Paraoxonase is an anti-oxidant enzyme, which circulates in the plasma, tightly bound to HDL. This enzyme is known to be synthesized in the liver. This study was carried out in order to ascertain the diagnostic utility of this enzyme in acute liver disease. Serum basal as well as salt (NaCl) stimulated paraoxonase was estimated in 50 patients with an established diagnosis of acute liver disease and also in 50 healthy blood donors. Paraoxonase levels were significantly lower in patients as compared with controls (P < 0.05). The ‘receiver operating characteristic’ plot showed that this enzyme has a high degree of sensitivity and specificity for the diagnosis acute liver disease. Serum PON is likely to emerge as an additional test of liver function, as it encompasses three different attributes of hepatic function namely, synthetic capacity, detoxication and secretory functions.  相似文献

目的:探讨中医个性化心理护理对肝硬化腹水患者一般症状,临床疗效,以及生活质量的影响。方法:在广西中医药大学第一附属医院肝病科住院病人70例,随机分为中医个性化心理护理组(38例)和普通护理组(32例)。其中中医个性化心理护理组在常规治疗和护理基础上,仅给予中医个性化心理护理;常规护理组仅进行常规的治疗和护理。分析比较两组干预前后的腹围、尿量、疗效,以及生活质量。结果:中医个性化心理护理可有效降低肝硬化腹水病人腹围、尿量,且中医个性化心理护理组的有效率和生活质量均高于普通护理组(均为P0.05)。结论:针对肝硬化腹水患者给予中医个性化心理护理,可有效提高疗效,提高生活质量,具有重要的临床推广价值。  相似文献

The correlation of serum arylesterase (PON1) activity on phenylacetate determined by an integrated method to classical biochemical indexes of liver damage was investigated for the use of PON1 activity to evaluate liver damage. PON1 reaction curve as absorbance at 270 nm for 0.20 mmol/L phenylacetate hydrolysis was analyzed by the integrated method to determine maximal PON1 reaction rate. Classical biochemical indexes of liver damage were determined routinely. The 95% confidence threshold of PON1 activity in sera from healthy individuals was 2.12 mkat/L [(4.73±1.31) mkat/L, n=105]. PON1 activity in clinical sera was closely correlated to serum albumin, total protein and the ratio of albumin to globulins, but was weakly correlated to both direct and total bilirubin in serum. There were no correlations of PON1 activity to γ-glutamyltransferase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase. Among 127 clinical sera with PON1 activity>2.12 mkat/L, there were 92% healthy individuals examined by albumin, 90% healthy individuals examined by total protein, 88% healthy individuals examined by total bilirubin, 86% healthy individuals examined by direct bilirubin and 64% healthy individuals examined by the ratio of albumin to globulins, respectively. In each group of healthy individuals judged by classical biochemical indexes of close correlation to PON1 activity, percentage of healthy individuals examined by PON1 activity was always >80%. These results suggested PON1 activity on phenylacetate estimated by the integrated method was also suitable for the evaluation of liver damage.  相似文献

The reference intervals (RIs) of serum aminotransferases and Gamma-glutamyl transferase (GGT) have been established many years ago. Recent RIs are not available. The prospective study was conducted to re-evaluate the RIs of liver enzymes and the effect of demographic and anthropometric variables on them in western Indian population. A total of 1059 blood donors comprised the study population. Anthropometry and serum liver enzymes levels were measured. Subjects were categorized into normal weight and overweight by using body mass index (BMI) and waist circumference (WC). For RI determination, non-parametric methodology recommended by IFCC/CLSI was adopted. Mann–Whitney test and Spearman’s rank correlation were used for statistical analysis. Upper limit of normal reference value of liver enzymes were lower in female compared to male. (ALT—23.55 F vs 36.00 M, GGT—34.58 F vs 36.20 M) When RI of liver enzymes were calculated according to body mass index, the upper limit of normal of ALT and GGT were higher in overweight group compared to normal weight group. (ALT—38.00 vs 27.00 IU/L and GGT—37.59 vs 35.26 IU/L). In both male and female, liver enzymes correlated significantly with age. WC and BMI were positively correlated with AST, ALT and GGT in both subgroups and the correlation was stronger in male. Demographic factors should be considered for making liver enzyme tests more clinically relevant. Gender based partitioning should be adopted for serum alanine aminotransferase (ALT) and GGT reference values for Western Indian population.  相似文献

Grapes are the richest source of antioxidants due to the presence of potent bioactive phytochemicals. In this study, the phytochemical contents, scavenging activities and protective role against H2O2-induced oxidative stress in liver tissue ex vivo of four grape (Vitis vinifera) cultivars extracts, namely Flame seedless (black), Kishmish chorni (black with reddish brown), Red globe (red) and Thompson seedless mutant (green), were evaluated. The total phenolics and flavonoids content in pulp or skin fractions of different grape cultivars were in the range of 47.6–310 mg gallic acid equivalent/g fresh weight (fw), and 46.6–733.3 µg catechin equivalent/g fw respectively. The scavenging activities in skin of different grape varieties against 2,2-diphenyl-1-picrylhydrazyl (44–58 %), hydrogen peroxide (15.3–18.6 %), and hydroxyl radicals (50–85 %), were higher than pulp of the corresponding cultivars. These scavenging activities of grape extracts were found to be significantly (p < 0.01) correlated with the levels of total phenols, flavonoids and ascorbic acid. Liver tissues from goat treated with H2O2 (500 μM) showed significantly decreased GSH content by 42.9 % and activities of catalase by 50 % and glutathione reductase by 66.6 %; while increased thiobarbituric acid reactive substances and nitric oxide level by 2.53- and 0.86-fold, respectively, and activity of glutathione S-transferase by 0.96-fold. Grape skin extracts showed the stronger protective activity against H2O2-induced oxidative stress in liver tissue ex vivo, than its pulp of any cultivar; and the Flame seedless (black) cultivar showed the highest potential. In conclusion, our study suggested that the higher antioxidant potential, phytochemical contents and significant scavenging capacities in pulp and skin of grape extracts showed the protective action of grape extracts against H2O2-induced oxidative stress in liver tissue ex vivo.  相似文献

目的:确立白藜芦醇脂质体比白藜芦醇是否具有更好的肝靶向性。方法:采用薄膜超声分散法制备白藜芦醇脂质体,从小鼠尾静脉注射白藜芦醇和白藜芦醇脂质体,以紫外检测法检测小鼠肝脏中白藜芦醇含量为考察指标。比较白藜芦醇与白藜芦醇脂质体在0.5h、2h、8h后在肝脏中的存留量确定肝靶向性差异。结果:白藜芦醇脂质体在尾静脉注射0.5h、2h、8h后在肝脏比单纯白藜芦醇具有更高的肝组织分布量。结论:白藜芦醇脂质体具有更好的肝靶向性。  相似文献

Anabasis articulata (Forssk) Moq. (Chenopodiaceae) is an herb, grows in Egypt, and used in folk medicine to treat diabetes, fever, and kidney infections. The protective and therapeutic effects of the ethanol extract of A. articulata aerial parts were evaluated against dimethylnitrosamine (DMN)-induced liver fibrosis, compared with the standard drug, silymarin. Hepatic hydroxyproline content, serum transforming growth factor-β1 (TGF-β1), interleukin 10 (IL-10) and fructosamine were measured as liver fibrosis markers. Hepatic malondialdehyde (MDA), nitric oxide (NO), catalase (CAT), glutathione reductase (GR) and glutathione content (GSH) were measured as oxidant/antioxidant markers. Parallel histopathological investigations were also performed. Protective and therapeutic administration of A. articulata (100 mg/kg daily for 4 weeks), markedly prevented DMN-induced loss in body and liver weights. The extract significantly inhibited the elevation of hepatic hydroxyproline, NO and MDA (P < 0.05), as well as serum fructosamine, and TGF-β1 (P < 0.05) induced by DMN while it restored IL-10 to normal level in both protective and therapeutic groups. Furthermore, A. articulata prevented the depletion in CAT, GR, and GSH levels (P ≤ 0.05). In addition, oral administration of A. articulata extract and silymarin to both protective and therapeutic groups reduced the increase in liver function enzyme activities; alanine and aspartate amintransferases, gamma-glutamyl transferase in addition to alkaline phosphatase, and caused significant increase in serum albumin concentration as compared to DMN group. These data corresponded closely with those obtained for the drug silymarin. Histopathological studies confirmed the biochemical data and revealed remarkable improvement in liver architecture. Thus, it could be concluded that, A. articulata extract exhibited in vivo hepatoprotective and therapeutic effects against DMN-induced liver injury and may act as a useful agent in controlling the progression of hepatic fibrosis through reduction of oxidative stress and improving liver function.  相似文献

[目的]观察双环醇对慢性乙型病毒性肝炎患者肝纤维化程度以及肝组织内转化生长因子β1(TGF-β1)和Ⅰ、Ⅲ型胶原含量的影响.[方法]20例慢性乙型病毒性肝炎患者以双环醇75~150 mg/d治疗24周,治疗前后分别肝活检进行常规病理学检查,并应用免疫组化技术和多媒体彩色病理图文分析系统观察双环醇治疗前后肝组织内TGF-β1和Ⅰ、Ⅲ型胶原含量的变化.[结果]慢性乙型病毒性肝炎肝纤维化患者经双环醇治疗后,肝组织炎症和纤维化程度明显改善,肝组织内TGF-β1和Ⅰ、Ⅲ型胶原含量均较治疗前明显降低,差异有显著性(P<0.05).[结论]双环醇治疗可以减轻慢性乙型病毒性肝炎患者肝脏炎症反应,同时明显降低慢性乙型病毒性肝炎患者肝组织内TGF-β1和Ⅰ、Ⅲ型胶原的含量,从而发挥抗纤维化作用.  相似文献

目的:探讨运动与CLA对青春期肥胖大鼠内脏脂肪及肝肾功能的影响。方法:高脂饲养大鼠建模7周后,取24只肥胖大鼠随机分成4组,每组6只,分别为对照组、CLA组、运动组、运动+CLA组。运动方案为跑台坡度0°,速度为21-25 m/min,总运动时间60min/天,5次/周,给药方案为运动后即刻灌喂1.6g/kg剂量的CLA,共持续干预8周。干预结束后麻醉大鼠采集血液及组织,测肝肾功能和内脏脂肪重量。结果:8周干预后,运动组、运动+CLA组的体重、内脏脂肪重量及体脂率均明显低于对照组、CLA 组(P<0.01,P<0.05),对照组和CLA组无明显差异(P〉0.05);对照组、运动组、运动+CLA组谷草转氨酶和谷草丙比数值明显高于CLA组(P<0.01),其他组别之间无明显差异(P>0.05);运动组、运动+CLA组血尿素氮和肌酐较对照组、CLA 组有变化,但无明显差异(P>0.05)。结论:8周运动与 CLA干预可有效降低青春期肥胖大鼠体重、内脏脂肪重量、体脂率,运动结合CLA能显著减少内脏脂肪含量,增加瘦体重,降低体脂率,具有明显的减肥效果。运动和CLA干预有一定的保肝作用,对肾脏功能无明显影响。  相似文献
