides theoretical and experimental basis for a new local ablative treatment for unresectable primary liver tumor.  相似文献

目的:观察脂肪肝大鼠在生化、病理、脂肪代谢、细胞因子、胰岛素及瘦素各方面指标的变化.方法:雄性Wistar大鼠22只,按体重层次随机分为2组,对照组(A)11只,脂肪肝组(B)11只.对照组饲以普通鼠饲料,脂肪肝组饲以高脂饲料.8周后,全部处死,取血及肝组织,测定血中AIT、AST、TG、CHO、INS、FBS、FFA、Ieptin、II-6、TNF.肝组织匀浆测定肝组织TC,TG,SOD、MDA,观察肝脏组织学变化.结果:脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素表达增高;细胞因子升高.结论:实验性高脂饮食脂肪肝大鼠存在瘦素抵抗及胰岛素抵抗.  相似文献

目的:观察姜黄素对胰岛素抵抗(IR)大鼠糖脂代谢及血清脂肪细胞因子的影响,探讨姜黄素干预IR的作用机制。方法:采用高脂饲料喂养,建立IR大鼠模型。将IR大鼠随机分为3组:模型组、吡咯列酮组和姜黄素组。给药4周后测定空腹血糖、胰岛素、血脂、TN F-α、Leptin等指标。结果:模型组大鼠血清FIN S、TC、TG和LDL水平均较空白组显著升高,而ISI及HDL水平显著下降;血清TN F-α、Leptin及FFA水平也较空白组明显升高;用姜黄素干预后,大鼠ISI较模型组显著升高,血脂得到改善,FINS、TNF-α、Leptin及FFA水平均显著下降。结论:姜黄素可改善IR大鼠糖脂代谢紊乱,增加胰岛素的敏感性,其机制可能与其调节脂肪细胞因子TNF-α、Leptin的分泌有关。  相似文献

The hepatoprotective potential of earthworm extract (EE) (Lampito mauritii, Kinberg) was evaluated against paracetamol-induced liver injury in Wistar albino rat, in comparison with silymarin, the standard hepatoprotective drug. We observed a reduction in liver antioxidants, such as glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx),and catalase (CAT) and in serum total protein, and an increase in serum alkaline phosphatase (ALP), serum aspertate aminotranferase (AST), serum alanine aminotranferase (ALT), bilirubin and liver thiobarbituric acid reactive substances (TBARS) due to liver injury in the paracetamol-administered rats (2 g/kg). On the contrary, increased activities of liver GSH, SOD, GPx,CAT and serum total protein level, and decrease in the contents of serum ALP, AST, ALT, bilirubin and liver TBARS were observed in rats administered with different doses of EE (100, 200 and 300 mg/kg), which are similar to the activities of hepatoprotective drug silymarin (150 mg/kg). The mode of action of EE as evidenced by the above parameters may suggest that EE, on the one hand, prevents the formation of the reactive oxygen groups, or scavenges these groups, thereby preventing the damage on the hepatic cells, and, on the other hand, modulates the genes responsible for synthesis of antioxidant enzymes such as GPx, CAT and SOD in liver tissue and decreases the serum enzymatic activities such as ALP, AST and ALT.  相似文献

144只一日龄的AA肉仔鸡随机分为4组(n=36),预饲一周后分别在试验组基础日粮中添加50mg/kg、100mg/kg、150mg/kg的半胱胺,分别于2、4、6周龄采颈静脉血,分离血清,测定四组血清钙、磷和葡萄糖含量及ALT、AST、ALP活性,研究半胱胺对肉鸡部分血清化学参数及酶活性的影响.结果表明,饲料中添加适量的Cs能升高AA肉鸡的血钙水平,而对鸡血清磷、葡萄糖含量没有明显影响;添加50~100mg/kg的Cs对ALT、AST活性没有明显的影响;而饲料中添加Cs对ALP活性有一定影响.  相似文献

40只雄性SD大鼠随机分成5组,分别是正常组、模型组和野鸦椿高、中、低剂量,采用50%乙醇灌胃造模,同时给相应剂量野鸦椿水提物,给药一周后检测大鼠血清AST、ALT、TBIL、DBIL以及TG含量,并取肝组织HE染色进行病理学检.通过以上处理后,野鸦椿水提物各剂量组大鼠血清AST、ALT、TBIL、DBIL及TG均不同程度的下降,病理组织学检查给药组肝组织病理损害也较模型组轻;以上结果说明野鸦椿水提物对大鼠急性酒精性肝损伤具有保护作用.  相似文献

文章研究了不同剂量甲醛等挥发性物质对发育期大鼠体重、血液常规指标、肝肾功能的影响。研究结果表明:甲醛等挥发性物质的混合物能显著降低大鼠体重,而对肝、肾重量无明显影响;可明显提高大鼠血液中血小板(PLT)和嗜酸性粒细胞(EOS)的含量,但对血液中其它常规指标无明显影响;能明显提高大鼠血清中的谷丙转氨酶(ALT)和天冬氨酸氨基转移酶(AST)的活性.对血清中碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(GGT)、血尿素氮(BUN)和肌酐(Cr)无明显影响。  相似文献

目的:探讨葡萄籽原花青素对大鼠肝缺血再灌注损伤的抗氧化作用、凋亡相关蛋白Bcl-2和Bax表达的影响。方法:SD雄性大鼠30只,复制肝缺血再灌注损伤模型。随机分为:假手术组;HIRI模型组;葡萄籽原花青素预处理(GSPE)组(160 mg/kg),每组10只。于再灌注后3 h检测肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量及血清谷丙转氨酶(ALT)、谷草转氨酶(AST)的含量;光镜下比较各组肝组织形态学的变化;免疫组化法检测GSPE对凋亡相关蛋白Bcl-2,Bax表达的影响。结果:与HIRI模型组比较,GSPE预处理组血清ALT、AST水平明显降低(P〈0.05);肝组织GSH-Px、SOD的活性明显升高(P〈0.05),而MDA含量下降(P〈0.05)。光镜下GSPE预处理组肝细胞损伤程度较模型组明显减轻;免疫组化结果显示,GSPE预处理组肝组织Bcl-2蛋白的表达量较模型组明显增多,而Bax蛋白的表达量则较模型组明显减少。结论:GSPE对缺血再灌注肝脏具有一定的抗氧化损伤和抗凋亡的作用。  相似文献

[目的]观察复方甘草甜素(美能)对慢性乙型肝炎的治疗作用.[方法]将病人随机分为为两组,治疗组10例,用美能40 m l/d.iv.3周后改为口服美能片75 mg,每日3次.对照组1 0例,使用甘利欣150 mg/d,VD.3周后改为口服甘利欣100 mg,每日3次.[结果]治疗组治疗后症状缓解率75%;血清ALT水平由306.70±229.33 u/L降为105.00±86.11 u/L;血清AST水平由137.90±71.44 u/L降为64.30±34.93 u/L;血清SB水平由31.80±28.90 umol/L降为16.20±3.43 umol/L.对照组治疗后症状缓解率为66.7%;血清ALT水平由208.20±160.13 u/L降为77.20±65.92 u/L;血清AST水平由92.60±61.19 u/L降为51.20±31.38 u/L;血清SB水平由20.60±5.08 umol/L降为11.70±2.06 umol/L.[结论]复方甘草甜素(美能)的临床疗效与国内同类产品(甘利欣)相仿,具有较好的保肝、降酶作用,有一定的优势.  相似文献

目的:探讨乙型肝炎患者机体抗氧化能力的变化及其与病情的关系.方法: 门诊乙型肝炎患者共124例和健康者30例,均经放射免疫方法检测后加以确诊.乙型肝炎患者分为三组:乙型肝炎病毒复制组、乙型肝炎病毒恢复组、乙型肝炎合并肝癌组.患者组和对照组均经7150型全自动生化分析仪测定血清ALT、AST含量,应用改良TBA微量法测定血清丙二醛(MDA),以黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)、Cu/Zn-SOD及Mn-SOD水平的变化.结果: 乙型肝炎病毒复制组、乙型肝炎病毒恢复组、乙型肝炎合并肝癌组三组患者血清MDA水平均显著高于对照组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于对照组(P<0.01);乙型肝炎病毒复制组、乙型肝炎合并肝癌组患者血清MDA水平均显著高于乙型肝炎病毒恢复组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于乙型肝炎病毒恢复组(P<0.01);乙型肝炎合并肝癌组患者血清MDA水平均显著高于乙型肝炎病毒复制组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于乙型肝炎病毒复制组(P<0.01).但血清MDA水平与ALT、AST水平(r=0.151),血清SOD、Cu/Zn-SOD及Mn-SOD水平与ALT、AST水平(r=-0.218)均无相关性(P>0.05).结论: 乙型肝炎患者出现明显抗氧化能力的异常.通过血清MDA、SOD、Cu/Zn-SOD及Mn-SOD水平的检测,可为乙型肝炎的临床防治及疗效观察提供一定的依据.  相似文献

目的观察慢性充血性心力衰竭(CHF)患者的血胰岛素和糖代谢变化,并探讨依那普利对CHF患者胰岛素敏感性的影响。方法CHF患者69例,依那普利(n=37)(第一周10mg,1次/d,后三周20mg,1次/d)和地高辛(n=32)(第一周0.25mg,1次/d,后三周0.125mg,1次/d),治疗前和治疗4周后检测空腹血糖(FPG)、空腹血清胰岛素(FINS)和空腹血清C肽(CP)水平,并计算出FPG/FINS比值、胰岛素敏感性指数IAI值[IAI=-ln(FPG×FINS)]作为机体胰岛素敏感性评价指标,分别比较两组患者治疗前后的胰岛素敏感性变化。结果治疗4周后,依那普利组CHF患者的胰岛素敏感性显著增加,而地高辛组患者胰岛素敏感性无显著变化。结论依那普利有改善CHF患者IR的作用。  相似文献
