The dense granule protein 4 (GRA4) is a granular protein from Toxoplasma gondii, and is a candidate for vaccination against this parasite. In this study, the plasmid pcDNA3. 1-GRA4 (pGRA4), encoding for the GRA4 antigen, was incorporated by the dehydration-rehydration method into liposomes composed of 16 mmol/L egg phosphatidylcholine (PC), 8 mmol/L dioleoyl phosphatidylethanolamine (DOPE), and 4 mmol/L 1,2-diodeoyl-3-(trimethylammonium) propane (DOTAP). C57BL/6 mice and BALB/c mice were immunized intramuscularly three times with liposome-encapsulated pGRA4 to determine whether DNA immunization could elicit a protective immune response to T. gondii. Enzyme-linked immunosorbent assay (ELISA) of sera from immunized mice showed that liposome-encapsulated pGRA4 generated high levels of IgG antibodies to GRA4. Production of primary interferon (IFN)-γ and interleukin (IL)-2 in GRA4-stimulated splenocytes from vaccinated mice suggested a modulated Th1-type response. 72.7% of C57BL/6 mice immunized with liposome-encapsulated pGRA4 survived the challenge with 80 tissue cysts of ME49 strain, whereas C57BL/6 mice immunized with pGRA4 had only a survival rate of 54.5%. When immunized BALB/c mice were intraperitoneally challenged with 103 tachyzoites of the highly virulent RH strain, the survival time of mice immunized with liposome-encapsulated pGRA4 was markedly longer than that of other groups. Our observations show that liposome-encapsulated pGRA4 enhanced the protective effect against infection of T. gondii.  相似文献

Objective: To investigate the anti-tumor efficacy of dendritic cell (DC)-based vaccines pulsed with tumor extracts or RNA in a mouse model of intracranial G422 glioblastoma. Methods: Bone marrow-derived DCs were pulsed ex vivo with tumor extracts or RNA. Ninety female mice harboring 4-day-old intracranial G422 glioblastomas and 126 normal mice were treated with three spaced one week apart subcutaneous injections either with PBS, unpulsed DCs, G422 tumor extracts, RNA, DCs pulsed with G422 tumor extracts (DC/extract) or with RNA (DC/RNA). Seven days after the third immunization of normal mice, the spleens of 36 of them were harvested for cytotoxic T lyphocyte (CTL) assays and the others were challenged in the brain with G422 tumor cells. All the treated mice were followed for survival. Some mice brains were removed and examined pathologically when they died. It has been proposed that activation of Toll-like receptors (TLRs) plays crucial roles in the polarization of adaptive immune responses. A synthetic Toll-like receptor 2 (TLR2) ligand, Pam3CSK4, has been reported to modulate the balance of Thl/Tn2 responses. We evaluated the modulation effect of Pam3CSK4 on allergic immune response in a mouse rhinitis model sensitized to house dust mite allergen (HDM). Mice were sensitized and challenged with Dermatophagoides farinae allergen (Der f), and then the allergic mice were treated by Pam3CSK4. Nasal allergic symptoms and eosinophils were scored. Der f-specific cytokine responses were examined in the splenocytes and bronchoalveolar lavage fluid (BALF). Serum level of total IgE was also detected. After establishing a mouse allergic rhinitis model with HDM, we have showed that Pam3CSK4 treatment not only ameliorated the nasal allergic symptoms remarkably but also decreased the eosinophils and total inflammation cells in BALF significantly. Analysis of cytokine profile found that' IFN-γ released from either BALF or stimulated splenocytes increased markedly in Pam3CSK4-treated mice, while IL-13 decreased significantly. Moreover, serum level of total IgE was significantly lower in Pam3CSK4-treated mice than in the untreated. Thus, in an allergic rhinitis mouse model developed with HDM, Pam3CSK4 was shown to exhibit an antiallergic effect, indicating its potential application in allergic diseases.  相似文献

摘要:观察食用菌——杏鲍菇(Pleurotus eryngii DC.ex Fr.)对小鼠小肠功能的影响.以正常、新斯的明负荷、肾上腺素负荷为研究模型,采用炭末小肠推进试验法.杏鲍菇提取液1次性ig给药12 g.kg-1可显著促进正常小鼠的小肠运动(P<0.05);杏鲍菇提取液对肾上腺素负荷引起小鼠小肠推进抑制有极显著的促进作用(P<0.01);对新斯的明负荷小鼠引起的小肠推进亢进没有明显拮抗作用(P>0.05).杏鲍菇提取液有加强小鼠小肠运动功能的功效.  相似文献

Objective: This study was aimed at evaluating and comparing the quality of life in patients who underwent laparo-scopic and open cholecystectomy for chronic cholecystolithiasis. Methods: The study included 25 patients with laparoscopic cholecystectomy (LC group) and 26 with open cholecystectomy (OC group). The quality of life was measured with the Gastrointestinal Quality of Life Index (GLQI) preoperatively, thereafter regularly at 2, 5, 10 and 16 weeks after the operation. Results: The mean preoperative overall GLQI scores were 112.5 and 110.3 in LC and OC group respectively (P>0.05). In the LC group, the mean overall GLQI score reduced slightly to 110.0 two weeks after the operation (P>0.05). The LC group showed significant improvement in overall score and in the aspects of symptomatology, emotional and physiological status from 5 to 16 weeks postoperatively. In the OC group, the GLQI score reduced to 102.0 two weeks after surgery (P<0.05). Significant reductions were shown in the aspects of symptomatology, physiological and social status. The GLQI scores returned to the preoperative level of 115.6 ten weeks after the operation (P>0.05). The patients experienced significant improvements of GLQI sixteen weeks after OC operation (P<0.01-0.05). Within the 10 postoperative weeks, the LC group had significantly higher GLQI scores than the OC group (P<0.05). Conclusions: LC can improve the quality of life postoperatively better and more rapidly than OC. The assessment of quality of life assessment is a valid method for measuring the effects of surgical treatment.  相似文献

目的:观察镍铬合金烤瓷冠修复后头发和血液中镍铬元素含量的变化,探讨镍铬合金长期存在于口腔中的安全性。方法:选取60名经体检合格的健康人作为正常对照组;60名上、下颌前牙有任意三颗行镍铬合金烤瓷冠修复的患者作为实验组。实验组修复后半年及一年和对照组均采用火焰原子吸收光谱法测定头发镍铬元素含量,采用石墨炉原子吸收光谱法测得其血清中镍铬元素含量。结果:头发镍含量:实验半年组与对照组比较无显著性差异(P>0.05);实验一年组与对照组相比较有显著性差异(P<0.05);实验一年组与半年组比较无显著性差异(P>0.05)。头发铬含量:各组之间比较均无显著性差异(P>0.05)。血清镍含量:实验半年组与对照组比较无显著性差异(P>0.05);实验一年组与对照组相比较有显著性差异(P<0.05);实验一年组与半年组比较无显著性差异(P>0.05)。血清铬含量:各组之间比较均无显著性差异(P>0.05)。结论:镍铬合金烤瓷冠修复后会在口腔环境中释放一定的镍、铬,且释放的量与时间有关,但量很微小,头发和血液中镍铬元素含量呈正相关。  相似文献

目的白细胞介素15(interleukin-15,IL-15)与炎症反应的关系。方法在含兔IL-15全长编码cDNA的质粒上缺失了部份片段.构建成竞争性RT—PCR的竞争子(competitor)。研究炎症反应后IL-15基因的转录情况。结果竞争性RT-PCR结果显示,IL-15与炎症反应密切相关,且脂多糖(LPS)增加了兔子内脏器官(心脏,肺,肾脏,脾脏和肝脏)中IL-15mRNA的表达。结论LPS可以增强IL-15的免疫反应。  相似文献

Objective: To construct a eukaryotic expression plasmid pcDNA3.1 (-)-Humanin. Methods: The recombinant plasmid pGEMEX-1-Humanin was digested with restriction endonucleases BamH I and Hind III and the Humanin gene fragments, about 100 bp length, were obtained. Then the Humanin gene fragments were inserted into eukaryotic expression vector pcDNA3.1 (-) and the recombinant plasmids pcDNA3, l(-)-Humanin were identified by sequencing. Results: Recombinant plasmid DNA successfully produced a band which had the same size as that of the Humanin positive control. The sequence of recombinant plasmids accorded with the Humnain gene sequence. Conclusions: A eukaryotic expression plasmid of Humanin was successfully constructed.  相似文献

This study was aimed to investigate the level of sEGF in CAG with different therapies of either Chinese traditional medicine or western medicine. Patients were divided into spleen-reinforcingⅠ, spleen-reinforcing Ⅱ,dampness-resolving and western medicine group by Differentiation Syndrome. Healthy volunteers acted as controls. The sEGF was investigated before and after treatment. The level of sEGF in CAG was higher than that in control group(P<0.01). In patients of spleen-reinforcingⅠgroup and spleen-reinforcing Ⅱgroup, the symptoms and pathological manifestations were improved dramatically (P<0.01) and sEGF dropped markedly after treatment(P<0.01 and P<0.05).The level of sEGF in the dampness-resolving group increased after treatment (P<0.05). But the level of sEGF in the western-medicine group dropped a little after treatment (P>0.05). The inflammation of gastric mucosa may cause the elevation of sEGF in CAG reflectively. After being effectively treated with Chinese traditional medicine, the symptoms of CAG improved simultaneously with the return of sEGF. SEGF is a sensitive index to prognosis of CAG.  相似文献

目的:探讨急性脑损伤患者血清乳酸脱氢酶变化的临床意义.方法:AbbottAeroset(2000)全自动生化分析仪测定乳酸脱氢酶(LDH).结果:2组病人在急性脑损伤后LDH明显高于对照组(P<0.01),组间比较差异不明显(P>0.05).结论:测定血清乳酸脱氢酶可在一定程度上反应脑损伤的程度,对预后和治疗有一定的指导意义.  相似文献

Resistin, a newly discovered peptide hormone mainly secreted by adipose tissues, is present at high levels in serum of obese mice and may be a potential link between obesity and insulin resistance in rodents. However, some studies of rat and mouse models have associated insulin resistance and obesity with decreased resistin expression. In humans, no relationship between resistin level and insulin resistance or adiposity was observed. This suggests that additional studies are necessary to determine the specific role of resistin in the regulation of energy metabolism and adipogenesis. In the present study, we investigated the effect of resistin in vivo on glucose and lipid metabolism by over-expressing resistin in mice by intramuscular injection of a recombinant eukaryotic expression vector pcDNA3.1-Retn encoding porcine resistin gene. After injection, serum resistin and serum glucose (GLU) levels were significantly increased in the pcDNA3.1-Retn-treated mice; there was an obvious difference in total cholesterol (TC) level between the experiment and the control groups on Day 30. In pcDNA3.1-Retn-treated mice, both free fatty acid (FFA) and high density lipoprotein (HDL) cholesterol levels were markedly lower than those of control, whereas HDL cholesterol and triglyceride (TG) levels did not differ between the two groups. Furthermore, lipase activity was expressly lower on Day 20. Our data suggest that resistin over-expressed in mice might be responsible for insulin resistance and parameters related to glucose and lipid metabolism were changed accordingly.  相似文献

应用溶血素测定方法检测润神口服液对小鼠抗体产生的影响,发现2号口服液比1号口服液有更好稳定体液免疫功能的作用,并且能使雌鼠有效拮抗环磷酰胺的免疫抑制作用(P<0.05)。伴随该实验的饮食测定,以及自然感染死亡率的观察、体力增强测定、常压耐缺氧试验等佐证资料说明,2号口服液确有“扶正固本”的功效,从而提高、稳定机体免疫功能。同时提示2号口服液如在抗肿瘤化疗中应用,可能会明显提高疗效,提高抗感染能力,减轻副作用。  相似文献
