锌诱导对力竭运动大鼠金属硫蛋白的影响及其对心肌的保护作用

摘    要:通过预先用锌诱导后大鼠做力竭性运动,来观察锌诱导金属硫蛋白(MT)合成对心肌的保护作用及其可能机制.方法:选用健康雄性SD大鼠30只,体重220~250 g,随机A、B、C分为3组,即:A组为对照组;B组为一次性力竭运动组;C组为锌 一次性力竭运动组,每组10只.满8周后,测定其心肌组织中的MT含量、组织中钙的含量、巯基(-SH)和丙二醛(MDA)含量、以及心肌中的谷胱甘肽过氧化物酶(CSH-px)和Na -K -ATP酶的活性.结果:1)与对照组相比,力竭运动组大鼠心肌组织中MT的含量较对照组显著降低(P<O.05);心肌组织中MDA含量和总钙量显著增高(P<0.05);-SH含量以及GSH-px活性和Na -K -ATP酶的活性显著降低(P<O.05).2)与力竭运动组相比;锌 力竭运动组大鼠心肌组织中MT含量较力竭运动组显著增高(P<O.05);心肌组织中MDA含量和总Ca2 量显著降低(P<0.05);-SH含量及GSH-px活性和Na -K -ATP酶的活性显著增高(P<0.05).结论:预先用锌诱导可减少力竭运动后心肌组织中MT含量的显著下降,降低心肌组织脂质过氧化水平,提高抗氧化酶的活性,防止钙超载,对力竭运动后心肌细胞的损伤具有保护作用.MT和金属锌密切相关,二者的相互作用可能共同参与保护力竭运动后机体组织的损伤,并促进损伤组织的快速修复.

Abstract:Objective :This paper discusses the protective effects of MT on heart, through analysis exhaustive exercise induced by zinc in rat heart.Method:30 Sprague-Dawley rats with weight between 220g to 250g are selected and divided into 3 Groups randomly: Group A is a controlled group without exercise,Group B is the exhaustive swimming exercise group,Group C is the zinc exhaustive swimming exercise group,with 10 rats in each group.After 8 weeks,the contents of MT,the levels of MDA,the contents-SH,the contents of Ca2 ,the activity of GSH-px and Na -K -ATP's enzyme in myocardial of rats are measured.Result : 1.Compared with the controlled Group A,the MT contents of rats in Group B are obviously decreased(P<0.05),the contents-SH and the activity of GSH-px and Na -K -ATP's enzyme are obviously decreased(P<0.05),while the levels of MDA and the contents of Ca2 increased(P<0.05).2.Compared with the exhaustive exercise group Group B,the contents of MT of rats in Group C are notably increased(P<0.05),the-SH contents and the activity of GSH-px and Na -K -ATP's enzyme are obviously increased(P<0.05),while the levels of MDA and Ca2 are obviously reduced(P<0.05).Cnclusion : By zinc inducing in exhaustive exercise may be reduce the contents of MT obviously descend in rats' myocardial,decrease lipid peroxidationl,increase the activitys of antioxidant enzymes,prevent calcium from overloading,protect cardiomyocytes against injury.MT and metal Zn are mutually related and interacted closely,they probably participate in protecting organism organization from injury and promoting the quickly repairing of the.injury organization after exhaustive exercise.
