Homoharringtonine (HHT) has currently been used successfully in the treatment of acute and chronic myeloid leukemias and has been shown to induce apoptosis of different types of leukemic cells in vitro. Emerging evidence suggests that angiogenesis may play an important role in hematological malignancies, such as leukemia. How ever, whether HHT can relieve leukemia by anti-angiogenesis is still unknown. We investigated the anti-angiogenesis potential of HHT with the human umbilical vein endothelial cell line (ECV304) and leukemic cell line (K562) in vitro. Cellular proliferation was determined by MTT assay and apoptosis was analyzed by flow cytometry. The mRNA expression of vascular endothelial growth factor (VEGF) was assessed by RT-PCR and VEGF protein production was detected by Western blot. Inhibition of cell proliferation and induction of apoptosis by HHT were discovered in ECV304 cells, and appeared in a dose- and time-dependent manner. Also, treatment with HHT caused down-regulation of VEGF mRNA expression in K562 cells in similar dose- and time-dependent manner and inhibition of VEGF protein production in K562 cells in response to the enhancing concentration of HHT. The results demonstrated that HHT could also induce apoptosis in endothelium and down-regulate VEGF expression in K562 cells. In conclusion, we believe HHT has anti-angiogenesis potential and speculate that HHT might exert its anti-leukemia effects via reduction of angiogenesis.  相似文献

摘要:目的:探讨间歇运动对心肌梗死(myocardial infarction,MI)大鼠白血病抑制因子(Leukemia inhibitory factor,LIF)表征和心脏血管新生的影响及其可能机制。方法:3月龄SD雄性大鼠,随机分为正常对照组(Control)、间歇运动组(CE)、假心梗组(Sham)、心梗安静组(MI)和心梗+间歇运动组(ME),每组12只。Control组和CE不手术。各心梗组采用左冠状动脉前降支结扎法制备MI模型。运动组在术后1周进行预适应运动(10~15 m/min,30 min/d×5 d/周)。间歇运动采用低强度和高强度依次交替的跑台训练,以10 m/min×10 min进行热身运动,以25 m/min×7 min和15 m/min×3 min依次进行中等强度与大强度交替的间歇运动(60 min/d×5 d/周×8周)。训练结束后次日,血流动力学检测心功能,迅速摘取心脏,RT-qPCR检测心肌LIF/LIFR mRNA表达,Western blot检测心肌LIF、LIFR、p-STAT3/STAT3、VEGF蛋白表达,免疫荧光和免疫组化检测vWF和CD31表达,酶活性试剂盒检测心肌细胞代谢指标。结果:1)间歇运动可激活正常大鼠心肌LIF/LIFR/STAT3通路;2)心梗大鼠心肌LIF/LIFR基因与蛋白表达和STAT3磷酸化水平升高,血管发生代偿性新生,心肌CVF%显著增加,心肌细胞代谢紊乱;3)间歇运动进一步显著上调心梗心肌LIF/LIFR基因与蛋白表达和STAT3磷酸化水平,促进血管新生,显著降低CVF%,改善心肌代谢紊乱。结论:间歇运动显著升高正常和心梗大鼠心肌LIF/LIFR基因和蛋白的表达,其下游信号通路蛋白STAT3的磷酸化水平显著升高,显著刺激心脏的血管再生,改善心梗大鼠心功能。  相似文献

A number of epidemiologic studies has been published in recent years showing an increase risk of death from cancer in subjects with low plasma cholesterol levels. Although several authors proposed that hypocholesterolemia is predisposing factor for cancer development, no causative relation has been established so far and that it may be that low plasma cholesterol is secondary to malignant disease. Hence, the present study was undertaken to examine the lipid profile in children patients with leukemia and Hodgkin's disease in comparison with age matched controls. The study included 52 normal healthy controls and 105 patients with leukemia and Hodgkin's disease. Lipid profile included serum cholesterol, HDL & LDL cholesterol and triglycerides. Serum cholesterol, HDL & LDL cholesterol were found to be inversely associated with incidence of cancer, whereas triglycerides were significantly elevated in cancer patients. The inverse association between cancer and serum cholesterol may reflect a physiological response to early undiagnosed stages of cancer.  相似文献

浙江省白血病患儿父母应对方式的调查研究   总被引:1,自引:0,他引:1  
目的:调查浙江省白血病患儿父母在患儿病程中的主要应对方式,筛选出白血病患儿父母在应对方式上有异常的父母,为护理干预提供依据。方法:用McCubbin的CHIP调查表面对面访谈白血病患儿父母,将调查结果使用SPSS 13.0软件处理。结果:66例患儿父母中有1例父亲和13例母亲在社会支持﹑自尊和稳定(Social Support,Esteem,and Stability,简写S)异常,1例母亲在家庭的完整﹑合作和乐观程度(family Intergration,Cooperation,and Optimism,简写F)异常,最有效应对方式在F方面,而最无效的应对方式是在S方面。结论:浙江省白血病患儿父母应对方式主要采用家庭的支持和合作,与家庭乐观程度有很大关系,而在社会支持方面的应对方式十分欠缺。  相似文献
