Because of the superior features of boron-10 isotope in absorbing hot neutrons, boric-10 acid(H310BO3)has been used widely in nuclear industry. Boric-10 acid is obtained from boron-10 trifluoride(10BF3) by esterification and hydrolyzation. In this study, trimethyl borate-10 [(CH3O)310B] was prepared from boron-10 trifluoride(10BF3)through chemical exchange rectification. In addition, the hydrolyzation of trimethyl borate-10 was investigated with different p H values, temperatures and molar ratios of water to trimethyl borate-10. Under the optimum process conditions, high yield of boric-10 acid with nuclear industrial purity grade was realized.  相似文献

In order to carry out decolorization, sludge protein solution, a dark brown close to black solution from activated sludge, was subjected to 60Co γ-ray irradiation in the presence of hydrogen peroxide. UV/Vis spectrophotometric method was used to investigate the effect of H2O2 on the coloration apparent kinetics and rate constants of sludge protein solution under γ-ray irradiation. In addition, the effects of irradiation dose, initial sludge protein solution concentration, and pH value on the decolorization efficiency of sludge protein solution were studied. Results showed that the decolorization apparent kinetics of sludge protein solution was a first-order reaction. The solution decolorization percentage increased with the increase of irradiation dose or the decrease of initial sludge protein solution concentration. The examination results of pH value showed that the sludge protein solution could be more efficiently decolorized in alkaline media than in acid media. Moreover, sensory evaluation and foamability analysis indicated that irradiated samples under H2O2 oxidation showed better sensory score and foamability.  相似文献

Using degenerate primers and RT-PCR,RACE techniques,a 1491 bp cDNA segment of stearoyl-acyl carrier protein desaturase(SAD)is cloned from developing seeds of Jatropha curcas L.The segment contains a 1191 bp of complete open reading frame(ORF).Analysis in the BLAST on NCB! shows that Jatropha curcas SAD(JSAD)gene encodes a protein precursor composed of a signal peptide of 33 amino acids and a mature peptide of 363 amino acids.The homological analysis shows that JSAD has high level of homology both in nucleotide sequence and in amino acid sequence to other plants SADs.The nucleotide and peptide identity of JSAD to Ricinus communis SAD(RSAD)is up to 89% and 96.2% respectively.Molecular modeling of JSAD indicates that its three-dimensional structure strongly resembled the crystal structure of RSAD.  相似文献

Objective:To evaluate the effect of chitosan(CH) and hydrogen peroxide(H2O2) seed coatings and seedling sprinklings on two different maize varieties by measuring their phenology,the H2O2 presence,the catalase(CAT) activity,and the protein quantity.Methods:Seven groups of ten seeds for each maize variety were treated with CH(2%(20 g/L) and 0.2%(2 g/L)) or H2O2(8 mmol/L) by coating,sprinkling,or both.Germination and seedling growth were measured.One month after germination,the presence of H2O2 in seedlings in the coated seed treatments was evaluated.Protein content and CAT activity were determined under all treatments.Results:H2O2 seed coating enhanced the germination rate and increased seedling and stem length in the quality protein maize(QPM) variety.Seedlings had a higher emergence velocity under this treatment in both varieties.CH and H2O2 sprinklings did not have an effect on seedling phenology.Exogenous application of H2O2 promoted an increase of endogenous H2O2.CH and H2O2 seedling sprinkling increased the protein content in both maize varieties,while there was no significant effect on the CAT activity of treated seeds and seedlings.Conclusions:CH and H2O2 enhance some phenological and biochemical features of maize depending on their method of application.  相似文献

Objective: To study the roles of different truncated hepatitis C virus (HCV) core proteins (CORE) in the pathogenesis of HCV persistent infection and hepatocellular carcinoma (HCC) and to assess intracellular localization in transiently transfected cells. Methods: Seven truncated CORE-GFP (green fluorescent protein) fusion protein expression plasmids were constructed,which contained HCV CORE sequences derived from tumor tissues (BT) and non-tumor tissues (BNT) from one patient infected with HCV. Amino acid (aa) lengths were BT: 1-172 aa, 1-126 aa, 1-58 aa, 59-126 aa, 127-172 aa; BNT: 1-172 aa and C191:1-172 aa respectively. Subcellular localization of CORE-GFP was analyzed by con-focal laser scanning microscope. Apoptosis and necrosis were quantified by flow cytometry. Results: Different truncated CORE-GFP localized mainly in the cytoplasm, but nuclear staining was also observed. HCV CORE could induce apoptosis and necrosis, and different truncated COREs could induce cell apoptosis and necrosis at different levels. Among the same length 1-172 aa of BT, BNT and C191, the cell apoptosis and necrosis percentage of BT is highest, and C191 is the lowest (BT>BNT>C191). To the different fragment COREs of BT,N-terminal of CORE induced apoptosis and necrosis higher, compared with that of C-terminal (1-172 aa>1-126 aa>1-58aa> 127-172 aa>59-126 aa). Conclusion: These results suggest HCV CORE could induce apoptosis and necrosis of cells, which might play an important role in the pathogenesis of HCV persistent infection and HCC and the different CORE domains of different HCV quasi-species might have some difference in their pathogenesis.  相似文献

To screen and evaluate protein biomarkers for the detection of gliomas (Astrocytoma grade Ⅰ-Ⅳ) from healthy individuals and gliomas from brain benign tumors by using surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS) coupled with an artificial neural network (ANN) algorithm. SELDI-TOF-MS protein fingerprinting of serum from 105 brain tumor patients and healthy individuals, included 28 patients with glioma (Astrocytoma Ⅰ-Ⅳ), 37 patients with brain benign tumor, and 40 age-matched healthy individuals. Two thirds of the total samples of every compared pair as training set were used to set up discriminating patterns, and one third of total samples of every compared pair as test set were used to cross-validate; simultaneously, discriminate-cluster analysis derived SPSS 10.0 software was used to compare Astrocytoma grade Ⅰ-Ⅱ with grade Ⅲ-Ⅳ ones. An accuracy of 95.7%, sensitivity of 88.9%, specificity of 100%, positive predictive value of 90% and negative predictive value of 100% were obtained in a blinded test set comparing gliomas patients with healthy individuals; an accuracy of 86.4%, sensitivity of 88.9%, specificity of 84.6%, positive predictive value of 90% and negative predictive value of 85.7% were obtained when patient's gliomas was compared with benign brain tumor. Total accuracy of 85.7%, accuracy of grade Ⅰ-Ⅱ Astrocytoma was 86.7%, accuracy ofⅢ-Ⅳ Astrocytoma was 84.6% were obtained when grade Ⅰ-Ⅱ Astrocytoma was compared with grade Ⅲ-Ⅳ ones (discriminant analysis). SELDI-TOF-MS combined with bioinformatics tools, could greatly facilitate the discovery of better biomarkers. The high sensitivity and specificity achieved by the use of selected biomarkers showed great potential application for the discrimination of gliomas patients from healthy individuals and glioma from brain benign tumors.  相似文献

A two-layer method based on support vector machines (SVMs) has been developed to distinguish epoxide hydrolases (EHs) from other enzymes and to classify its subfamilies using its primary protein sequences. SVM classifiers were built using three different feature vectors extracted from the primary sequence of EHs: the amino acid composition (AAC), the dipeptide composition (DPC), and the pseudo-amino acid composition (PAAC). Validated by 5-fold cross tests, the first layer SVM classifter can differentiate EHs and non-EHs with an accuracy of 94.2% and has a Matthew's correlation coefficient (MCC) of 0.84. Using 2-fold cross validation, PAAC-based second layer SVM can further classify EH subfamilies with an overall accuracy of 90.7% and MCC of 0.87 as compared to AAC (80.0%) and DPC (84.9%). A program called EHPred has also been developed to assist readers to recognize EHs and to classify their subfamilies using primary protein sequences with greater accuracy.  相似文献

Good quality deoxyribonucleic acid (DNA) is the pre-requisite for its downstream applications. The presence of high concentrations of polysaccharides, polyphenols, proteins, and other secondary me- tabolites in mango leaves poses problem in getting good quality DNA fit for polymerase chain reaction (PCR) applications. The problem is exacerbated when DNA is extracted from mature mango leaves. A reliable and modified protocol based on the cetyl- trimethylammonium bromide (CTAB) method for DNA extraction from mature mango leaves is described here. High concentrations of inert salt were used to remove polysaccharides; Polyvinylpyrrolidone (PVP) and β-mercaptoethanol were employed to manage phenolic compounds. Extended chloroform-isoamyl alcohol treatment followed by RNase treatment yielded 950?1050 μg of good quality DNA, free of protein and RNA. The problems of DNA degradation,contamination, and low yield due to irreversible binding of phenolic compounds and coprecipitation of polysaccharides with DNA were avoided by this method. The DNA isolated by the modified method showed good PCR amplification using simple se- quence repeat (SSR) primers. This modified protocol can also be used to extract DNA from other woody plants having similar problems.  相似文献

不久前接邹立志君长途电话,说她的博士论《诗歌语体论》经过修改、补充,即将问世了,希望我为之写一篇序言。高兴之余,二年半之前她的论顺利通过答辩的情景,又一次在面前映现。记得当时评委会的专家一致给予高度评价,“决议书”写道:“这是一篇功夫扎实,富有创见,学术价值较高的论。它不仅对于诗歌语体学的建设具有一定的理论和应用价值,对于修辞学科学化、现代化具有促进作用,而且对于语言学理论的研究也具有启发意义。”我认为上述评价是比较恰切的,该书的问世不仅对诗歌语体研究是一次较大的开拓,对整个语体学、修辞学甚至对语言学和学,也都“具有促进作用”和“启发意义”。  相似文献

中学课本中介绍苯的取代反应性质时是以苯和溴反应为例进行实验的.若按课本上的装置和操作,将苯和溴混和好后,再加入催化剂——铁粉,烧瓶内液体急剧沸腾,大量的溴蒸气和溴化氢气体充满烧瓶上面的空间,并通过长导管进入锥形瓶,进一步弥漫到空气中,产生呛人的白雾和窒息的恶臭,造成严重的污染,锥形瓶中的蒸馏水也会因溶有溴呈棕黄色,这时再用AgNO3溶液来检验蒸馏水中的溴离子缺乏说服力.另外,实验结束后,把烧瓶内生成物倒入水中,生成的溴苯因溶有溴而呈褐色,尽管在教学中强调是因为溶有溴而发生了颜色变化。  相似文献
